Comprehensive proteomics for Discovery vaccine candidates against Streptococcus equi Infections

ABSTRACT: Streptococcus equi subspecies equi (S. equi) is a major pathogen which cause strangles, a highly contagious respiratory infection, in horses and other equines. In this study, we purified the extracellular vesicles (EVs) of S. equi ATCC 39506 and evaluated them as vaccine candidates against S. equi infections in mice. Through immunization in an animal model and immunoprecipitation-mass spectrometry, we evaluated EV as vaccine candidates against S. equi infections and identified novel immunogenic proteins.

Project description:Streptococcus equi subspecies equi (S. equi) is a major pathogen which cause strangles, a highly contagious respiratory infection, in horses and other equines.In this study, we discovered potential vaccine candidates using comprehensive proteomics and reverse vaccinology. As the initial step, we divided proteome of S. equi ATCC 39506 into whole cell lysate, secretory proteome, membrane proteome and extracellular vesicle and then, comparative proteomic analysis was performed to characterize the functional features of the proteome. Especially, extracellular vesicle of S. equi was evaluated at the first time. Total 114 potential vaccine candidates (PVCs) were selected using reverse vaccinology and knowledge based annotations. Comprehensive proteomic analysis confirmed that 60 PVCs were identified in S. equi ATCC 39506. Particularly, 32 PVCs were enriched in the EV proteome, suggesting that this cellular fraction may serve as vaccine.

Identification of Amblyomma americanum antigens after vaccination with tick extracellular vesicles in white-tailed deer

Project description:Anti-tick vaccines represent a promising alternative to chemical acaricides for the management of ticks on wildlife; however little progress has been made to produce an effective vaccine in hosts relevant to tick reproduction. Further, most of the antigens described are expressed during feeding in domesticated hosts and may not activate the immune system in wild hosts. We recently tested Amblyomma americanum salivary (SG) and midgut (MG) extracellular vesicles (EVs) as vaccine candidates in white-tailed deer (Odocoileus virginianus; WTD), which resulted in on-host female tick mortality. Herein, we describe our efforts to identify the antigenic proteins found within these vesicles. Within both SG- and MG-EVs, an enriched “core- cargo” was identified within both SG- and MG-EVs, including chaperones, small GTPases, and other proteins previously reported in small EVs was discovered. Label-free quantitative proteomics showed significant differences in protein cargo between MG and SG-EVs (333 proteins out of 516). Serum from three vaccinated WTD (pre-vaccinated and day 57 post vaccination) and one control WTD was used to immunoprecipitated antigenic proteins from SG- and MG-EV preparations. Proteomic analysis of immunoprecipitated proteins identified seven antigenic proteins in SG-EVs and one in MG-EVs that were statistically significantly different from the pre-vaccinated serum. Moreover, these proteins were unique to vaccinated animals (not precipitated by control serums). Additionally, two MG-EVs and 24 SG-EV proteins show antigenic potential. These proteins represent promising candidates for anti-tick vaccine design in WTD and other wildlife.

Mass spectrometry of circulating antigens products isolated from sera of mice infected with Toxoplasma gondii tachyzoites

Project description:Toxoplasma gondii is a ubiquitous apicomplexan protozoan parasite that can infect all warm-blooded animals and lead to toxoplasmosis. Discovering efficient methods for diagnosing T. gondii infection and developing vaccine candidates are essential for controlling this disease. Circulating antigens (CAg) are reliable diagnostic indicators of acute infection and highly immunogenic compounds that induce protective immunity. We aimed to identify the CAg spectrum in infected mouse serum. Among 31 identified CAg, we evaluated the ability of RuvB-like helicase (TgRuvBL1), ribonuclease (TgRNaseH1), and ribosomal protein RPS2 (TgRPS2) to protect against T. gondii infection. These three proteins were found in the serum of infected mice. Vaccination with rTgRuvBL1 or rTgRPS2 elicited powerful humoral and cellular immune responses in mice, as levels of total IgG, IgG2a, IgG1, and CD4+ and CD8+ T lymphocytes were significantly increased. The production of proinﬂammatory gamma-interferon, interleukin-2, and interleukin-12 was also significantly increased. The survival of mice immunized with rTgRuvBL1 (10.0 ± 0.30 days) or rTgRPS2 (11.5 ± 0.34 days) was significantly prolonged after challenge with virulent T. gondii RH strain (P < 0.05 vs. control). The three proteins are potential diagnostic candidates for acute T. gondii and TgRuvBL1 and TgRPS2 might serve as candidate vaccine antigens against toxoplasmosis.

