LYSET/TMEM251 KO cellular and lysosomal proteomes from MIA PaCa-2 cells

ABSTRACT: Proteome analysis of post-nuclear supernatants (PNS) and enriched lysosomal fractions from LYSET/TMEM251 knockout and control MIA PaCa-2 cells. Lysosomes were loaded with iron nanoparticles (DexoMAG C, Liquids Research). PNS were generated from cell homogenates and used as input to enrich lysosomal fractions over magnetic LS MACS columns (Miltenyi).

The human disease gene TMEM251 is essential for lysosomal transport and viral infection

Project description:After gaining access to the endo-lysosomal pathway, several viruses depend on lysosomal cathepsin proteases to cleave their structural proteins triggering productive entry. Targeting of cathepsins and other luminal lysosomal proteins to lysosomes requires their modification with mannose 6-phosphate (M6P) signals. Key to M6P tagging is N-acetylglucosamine (GlcNAc)-1-phosphotransferase whose deficiency leads to the severe lysosomal storage disorder mucolipidosis II (MLII). Here, using genome-scale CRISPR screens, we identify the transmembrane protein TMEM251 as critically important for viral infection by cathepsin-dependent viruses including reovirus, Ebola virus, and SARS-CoV-2. We demonstrate that Golgi-resident TMEM251 is essential for lysosomal sorting and activity of cathepsins. Mechanistically, we show that TMEM251 deficiency in human cells results in global loss of M6P on luminal lysosomal proteins by destabilizing GlcNAc-1-phosphotransferase. Tmem251 knockout mice reveal characteristics typical of MLII including hypersecretion of lysosomal enzymes and accumulation of lysosomal storage material in isolated fibroblasts. Finally, we demonstrate that human pathogenic TMEM251 alleles fail to rescue lysosomal sorting defects in knockout cells. Our results uncover a crucial role of TMEM251 in M6P-dependent lysosomal transport and viral infection. Overall, work here reveals the biochemical basis of an inherited MLII-like disease caused by mutations in TMEM251 and provides insights into infection mechanisms of a broad class of medically important viruses.

Project description:Lysosomes are key degradative compartments of the cell. Transport to lysosomes ismediated by tagging soluble enzymes with mannose 6-phosphate (M6P) by GlcNAc-1-phosphotransferase whose deficiency leads to the severe lysosomal storage disorder mucolipidosisII (MLII). Several viruses require lysosomal cathepsins to cleave structural proteins and thusdepend on functional GlcNAc-1-phosphotransferase. Using genome-scale CRISPR screens, weidentify LYSET as essential for infection by cathepsin-dependent viruses including SARS-CoV-2. We show that LYSET deficiency results in global loss of M6P tagging and mislocalization ofGlcNAc-1-phosphotransferase to lysosomes. Lyset knockout mice exhibit MLII-like phenotypesand human pathogenic LYSET alleles fail to restore lysosomal sorting defects. Thus, we uncoverLYSET as an indispensable component of the M6P trafficking machinery and reveal thebiochemical basis of an inherited disease caused by mutations in LYSET.

Extracellular proteins enhance Cupriavidus pauculus nickel tolerance and cell aggregate formation

Project description:Heavy metal-resistant bacteria secrete extracellular proteins (e-PNs). However, the role of e-PNs in heavy metal resistance remains elusive. Here Fourier Transform Infrared Spectroscopy implied that N-H, C = O and NH2-R played a crucial role in the adsorption and resistance of Ni2+ in the model organism Cuprividus pauculus 1490 (C. pauculus). Proteinase K treatment reduced Ni2+ resistance of C. pauculus underlining the essential role of e-PNs. Further three-dimension excitation-emission matrix fluorescence spectroscopy analysis demonstrated that tryptophan proteins as part of the e-PNs increased significantly with Ni2+ treatment. Proteomic and quantitative real-time polymerase chain reaction data indicated that major changes were induced in the metabolism of C. pauculus in response to Ni2+. Among those lipopolysaccharide biosynthesis, general secretion pathways, Ni2+-affiliated transporters and multidrug efflux play an essential role in Ni2+ resistance. Altogether the results provide a conceptual model for comprehending how e-PNs contribute to bacterial resistance and adsorption of Ni2+.

Systematic comparison of strategies for the enrichment of lysosomes by data independent acquisition

Project description:We describe a comparison of individual approaches for the enrichment of lysosomes comparing four different strategies using data independent acquisition (DIA). As a benchmark, we chose a crude enrichment strategy, which is the centrifugation of the PNS at 20,000 x g termed organelle enriched pellet (OEP), a two-step density gradient centrifugation (SDGC) approach employing sucrose, histodenz, and percoll, the SPIONs approach, which is based on the endocytosis of dextran coated nano-particles and their delivery to the lysosomal compartment, and the TMEM192-HA immunoprecipitation a 3 x HA tagged version of the lysosomal membrane protein TMEM192 used for lysosome isolation by anti-HA antibodies immobilized on magnetic beads. We evaluated the abundance and distributions of lysosomal, as well total proteins, allowing for the determination of lysosomal enrichment among the different strategies.
