Arabidopsis proteome in the tripartite interaction between cytokinin, light and temperature

ABSTRACT: Transgenic Arabidopsis plants carrying dexamethasone-inducible barley cytokinin oxidase/dehydrogenase (CaMV35S>GR>HvCKX2) and agrobacterial isopentenyl transferase (CaMV35S>GR>ipt) constructs were profiled to elucidate proteome-wide response to cold stress under different light conditions.

Barley root proteome and metabolome in response to cytokinin

Project description:Cytokinin is a phytohormone involved in the regulation of diverse developmental and physiological processes in plants. Its potential for biotechnology and development of high-yield and more resilient plants has been recognized, but the molecular mechanisms behind its action are far from understood. In this report, the roots of barley seedling were explored as a new tool to reveal as yet unknown cytokinin-responsive proteins. Significant differences were reproducibly observed for 176 proteins, and at least some of the revealed cytokinin-responsive pathways were confirmed in metabolome analysis, including alterations in phenylpropanoid pathway, amino acid biosynthesis or ROS metabolism. Bioinformatics analysis indicated a significant overlap between cytokinin response and response to abiotic stress. This was confirmed by comparing proteome and metabolome profiles in response to drought, salinity or a period of temperature stress. The results illustrate complex abiotic stress response in the early development of model crop plant and confirm an extensive crosstalk between plant hormone cytokinin and response to temperature stimuli, water availability or salinity.

Identification of genes cooperatively regulated by cytokinin and HD-ZIP III transcription factors

Project description:In Arabidopsis thaliana, cytokinin responsive B-type ARR transcription factors and HD-ZIP III transcription factors such as REVOLUTA (REV), act cooperatively as master regulators of shoot regeneration. To identify the downstream targets of ARR-HD-ZIP III transcriptional complex, we used an inducible line of REV, 35S::FLAG-GR-rREV, in which FLAG-tagged miR165/6-non-targetable form of REV (rREV)-GR fusion protein was expressed from 35S promoter. DEX treatment induced activation of REV by translocation of FLAG-GR-rREV fusion protein from cytoplasm to the nucleus. We treated 35S::FLAG-GR-rREV seedlings with 6-benzylaminopurine (6-BA, a cytokinin), dexamethasone (DEX), or 6-BA+DEX for 2 hours. Total RNAs were extracted and subjected to Agilent Arabidopsis Gene Expression Microarray analyses. The differentially expressed genes (>1.5-fold, p<0.05) were identified. 10-day-old 35S::FLAG-GR-rREV plants were treated with 6-benzylaminopurine (6-BA), dexamethasone (DEX), or 6-BA+DEX for 2 hours. DEX treatment induced activation of REV by translocation of FLAG-GR-rREV fusion protein from cytoplasm to the nucleus. Total RNA was extracted with RNeasy Mini Kit and hybridized to Agilent Arabidopsis Gene Expression Microarray. Differentially expressed genes were defined by a 1.5-fold expression difference with a P value<0.05. Biological replicates were performed.

Multifaceted activity of cytokinin at distinct phases of leaf development shapes its size and structure in Arabidopsis thaliana

Project description:To get an insight into cytokinin-induced alterations of molecular networks underlying size and structure of a leaf, transgenic Arabidopsis lines (Col-0 background) CaMV35S>GR>ipt, and CaMV35S>GR>HvCKX2 were cultivated in a growth chamber under controlled environmental conditions (50-60% relative humidity, long day, 21/19 °C day/night temperature, 110 µmol m-2 s-1). Plants were activated at 14 DAS by watering once with 50 ml of distilled water supplemented with 10 µM dexamethasone dissolved in 5x10-4% (v/v) DMSO (DEX samples). Corresponding mock-treated control plants were watered with DMSO in water.

Ethylene-independent Promotion of Photomorphogenesis by Cytokinin Requires a Functional Cytokinin and Light Signaling Pathway.

Project description:The transition of skotomorphogenesis to photomorphogenesis is induced by the perception of light and is characterized by the inhibition of hypocotyl elongation and opening of cotyledons. Although it is known that the plant hormone cytokinin, when applied in high concentrations, inhibits hypocotyl elongation in the dark-grown Arabidopsis plants, it is unclear to what extent this response is the result of cytokinin alone or cytokinin-induced ethylene production. We show that treatment of etiolated seedlings in presence of ethylene inhibitors (eg. AgNO3) or treatment of the ethylene-resistant mutant ein2, resulted in a significant inhibition of hypocotyl elongation. This indicates that cytokinin induced de-etiolation is largely independent of ethylene and suggests a close connection between the cytokinin two component system and the light singalling networks. We show that this cytokinin signal is mainly mediated through the cytokinin receptor ARIBIDOPSIS HISTIDIN KINASE 3 (AHK3) and the ARABIDOPSIS RESPONSE REGULATORS 1 (ARR1) in combination with ARR12. Interestingly, mutation of COP1, DET1 and CIN4/COP10 renders plants insensitive to cytokinin and these factors are indispensable for the transcriptional response during cytokinin induced de-etiolation which indicates that a functional light signaling pathway is essential for this cytokinin response. In addition, the cytokinin effect on hypocotyl elongation is highly dependent on the ambient light conditions where higher light intensities causes a switch in the response to CK from an inhibitor to a promoter of hypocotyl elongation.

Trans-Zeatin-N-Glucosides have biological activity in Arabidopsis thaliana

Project description:Cytokinin is an indispensable phytohormone responsible for a number of physiological processes ranging from root development to leaf senescence. The term “cytokinin” refers to several dozen adenine-derived compounds which occur naturally in plants, including the model plant Arabidopsis thaliana. Cytokinins (CKs) can be divided up into various classes and forms; the base forms are generally considered to be active while highly abundant cytokinin-N-glucosides (CKNGs), which are composed of a CK base irreversibly conjugated to a glucose molecule, are considered inactive. However, results from early CK studies suggest CKNGs do not always lack activity despite the perpetuation in the literature that they are inactive. Here we show that exogenous application of CKNGs to Arabidopsis tissue results in a CK response comparable to the application of an active CK base. These results are most apparent in senescence assays in which both a CK base (tZ) and CKNGs (tZ7G, tZ9G) delay senescence in cotyledons. Further physiological experiments involving root growth and shoot regeneration revealed CKNGs do not always have the same effects as CK bases, and these compounds have largely distinct effects on the transcriptome, as well as the proteome. These data are in direct contradiction to previous reports of CKNGs being inactive and raise questions about the function of these compounds as well as their mechanism of action. Because CKNGs make up the majority of CKs in Arabidopsis and other Angiosperms, it is especially important to understand their physiological effects in order to have a more holistic understanding of this essential phytohormone.

Project description:Barley is one of the most important cereals, which is used for breweries, animal and human feeds. Genetic manipulation of plant hormone cytokinin (CK) levels may influence several physiological processes, besides others stress tolerance, root formation and crop yield. In planta, endogenous CK status is finely regulated by the enzyme cytokinin dehydrogenase (EC 1.5.99.12; CKX), that irreversibly cleaves the N6-side chain of adenine-derived CKs. Increasing grain yield by mean of manipulation of endogenous CK content was assayed by the silencing of the HvCKX1 gene. Moreover, to elucidate the putative role of HvCKX1 gene on grain production, knocked-out Hvckx1 mutant plants were generated using RNA-guided Cas9 system.