Epigenetic therapy activates TE-chimeric transcripts to provide additional source of antigens in glioblastoma stem cells

Project description:Cancer-specific coding mutations can create neoantigens that can be presented on the cell surface of tumors to trigger immunogenic clearance1–4. However, current cancer vaccine approaches have not been universally effective5; this is especially true in tumors with a low mutational burden which, in turn, carry a low conventional neoantigen load6. Transposable elements (TEs) make up approximately 50% of the human genome and have been discovered to provide cryptic promoters, which can be reactivated with epigenetic manipulations to generate TE-gene chimeric transcripts that can be translated into noncanonical peptides7. Here, we focus on glioblastoma, an aggressive brain cancer with low mutation burden, to explore whether epigenetic therapy can induce TE-chimeric antigens (TEAs) to appreciably increase the antigen repertoire that can be targeted with immunotherapy. We perform comprehensive epigenetic and transcriptomic profiling of three patient-derived glioblastoma stem cell lines (GSCs) and, more importantly, astrocyte and fibroblast primary cell lines that are either proliferating or quiescent, treated with epigenetic therapy drugs to identify treatment-induced TEA (TI-TEA) candidates that are preferentially expressed in cancer cells. Although we verify TI-TEAs are indeed presented on HLA molecules in GSCs thus are promising cancer vaccine candidates, many TEs were also transcriptionally activated in proliferating primary cell lines after epigenetic therapy. This work presents a cautionary but optimistic tale for future efforts in harnessing TI-TEAs for targeted immunotherapy approaches.

A Subtractive Proteomics Approach for the Identification of Immunodominant Acinetobacter baumannii Vaccine Candidate Proteins

Project description:Background: Acinetobacter baumannii is one of the most dangerous multidrug-resistant pathogens worldwide. Currently, 50-70% of clinical isolates of A. baumannii are extensively drug-resistant (XDR) and available antibiotic options against A. baumannii infections are limited. There are still needs to discover specific de facto bacterial antigenic proteins that could be effective vaccine candidates in human infection. With the growth of research in recent years, several candidate molecules have been identified for vaccine development. So far, there is no public health authorities approved vaccine against A. baumannii. Methods: The purpose of this study was to identify immunodominant vaccine candidate proteins that can be immunoprecipitated specifically with patients’ IgGs. Relaying on hypothesis that IgGs of infected person have capacity to capture immunodominant bacterial proteins. Herein, outer membrane and secreted proteins of sensitive and drug resistant A. baumannii were captured by using IgGs obtained from patient and healthy control sera and were identified by LC-MS/MS analysis. Results: By using subtractive proteomic approach, we determined 34 unique proteins which were captured only in drug-resistant A. baumannii strain via patient sera. After extensive evaluation of predicted epitope regions, solubility, membrane transverse characteristics, and structural properties, we selected several notable vaccine candidates. Conclusion: We identified vaccine candidate proteins that triggered de facto response of human immune system against the antibiotic-resistant A. baumannii. Precipitation of bacterial proteins via patient immunoglobulins was a novel approach to identify the proteins which have potential to trigger to response in patient immune system.

