Environment and its impact on barley grain composition

Project description:To provide a more detailed insight into molecular changes in seeds exposed to the accelerated ageing process, we excised embryos and compared the differences in the proteome composition. Seeds collected from four contrasting locations were exposed to accelerated ageing. Next, embryos were dissected and analyzed in five biological replicates [1-5]. Transcriptome Profiling for Escherichia coli K12 wild type and ΔcsdA mutant before and after rifampicin-treatment

Project description:The cell cycle coordinates with growth to ensure suitable genome inheritance. The CsdA protein is a highly conserved, DEAD-box RNA helicase and assists RNA structural remodeling at low temperature. The absence of CsdA yields a severe growth defect manifested by increased doubling time, more extensive cell size, or slower colony growth at low temperature. Nevertheless, it is unknown how the lack of CsdA at low temperature affects the cell cycle and the causes of the yield of severe growth defect phenotypes. This work shows that the lack of CsdA results in incoordination of the cell cycle to growth at low temperature. We performed microarray analyses to compare the transcriptome profiles of ΔcsdA mutant with WT cell before and after rifampicin-treatment in LB to understand how the lack of CsdA at low temperature affects the cell cycle and the causes of the yield of severe growth defect phenotypes.

Photooxidative stress and light acclimation in the PSII antenna-deficient ch1 mutant

Transcriptome and physiological analysis of rapeseed tolerance to post-flowering temperature increase

Project description:This study investigates the transcriptome and physiological responses of rapeseed to post-flowering temperature increases, providing valuable insights into the molecular mechanisms underlying rapeseed tolerance to heat stress. Two rapeseed genotypes, Lumen and Solar, were assessed under control and heat stress conditions in field experiments conducted in Valdivia, Chile. Results showed that seed yield and seed number were negatively affected by heat stress, with genotype-specific responses. Lumen exhibited a 9.3% average seed yield reduction, while Solar showed a 28.7% reduction. RNA-seq analysis of siliques and seeds revealed tissue-specific responses to heat stress, with siliques being more sensitive to temperature stress. Hierarchical clustering analysis identified distinct gene clusters reflecting different aspects of heat stress adaptation in siliques, with a role for protein folding in maintaining silique development and seed quality under high temperature conditions. In seeds, three distinct patterns of heat-responsive gene expression were observed, with genes involved in protein folding and response to heat showing genotype-specific expression. Gene coexpression network analysis revealed major modules for rapeseed yield and quality, as well as the trade-off between seed number and seed weight. Overall, this study contributes to understanding the molecular mechanisms underlying rapeseed tolerance to heat stress and can inform crop improvement strategies targeting yield optimization under changing environmental conditions.

Roles of RDR701 and DCLs in Rice PhasiRNA biogenesis

Project description:In this study, we analyzed function of the rice RDR701 and various DCLs proteins in small RNA processing. small RNA deep sequencing analyses revealed that RDR701 and OsDCL4 is involved in tasiRNAs and 21- and 24-nt phasiRNAs biogenesis. Further analyses indicate RDR701 coupling with OsDCL4 and OsDCL3b are required for the biogenesis of 21- and 24-nt phasiRNAs, respectively. The triggering miRNAs for intial both size phasiRNAs,are also shown RDR701- and OsDCL4-dependent, suggesting two sequential round activity of RDR701 in associated with different DCLs are essential for rice phasiRNAs biogenesis. Four small RNA libraries were constructed from panicles of zhonghua11 (a wild-type Japonica rice variety) and rdr701-1 at high temperature (Day average temperature 32M-bM-^DM-^C) and low temperature (Day average temperature 28M-bM-^DM-^C).

PIF7 is the master regulator of thermomorphogenesis in shade [ChIP-seq]

Project description:The size and shape of plant organs are highly responsive to environmental conditions. The plant's embryonic stem, or hypocotyl, displays phenotypic plasticity, in response to light and temperature. The, hypocotyl of shade avoiding species elongate to outcompete neighbouring plants and secure access to sunlight. Similar elongation occurs in high temperature. PHYTOCHROME-INTERACTING FACTORS (PIFs) family transcription are known to be importenet players in these responses. However, it is poorly understood how environmental light and temperature interact to affect plants development. We found that low R/FR combined with warm temperature produces a synergistic hypocotyl growth response that dependent on PIF7 and the hormone auxin. We demonstrate that additional, unknown factor/s must be working downstream of the phyB-PIF-auxin module. As shade responses are known to affect yield, susceptibility to pathogens, and fruit quality in many species, our findings will improve the predictions of how plants will respond to increased ambient temperatures when grown at high density, a condition in which mutual shading occurs.
