Proteomics Analysis of Saliva from Children with Autism Spectrum Disorder

ABSTRACT: In the current work, saliva samples were collected from children with different degrees of ASD and healthy children and proteomics approaches were applied to generate data on differentially expressed proteins between groups which will serve as a basis for future validation studies as protein markers.

Project description:Systemic and local (oral mucosal) immune responses in acutely infected HIV individuals before the initiation of HAART have not been well characterized. Protein microarrays were used to analyze saliva and plasma from HIV infected and HIV uninfected subjects to identify new biomarkers for HIV disease progression and pathogenesis. A 507 protein microarray was employed to provide a broad view of cytokines and chemokines in saliva and plasma in acutely HIV infected subjects as compared to uninfected subjects. A custom array derived from the initial results was refined to highlight those molecules with significant change relative to control subjects indicating the potential for biological impact.

Top Down Proteomic analysis of Gingival Crevicular Fluid in deciduous, exfoliating and permanent teeth in children

Project description:Gingival Crevicular Fluid, a plasma-derived exudate present in the gingival crevice was collected from deciduous, exfoliating and permanent teeth from 20 children (60 samples) with the aim to characterize and quantify by a mass spectrometry based top-down proteomic approach, the peptide/proteins in the fluid and verify possible variations occurring during the exfoliating process. The results obtained confirmed the presence in Gingival Crevicular Fluid of α-Defensins 1-4, Thymosin β4 and Thymosin β10, as described in previous works and revealed the presence of other interesting peptides never described before in Gingival Crevicular Fluid, such as specific fragments of α-1-antitrypsin, α-1-antichymotrypsin, Thymosin β4 and Thymosin β10 fragments, Fibrinopeptide A, Fibrinopeptide B, S100A, LVV Hemorphin-7 (hemoglobin chain β fragment), as well as some other peptides deriving from α and β subunits of hemoglobin. Statistical analysis evidenced different levels in 5 proteins/peptides in the three groups in particular with higher level in exfoliating teeth. Our study demonstrate that an in-depth analysis of a biological fluid like Gingival Crevicular Fluid, present in small amount, can provide useful information for the understanding of different biological processes like teeth eruption.

The salivary proteome in relation to oral mucositis in autologous hematopoietic stem cell transplantation recipients: a labelled and label-free proteomics approach-DDALFQ

Project description:Decreased salivary flow rates and/or changes in protein composition reported after autologous hematopoietic stem cell transplantation (ASCT) reduces the protective function of saliva. This might be associated with the development of oral mucositis (OM), an inflammation of the oral mucosa as a result of chemotherapy before ASCT which affects patients? quality of life and risk factor for systemic infections. In this study, a TMT-labelled proteomics experiment, a label-free quantification (LFQ) proteomics experiment and a DIA-MS proteomics experiment were used to identify differences in the salivary proteome between patients with ulcerative OM (uOM; WHO score ? 2) and those without (nOM). For the TMT-labelled experiment, saliva samples of 5 uOM and 5 nOM were pooled at different time points: baseline, 1, 2, and 3 weeks after ASCT and 3 months after ASCT. A different salivary proteome was suggested by a distinct clustering of the OM pools at baseline, week 2 and week 3 after ASCT.

Project description:he aim of the project is to characterize qualitatively and quantitatively the interaction of cystatin D in other protein partner and the formation of a complex in whole saliva of patients affected by different forms of systemic mastocytosis (SM), with and without skin involvement. The different composition of the complex could be useful to distinguish patients from healthy subject and the different forms of systemic mastocytosis. Pools of whole saliva from SM (SM-C and SM+C) patients, and from sex/age matched healthy controls were submitted to immunoprecipitation assay using cystatin D antibody followed by SDS-PAGE in reducing conditions, tryptic digestion, and nano-HPLC-high-resolution-MS/MS analysis.

Immunorecognition of Streptococcus mutans secreted proteins protects against caries by limiting tooth adhesion

Project description:Childhood caries is an extremely common childhood chronic disease, affecting 60–90% of children in industrialized countries. It results in lesions in both the primary and permanent dentitions, hospitalizations and emergency room visits, high treatment costs, loss of school days, diminished ability to learn increases the risk of caries in adulthood. Streptococcus mutans is a key bacteria in caries development. While multiple caries risk factors have been identified, significant interpersonal variability not explained by known risk factors still exists. The immune system generates a personal antibody repertoire that helps maintain a balanced and healthy oral microbiome. Using mass-spectrometry, we probed in an hypothesis-free manner which S. mutans proteins are identified by antibodies of children with low and high DMFT (decayed, missing, filled teeth) scores. We identified a core set of proteins, recognized by the immune system of most individuals. This set was enriched with proteins enabling bacterial adhesion, and included glucosyltransferases and glucan-binding proteins known to be important for S. mutans cariogenicity. To explore the physiological relevance of these findings, we tested the ability of saliva from caries free individuals in preventing S. mutans from binding to the tooth surface. Indeed, saliva from individuals with caries free prevented S. mutans binding to teeth. These findings map the S. mutans proteome targeted by the immune system and suggest that inhibiting tooth attachment is a primary mechanism used by the immune system to maintain oral balance and prevent caries. These findings provide new insights into the role of the immune system in maintaining oral health and preventing caries development.
