Proteomic analysis of S-nitrosylation in HUVECs

ABSTRACT: HUVECs stimulated with HG+oxLDL or Mannitol+nLDL were subjected to the biotin-switch assay to enrich the S-nitrosylated proteins, the S-nitrosylated proteins were resolved by SDS-PAGE and gels were silver stained. Mass spectrometry was conducted to determine proteins with S-nitrosylation.

We hypothesized that SNO-GNAI2 induced Hippo-YAP dysfunction through enhancing coupling and activating G-protein coupling receptors (GPCRs).

RNA-seq of young and senescent human umbilical vein endothelial cells (HUVECs) under normal or high glucose conditions

Project description:Human endothelial cellular models are useful to disentangle the pathophysiological role of dysfunctional endothelium in the development of cardiovascular (CV) disease and organ damage in T2D. Here, we performed a RNAseq of human umbilical vein endothelial cells (HUVECs) undergoing replicative senescence and exposed to high glucose (25 mM) to investigate the combined effects of replicative senescence and high glucose on the transcriptional landscape of these cells. To gain insight into the molecular mechanisms driving the acquisition of a senescent phenotype following exposure to HG, we performed a RNA-seq assay on control (Ctr) and replicative senescent (Sen) HUVECs cultivated in presence/absence of HG culture medium (total number of samples = 12; number of samples in each cell type-medium condition group = 3) using the NovaSeq 6000 Illumina system. Differential expression analysis was performed in R environment (version 4.2.2) using the limma and edgeR Bioconductor R packages.

Proteomic analysis of S-nitrosylated nuclear proteins in rat cortical neurons

Project description:S-nitrosothiol Resin Assisted Capture(SNORAC) coupled with mass spectrometry analysis has been used to study S-nitrosylated proteins in rat cortical neurons. Proteins were captured on the resin by covalently attaching their former S-NO moiety. Then, nistrosylated proteins were eluted by on-bead digestion. Formerly S-nitrosylated peptides were also eluted from the beads in a second step, and analyzed separately for assessment of the nitrosylation site (qualitative analysis). Out of over 600 nitrosyltated proteins, 131 proteins have never been shown to be S-nitrosylated in any system and 612 are new targets of S-nitrosylation in cortical neurons. The site(s) of Snitrosylation were also identified for 59% of the targets.

Nitric oxide prevents aortic valve calcification by S-nitrosylation of USP9X to activate NOTCH signaling

Project description:Calcific aortic valve disease (CAVD) is an increasingly prevalent condition and endothelial dysfunction is implicated in its etiology. We previously identified nitric oxide (NO) as a calcification inhibitor by its activation of NOTCH1, which is genetically linked to human CAVD. Here, we show that NO rescues calcification by a S-nitrosylation-mediated mechanism in porcine aortic valve interstitial cells (pAVICs) and single cell RNA-seq demonstrated regulation of NOTCH pathway by NO. A unbiased proteomic approach to identify S-nitrosylated proteins in valve cells found enrichment of the ubiquitin proteasome pathway and implicated S-nitrosylation of USP9X in NOTCH regulation during calcification. Furthermore, S-nitrosylated USP9X was shown to deubiquitinate and stabilize MIB1 for NOTCH1 activation. Consistent with this, genetic deletion of Usp9x in mice demonstrated aortic valve disease and human calcified aortic valves displayed reduced S-nitrosylation of USP9X. These results demonstrate a novel mechanism by which S-nitrosylation dependent regulation of ubiquitin-associated pathway prevents CAVD.

A protein microarray-based analysis of S-nitrosylation

Project description:Yeast protein microarrays were utilized to investigate determinants of S-nitrosylation by biologically relevant low-mass S-nitrosothiols (SNOs). Large numbers of S-nitrosylated yeast proteins were identified after treatment with SNOs, among which those with active-site Cys thiols residing at N termini of alpha-helices or within catalytic loops were particularly prominent. However, S-nitrosylation varied substantially even within these families of proteins (e.g., papain-related Cys-dependent hydrolases and rhodanese/Cdc25 phosphatases), suggesting that neither secondary structure nor intrinsic nucleophilicity of Cys thiols was sufficient to explain specificity. Further analyses revealed a substantial influence of NO-donor stereochemistry and structure on efficiency of S-nitrosylation as well as an unanticipated and important role for allosteric effectors. Thus, high-throughput screening and unbiased proteome coverage reveal multifactorial determinants of S-nitrosylation (which may be overlooked in alternative proteomic analyses), and support the idea that target specificity can be achieved through rational design of S-nitrosothiols
