Extracellular vesicles as prognostic biomarkers for Multiple Myeloma - a clinical population-based study

ABSTRACT: Multiple myeloma (MM) is an incurable hematological malignancy of mature B lymphocytes. In this study patients with MGUS, SMM and MM followed in two hematology departments were enrolled. Extracellular vesicles were isolated from bone marrow (BM) and peripheral blood (PB) followed by mass spectrometry based bottom up proteomics.

Project description:The aim of the project was to explore large extracellular vesicles capacity to seperate non lung cancer from lung cancer cases. A case control study of patients suspected for lung cancer (12 non lung cancer and 12 lung cancers). The raw files are labeled according to clinical status. Small and large extracellular vesicles were isolated by differential centrifugation. The proteome of full BAL, vesicle depleted BAL, small and large extracellular vesicles were characterized by LC-MS. Small extracellular vesicles were further analyzed by nanoparticle tracking analysis, Transmission electron microscopy and western blots.

Unbiased undepleted malignant pleural effusion proteomics study

Project description:Pleural effusion (PE) occurs as a consequence of various pathologies. Malignant effusion due to lung cancer is one of the most frequent causes. Methods for accurate differentiation of malignant from benign PE cases are an unmet clinical need. Proteomics profiling of PE has shown promising results. However, mass spectrometry (MS) analysis typically involves the tedious elimination of abundant proteins before analysis, and clinical annotation of proteomics profiled cohorts is limited. In this study, PE from 97 patients was investigated by applying label-free state-of-the-art liquid chromatography-mass spectrometry (LC-MS) to find potential novel biomarkers that correlate with immunohistochemistry assessment of tumor biopsy or survival data. The data set consists of 214 LC-MS runs.

Project description:The aggressive B cell lymphoma diffuse large B cell lymphoma (DLBCL) is a heterogeneous entity that requires more precise monitoring and prognosis molecular tools. Extracellular vesicles that are secreted by all cell types are currently recognized as serving as a proxy for the cell of origin. Utilizing cutting-edge mass spectrometry, the current study described and assessed the plasma small extracellular vesicles (sEVs) proteome's diagnostic and prognostic potential. The presence of DLBCL has a significant impact on the sEV proteome, and several proteins substantially correlate with DLBCL.Nevertheless, no proteins that highly correlated with non-GCB or GCB were found.

Multiple myeloma-derived extracellular vesicles modulate the bone marrow immune microenvironment

Project description:One of the hallmarks of multiple myeloma (MM) is a permissive BM microenvironment. Increasing evidence suggest that cell-to-cell communication between myeloma and immune cells via tumor cell-derived extracellular vesicles (EV) plays a key role in the pathogenesis of MM. Hence, we aimed to explore BM immune alterations induced by MM-derived EV. For this, we inoculated immunocompetent BALB/cByJ mice with a myeloma cell line - MOPC315.BM -, inducing a MM phenotype. Upon tumor establishment, characterization of the BM microenvironment revealed the expression of both activation and suppressive markers by lymphocytes, such as Granzyme b and PD-1, respectively. In addition, conditioning of the animals with MOPC315.BM-derived EV, before transplantation of the MOPC315.BM tumor cells, did not anticipate the disease phenotype. However, it induced features of suppression in the BM milieu, such as an increase in PD-1 expression by CD4+ T cells. Overall, our findings reveal the involvement of MOPC315.BM-derived EV protein content as promoters of immune niche remodeling, strengthening the importance of assessing the mechanisms by which MM may impact the immune microenvironment.

Project description:In C. albicans, after yeast-macrophage interaction up to 30% of yeast cells suffers oxidative stress mediated-apoptosis. In previous studies to reveal the response mechanisms to oxidative stress of this yeast we performed a quantitative proteomic analysis in cells exposed to hydrogen peroxide to evaluate changes in their proteome. In the analysis we identified a high increase in different oxidoreductase proteins like Oye32 but also in new undescribed proteins like Prn1, a protein similar to pirins. Pirins has already been described as a nuclear redox sensor in mammal cells which are implicated in oxidative stress response and as coregulator of different the transcription factors implicated in proliferation. Therefore, is probably that C. albicans Prn1 function will be related with oxidative stress response and also working as a regulator of different protein expression in response to this stress. In addition, prn1 lacks a homologue in S. cerevisiae, suggesting that it may be specific to pathogenic fungi, which makes important to study the role of this protein in the cell to finally be considered a possible drug target. In order to uncover the possible function of Prn1 in response to oxidative stress in C. albicans we carried out a quantitative proteomic assay in a wild type (SN250) and a prn1 strain treated with hydrogen peroxide. The main objective of the experiment was to identify which proteins decrease its abundance in prn1 cells in comparation with wild type cells in response to oxidative stress which means that their expression could be coregulated by Prn1. The second objective was to identify which proteins are overexpressed in response to oxidative stress to supply the lack or Prn1 which is very useful to identify new proteins implicated in oxidative stress response. Finally, these deceased and overexpressed proteins allowed us to identify which signalling pathways are regulated directly by Prn1 in response to oxidative stress.
