Egg proteome in vtg1 and vtg3 knocked-out zebrafish females

ABSTRACT: CRISPR/Cas9 knock-out of the vtg1 and vtg3 genes was performed in zebrafish separately. This project involves proteomics screening for validation of the absence of targeted proteins in vtg1 and vtg3 and their variants in knocked out female zebrafish in eggs and investigating possible changes within the corresponding proteomes as a results of the absence of these specific vitellogenins and their variants.

Background
We previously reported the results of CRISPR/Cas9 knock-out (KO) of type-I and type-III vitellogenins (Vtgs) in zebrafish, which provided the first experimental evidence on essentiality and disparate functioning of Vtgs at different stages during early development. However, the specific contributions of different types of Vtg to major cellular processes remained to be investigated. The present study employed liquid chromatography and tandem mass spectrometry (LC-MS/MS) to meet ...[more]