Decreased Fc fucosylation aligns with enhanced RSV-specific antibody functionality in adults compared to young children

Project description:Respiratory syncytial virus (RSV) is a major cause of severe respiratory tract infections in infants and older adults. While vaccines have recently been approved for older adults and pregnant women, vaccine development for infants remains challenging due to the risk of vaccine-enhanced disease. Understanding immunological differences between target groups is essential for improving vaccination strategies, but no clear correlates of protection have been identified yet. Antibody Fc-mediated effector functions may play an important role. In a cohort of 24-month old children and adults (n=46 per group), we assessed RSV-specific IgG/IgA levels, IgG subclasses, avidity, Fc glycosylation, and neutralization, antibody-dependent NK cell activation (ADNKA), cellular phagocytosis (ADCP), and complement deposition (ADCD). Compared to children, antibodies from adults showed enhanced functionality. Furthermore, RSV-specific antibodies displayed differences in avidity and glycosylation patterns between the two groups, which might relate to differences in the number of previous exposures. Importantly, our data strongly suggest that IgG Fc afucosylation drives enhanced ADNKA capacity of RSV-specific antibodies. Our data provide a detailed overview of similarities and differences between the RSV-specific antibodies in children and adults. This information will support the ongoing quest for correlates of protection and the design of future vaccination strategies in different target populations.

miRNAs expression profile in goat peripheral blood mononuclear cells (PBMCs) infected with Nigeria 75/1 vaccine virus

Project description:Peste des petits ruminants virus (PPRV) belongs to the genus Morbillivirus that causes an acute and highly contagious disease in goats and sheep. Virus infection can trigger the change in the cellular microRNA (miRNA) expression profile, which play important post-transcriptional regulatory roles in gene expression and can greatly influence viral replication and pathogenesis. Here, we employed deep sequencing technology to determine cellular miRNAs expression profile in goat peripheral blood mononuclear cells (PBMCs) infected with Nigeria 75/1 vaccine virus, a widely used vaccine strain for mass vaccination programs against Peste des petits ruminants (PPR). Expression analysis demonstrated that PPRV infection can elicit 316 significantly differentially expressed (DE) miRNAs including 103 known and 213 novel miRNAs candidates in infected PBMCs at 24 hours post-infection as compared with mock control. Target prediction and functional analysis of these DEmiRNAs revealed significant enrichment for several signaling pathways including TLR signaling pathways, PI3K-Akt, endocytosis, viral carcinogenesis, and JAK-STAT signaling pathways. This study provides a valuable basis for further investigation on the roles of miRNAs in PPRV replication and pathogenesis.

African swine fever virus proteins MGF360-21R and A151R interact with barrier-to-autointegration factor 1 (BANF1)

Project description:African swine fever virus (ASFV) is a large DNA virus causing a highly contagious disease in domestic and wild boar, for which no treatment is available. ASFV vaccine development is hindered by large gaps in knowledge considering virus protein function and virus-host interaction. This study identifies the interaction between two ASFV proteins, MGF360-21R and A151R, and a host barrier-to-autointegration factor 1 (BANF1). We showed that proteins MGF360-21R and A151R can inhibit the induction of the IFN response. Additionally, BANF1 was determined as an important factor in ASFV infection, as the downregulation of BANF1 reduces virus replication. These results uncover a novel function of viral proteins MGF360-21R and A151R and extend our understanding of complex interaction between virus and host, providing insights for developing a vaccine to prevent and control ASFV.

Transcriptome analysis reveals an activation of MHC-I and MHC-II pathways in chicken trachea immunized with infectious laryngotracheitis virus vaccine

Project description:Infectious laryngotracheitis (ILT) is an acute, contagious, upper respiratory disease, which is caused by gallid herpesvirus 1 (GaHV-1). Due to the mortality rates up to 70% depending on the virulence of the virus, it is of economic importance of the disease to explore the etiology of the ILT in the poultry industry. In this study, 15-day-old SPF white leghorn chickens were used to transcriptome analysis in chicken trachea immunized with infectious laryngotracheitis virus vaccine. In conclusion, chicken embryo origin (CEO) vaccine activation of the MHC-I and MHC-II pathways provides insight into the molecular mechanism of immune response in chickens, and holds potential for evaluation and design of new ILT vaccines in a manner adapted to the host immune response to the virus. Ten vaccine inoculated birds were randomly divided in two groups. Each group represents one replication of five pooled tissues, for inoculated birds. Control group consists of five birds that received sterile vaccine diluent.
