Differential phosphoproteomics upon chitin-treatment in Marchantia polymorpha

Project description:Pattern-recognition receptor (PRR)-triggered immunity (PTI) plays a pivotal role in plant immunity to ward off a wide range of pathogenic microbes. The model liverwort Marchantia polymorpha is gaining popularity in investigating the evolution of plant-microbe interactions. The M. polymorpha is capable of triggering defense-related gene expression by sensing components in bacterial and fungal extracts, suggesting existence of PTI in this plant model. However, the molecular components that would form PTI in M. polymorpha have not yet been described. We show that, in M. polymorpha, lysin motif (LysM) receptor-like kinase (LYK) MpLYK1 and LYK-related (LYR) MpLYR, among four LysM receptor homologs, are required for sensing chitin and peptidoglycan (PGN) fragments and thereby triggering a series of immune responses. Phosphoproteomic analysis of M. polymorpha in response to chitin treatment comprehensively identified regulatory proteins that would shape LysM-mediated PTI. The identified proteins covered homologs of well-described PTI components in angiosperms as well as proteins whose roles in PTI are not yet determined including the blue-light receptor phototropin MpPHOT. We revealed that MpPHOT is required for a negative feedback of defense-related gene expression during PTI. Taken together, this study provides the basic framework of LysM-mediated PTI in M. polymorpha and demonstrates the utility of M. polymorpha as a plant model for discovering novel or fundamental molecular mechanisms underlying PRR-triggered immune signaling in plants.

OXI1 coordinates programmed cell death by promoting salicylic acid, N-hydroxypipecolic acid and reactive oxygen species accumulation in Arabidopsis

Project description:The protein kinase OXI1 is induced by a large number of stress conditions and regulates the interaction of plants with pathogenic and beneficial microbes but little is yet known on the underlying mechanisms. In this work, we generated Arabidopsis OXI1 knock out and overexpression lines and show by transcriptome, proteome and metabolome analysis. Our work revealed that OXI1 regulates biosynthesis of SA via CBP60g-induced expression of isochorismate synthase SID2. OXI1 also induces the transcription factor WRKY33 and its downstream target PAD3, resulting in accumulation of camalexin. Moreover, OXI1 regulates ALD1, SARD4 and FMO1 to promote the biosynthesis of pipecolic acid (Pip) and N-hydroxy pipecolic acid (NHP), mediating systemic acquired resistance to bacterial infection. In contrast to OXI1 overexpressor plants, OXI1 knock out plants show enhanced expression of nuclear and chloroplast genes of photosynthesis, and accordingly enhanced growth under ambient conditions. Overall, these results show that OXI1 plays a key role in regulating the trade-off between growth and defense in plants.

Project description:To evaluate the impact of the RNA purification method on extracellular RNA (exRNA) sequencing, 8 different RNA purification kits were compared by applying Small RNA sequencing (Illumina) to exRNA from human healthy donor plasma. Minimum and maximum plasma input volumes recommended by the manufacturers were tested in triplicate. Due to donor privacy concerns the raw data for this study have been submitted to the controlled-access archive EGA under the accession EGAS00001005263.

Transcription profiling of thrombin stimulated human microvascular endothelial cell line-1 (HMEC1) over time and vs untreated cells

Project description:Thrombin exerts pleiotropic effects on the endothelium. Nevertheless, little is known about its capacity to generate endothelial microparticles, a hallmark of endothelial activation. To gain insight into the mechanisms involved in microparticles release, the human microvascular endothelial cell line-1 (HMEC1) was stimulated with thrombin at 1 IU/ml for different times. The gene expression profiling in stimulated HMEC-1 was compared to that obtain with untreated cells.
