Uveal melanoma-derived extracellar vesicles displayed transforming potential and carried portein cargo involved in metastatic niche preparation

ABSTRACT: Extracellular Vesicles (EVs) are oval membrane-enclosed structures with highlyheterogeneous content. EVs are known to play important roles in intercellular communication by transporting their contents between cells whereby they alter the properties of both donor and recipient cells.Uveal melanoma (UM) is the most common primary intraoculartumor in adults andit displays a high frequency of metastases to the liver. The aim of this study was to characterize the protein signature, determine the potential biomarker of circulating EVs derived from UM cell lines.Mass spectrometry of proteins from UM-EVs was performed and shwed the presence of exosomal markers and proteins involved in cell-cell and focal adhesion. In addition, we observed high expression of HSP70, HSP90 and integrin alpha V.

Extracellular vesicles (EVs) carry molecules derived from donor cells and are able to alter the properties of recipient cells. They are important players during the genesis and progression of tumors. Uveal melanoma (UM) is the most common primary intraocular tumor in adults and is associated with a high rate of metastasis, primarily to the liver. However, the mechanisms underlying this process are poorly understood. In the present study, we analyzed the oncogenic potential of UM-derived EVs and  ...[more]
