The infraorder Brachyura (true or short-tailed crabs) represents a successful group of marine invertebrates yet with limited genomic resources. Here we report a chromosome-anchored reference genome and transcriptomes of the Chinese mitten crab Eriocheir sinensis, a catadromous crab and invasive species with wide environmental tolerance, strong osmoregulatory capacity and high fertility. Proteomic analysis of accessory gland in sexually mature Eriocheir sinensis

Project description:The accessory gland (AG), an important component of the male reproductive system, enhances the fertility of spermatozoa during male reproduction. Some AG proteins are known to bind to the spermatozoa membrane and affect its function and properties. In this study, we describe, for the first time, a comprehensive catalog of proteins of the AG secreted fluid during the mature phase of the Chinese mitten crab (Eriocheir sinensis). AG secreted proteins were separated by one-dimensional gel electrophoresis and analyzed by reverse phase high-performance liquid chromatography coupled with tandem mass spectrometry (HPLC-MS/MS).

Transcriptome profiling reveals cleft palate-related genes in Chinese sturgeon (Acipenser sinensis)

Project description:The Chinese sturgeon (Acipenser sinensis) is anadromous fish distributed in Yangtze River and East China Sea. In this study, we reported cleft-palate Chinese sturgeons in artificial population for the first time. In order to explore the genetic basis of palate malformation in A. sinensis, Illumina RNA-seq technology was used to analyze the transcriptome data of normal and cleft-palate individuals in farmed Chinese sturgeons. Raw reads were obtained and assembled into 808,612 unigenes, with an average length of 509.33 bp and an N50 of 574 bp. Sequence similarity analyses against four public databases (Nr, Uniprot, KEGG and COGs) found 158,642 unigenes that can be annotated. GABAergic synapse and TGF-β signal pathway were the most two enriched pathways with high Richfactor in the analyses of different expressed genes. In these two signal pathways, six genes (GABRA4, GS, GNS, S6K, PITX2, and BMP8) were found as cleft-palate genes in Chinese sturgeon. These findings contribute to our understanding of the genetic basis of cleft palate in sturgeon, while simultaneously adding to our knowledge about craniofacial development.

Antibacterial properties and mechanism of action for serum obtained from the Chinese alligator (Alligator sinensis)

Project description:Chinese alligator (Alligator sinensis) is one of the rarest endangered reptiles found in China and possesses strong immune potential. This study tested the antibacterial ability of Chinese alligator serum (CAS) against Klebsiella pneumoniae, Escherichia coli, Staphylococcus aureus, and Pseudomonas aeruginosa and analyzed the underlying mechanisms. Results showed that the CAS had a marked antibacterial effect on K. pneumoniae, E. coli, and P. aeruginosa. However, S. aureus was only mildly affected, and this effect disappeared when incubated with Protease K. The serum proteome revealed that the antibacterial ability of CAS was produced by interactions between various proteins and that the complement proteins played a major antibacterial role. Furthermore, the prediction of the structure and function of complement component 3 revealed eight potential protein binding sites and one nucleic acid binding site that were likely related to the broad-spectrum antibacterial ability of this serum. This study provided evidence that CAS elicits significant antibacterial effects against some pathogens and provides the basis for further development of novel antibacterial drugs.

The tegument isolated using a freeze/thaw method, extraction with succession extracted buffers. The outer surface proteins of C. sinensis were labeled with biotin, and the biotinylated proteins were purified. The ES, tegumental protein and outer surface labeling protein were identified and analysis by high-resolution LC-MS/MS.

Proteome reveal abnormal spermatozoa in precocious Chinese mitten crab, Eriocheir sinensis

Project description:In juvenile Chinese mitten crabs, Eriocheir sinensis, the occurrence of sexual gland maturation in the first year is known as precocity and significantly affects culture productivity and profitability. The miniaturization of individuals is a key characteristic of precocious crabs. Most related studies focus on the underlying causes, nutritional quality, biochemical components and differentially expressed mRNAs of precocious crab individuals and tissues. The reproductive performance of precocious crabs is evaluated based on offspring quality. However, the status of precocious parental gametes (especially spermatozoa) directly related to the offspring has not been reported. To clarify the differences in spermatozoa between precocious and normal mature crabs, spermatozoal proteome data was obtained via high-throughput sequencing technology. The results revealed a total of 856 differentially expressed genes and 150 differentially expressed proteins in spermatozoa. Functional analysis showed that these genes and proteins identified in the spermatozoa of precocious E. sinensis were significantly enriched in the categories of substance and energy metabolism. And more, the miniaturization of spermatogenic cells was also identified in precocious crabs. This indicates that metabolic disorders might be the main reason for the miniaturization of spermatogenic cells in precocious E. sinensis males. The molecular defects in precocious sperm were closely related to the imbalances in essential substance and energy metabolism. These findings provide foundational information and insights into hidden biological irregularities associated with precocious phenotypes and will contribute to the further study of precocious E. sinensis males and even other crustaceans.
