MKRN3 interacting proteins identified by LC-MS/MS

ABSTRACT: MKRN3, whose deletion or loss-of-function mutations were genetically associated with human centeral precocious puberty (CPP). To identify the potential substrates for MKRN3, MKRN3 was transfected into HEK293T cells as bait, and its interacting proteins were identified by mass spectrum and functions were extensively studied.

Project description:Hypothalamic hamartomas (HHs) are congenital lesions of the neuroendocrine brain composed of neurons and astroglia. Frequently, HHs are associated with central precocious puberty (CPP) and/or gelastic seizures. Because HHs might express genes similar to those required for the initiation of normal puberty we used cDNA arrays to compare the gene expression profile of a HH associated with CPP with three HHs not accompanied by sexual precocity. Our aim was to identify genes whose expression may be selectively altered in the HH with CPP and hence, involved in the onset of puberty. Experiment Overall Design: Affymetrix arrays were used to detect global changes in gene expression. The results of this analysis were confirmed by semi-quantitative PCR.

Integrated Analysis of Proteomics and Metabolomics in Girls with Central Precocious Puberty

Project description:Central precocious puberty (CPP) is a multifactorial and complex condition. Traditional studies focusing on a single indicator cannot always elucidate this panoramic condition but these may be revealed by using omics techniques. Proteomics and metabolomics analysis of girls with CPP were compared to normal controls and the potential biomarkers and pathways involved were explored. Serum proteins and metabolites from normal girls and those with CPP were compared by LC-MS/MS. Multivariate and univariate statistical analysis were used to identify the differentially expressed proteins (DEPs) and differentially expressed metabolites (DEMs). Functional annotation and pathway enrichment analysis were performed by using GO and KEGG databases, and candidate markers were screened. Finally, bioinformatic analysis was used to integrate the results of proteomics and metabolomics to find the key differential proteins, metabolites and potential biomarkers of CPP.

Interaction partners of arginyl-tRNA protein transferase in the moss Physcomitrella patens

Project description:Protein arginylation is a post-translational modification at the N-terminus of proteins and is crucial for viability and physiology in higher eukaryotes. The lack of arginylation causes severe developmental defects in plants and embryo lethality in Drosophila and in mice. Although several studies investigated impact and function of the responsible enzyme, the arginyl-tRNA protein transferase (ATE) in plants, identification of arginylated proteins by mass spectrometry was not hitherto achieved. In the present study, we report the identification of targets and interaction partners of ATE in the model plant Physcomitrella patens by mass spectrometry employing two different immuno-affinity strategies and a recently established transgenic ATE:GUS reporter line (Schuessele et al., 2015 New Phytol., DOI: 10.1111/nph.13656). Here we use a commercially available antibody against the fused reporter protein (GUS) to pull down ATE and its interacting proteins and validate the in vivo interaction with a class I small heatshock protein via Förster resonance energy transfer (FRET).

RNA Sequencing for MKRN3 regulating genes identification in GT1-7 cells

Project description:Purpose: The deletion or loss-of-function of MKRN3 were genetically associated with human centeral precocious puberty (CPP).The goal of this study is to explore the global genes expression changes on MKRN3 overexpessing by high-throughput RNA sequencing analysis. Methods: The mRNA profiles of wild-type (WT) and MKRN3 overexpressed GT1-7 cells were generated by deep sequencing, in triplicate, using Illumina GAIIx. The sequence reads that passed quality filters were aligned to mus musculus genomic reference (GRCm38) using HISAT2. Read counts were summarized at the gene level in each sample with HTSeq. Results: Using an optimized data analysis workflow, we mapped about 19 million sequence reads per sample to the mouse genome (build mm10) and identified 13,041 transcripts in the GT1-7 cells of WT and MKRN3. Approximately 6% of the transcripts showed differential expression between the WT and MKRN3 overexpressed cells, with a fold change ≥2 and p-adjust value <0.05. 354 genes were up-regulated by MKRN3, while 395 genes were down-regulated by MKRN3 in this study. Conclusions: Our study is the first effort to explore MKRN3 on gobal genes expression, and hundreds of different expressed genes have been identified.

Effect of MKRN3 deletion on gene expression in hypothalamic neurons derived from human induced pluripotent stem cells (hiPSCs)

Project description:Makorin ring finger protein 3 (MKRN3) was identified as an inhibitor of puberty initiation with the report of loss-of-function mutations in association with central precocious puberty. To investigate the roles and mechanisms of action of MKRN3 within the human hypothalamus, we used hypothalamic neurons derived from human induced pluripotent stem cells (hiPSCs). MKRN3 deletion was introduced into hiPSCs using CRISPR interference (CRISPRi) technology. Three separated hypothalamic differentiation of MKRN3-wildtype hiPSCs (MKRN3-WT I, II and III) and MKRN3-deficient hiPSCs (MKRN3-KO I, II and III) were performed using an established hypothalamic neuron differentiation protocol. To identify hypothalamic targets of MKRN3, we performed comparative transcriptome analysis by RNA sequencing (RNA-seq) of MKRN3-WT and MKRN3-KO hypothalamic neurons after 30 days of differentiation.
