Human monocyte-derived macrophages LC-MS/MS

ABSTRACT: We utilized proteomics analysis to ascertain the protein expression profile regulated by MALAT1 of macrophages and find out the protein network of MALAT1 regulation in human macrophages.

Long noncoding RNAs (lncRNAs) are effective regulators of both RNA and protein functions throughout cell biology, including viral replication. Inhibition of BET proteins during adolescence affects prefrontal cortex development: relevance to schizophrenia

Project description:Background: The present study investigated a role of proteins from BET family (epigenetic readers) in schizophrenia-like abnormalities in MAM-E17 model of schizophrenia. Methods: An inhibitor of BET proteins, JQ1, was given during adolescence in postnatal days (P) 23-P30, and behavioral response (sensorimotor gating, recognition memory) and prefrontal cortex (mPFC) function (long-term potentiation (LTP), molecular and proteomic studies) were performed in adult males and females. Results: Deficits in sensorimotor gating and recognition memory were observed only in MAM-treated males. However, adolescent JQ1 treatment affected control, but MAM-treated groups in both sexes. Electrophysiological study showed an LTP impairment only in male MAM-treated animals, and JQ1 did not have any effect on LTP in the mPFC. In contrast, MAM did not affect immediate early gene expression (markers of neuronal and synaptic activity), but JQ1 altered them in both sexes. Proteomic study revealed alterations in MAM-treated groups only in males, while JQ1 affected both sexes. Conclusions: Prenatal MAM administration induced schizophrenia-like abnormalities only in males. In contrast, adolescent JQ1 treatment affected both sexes and induced behavioral changes in control groups, altered a markers of neuronal and synaptic activity and proteomic landscape in the mPFC of both groups (VEH- and MAM-treated). Thus, adolescent inhibition of BET family might change neuroplasticity in the mPFC.

Project description:We were interested in the effects of TLR8 triggering of human monocyte-derived macrophages. We observed a striking upregulation of those genes. This work was done in the context of examining the effects of TLR8 triggering for the activation of HIV. We generated monocyte-derived macrophages (MDM) by initially isolating monocytes and subsequently culturing them in RPMI supplemented with human AB serum. MDM were stimulated for 18 h with the TLR8 agonist 3M-002, and subsequently harvested for extraction of total RNA. Total RNA was then used to quantify the TLR-dependent genes using a PCR array pathway.

E. coli S30 lysate proteome - The E

Project description:Protein production using processed cell lysates, defined precursor cocktails and target specific additives has become a core technology in the field of synthetic biology. Due to their open nature, these artificial systems are excellent to produce difficult proteins such as toxins or membrane proteins. However, the composition of cell lysates as the core component of cell-free systems is still widely a black box. Lysates of Escherichia coli are most productive for cell-free expression, yielding several milligrams of protein per milliliter of reaction. Although proteomics studies of several E. coli strains are available, the preparation of cell-free lysates implies specific fractionation resulting into yet unknown final lysate compositions. Therefore, the identification of prevalent and reliable lysate compounds will be an indispensable prerequisite for directed lysate engineering as an emerging strategy for new applications in synthetic biology. In this study, the S30 lysate proteome from E. coli strain A19 has been analyzed for the first time by using a GeLC-MS/MS approach. A total of 821 proteins were identified in processed S30 lysates. We provide detailed lists of detected proteins relevant for transcription/translation, folding, stability and enzymes involved in metabolic processes. In addition, we also performed a quantitative proteome analysis of modified S30 lysates, generated by induction of the SOS response during cultivation. These altered lysates contain 3-10-fold more folding factors and exhibit improved solubility and folding of difficult proteins. Therefore, the manipulation of the S30 lysate proteome by modified cultivation conditions is an effective approach for the generation of customized cell-free systems.

In line with this, in a 2D DIGE based redox proteomics approach, proteins were identified showing (1) differences in their thiol state upon knockdown of PRDX1 and (2) proteins showing differences in reduded and unreduced samples.

Overexpressed Malat1 drives metastasis through inflammatory reprogramming of lung adenocarcinoma microenvironment

Project description:Although the expression of some long noncoding RNAs (lncRNAs), including MALAT1 (metastasis-associated lung adenocarcinoma transcript 1), is predictive of metastasis, their impact and mechanism of action remain elusive. Here we use CRISPR activation (CRISPRa) to model MALAT1/Malat1 overexpression in patient-derived lung adenocarcinoma (LUAD) cell lines and in the autochthonous K-ras/p53 LUAD mouse model. The results indicate that Malat1 overexpression alone is sufficient to enable the progression of LUAD to metastatic disease. We show that overexpressed MALAT1/Malat1 enhances cell mobility and promotes the recruitment of pro-tumor macrophages to the tumor microenvironment through paracrine secretion of the CCL2/Ccl2 cytokine. We determine that Ccl2 upregulation results from an increase in global chromatin accessibility upon Malat1 overexpression. Importantly, macrophage depletion and Ccl2 blockade counteracted the effects of Malat1 overexpression. These data demonstrate that a single lncRNA can drive LUAD metastasis through reprogramming of the tumor microenvironment.
