Proteomic analysis of two populations of Schistosoma mansoni-derived extracellular vesicles: exosome-like vesicles and microvesicles

Project description:Extracellular vesicles (EVs) have been characterized from many species of parasitic helminths, and recent experimental evidence supports important functions for their cargo in host-parasite relationships. Here, we carried out a detailed proteomic analysis of two populations of EVs – exosome like vesicles (ELVs) and microvesicles (MVs) – from Schistosoma mansoni. We profiled the proteins present in these EVs (including external trypsin-liberated peptides, cargo proteins, integral membrane proteins (IMPs) and peripheral membrane proteins (PMPs) using LC-MS/MS.

Proteomic variation in the oral secretion of Spodoptera exigua and Spodoptera littoralis larvae in response to different food sources

Project description:The Spodoptera genus is defined as the pest-rich genus because it contains some of the most destructive lepidopteran crop pests, characterized by a wide host range. During feeding, the caterpillars release small amounts of oral secretion (OS) onto the wounded leaves. This secretion contains herbivore-induced molecular patterns (HAMPs) that activate the plant defense response, as well as effectors that may inhibit or diminish the plant’s anti-herbivory response. In this study, we explored the protein components of the OS of two Spodoptera species, Spodoptera exigua and Spodoptera littoralis.

Sex-biased expression profile of Schistosoma mansoni microRNAs

Project description:Schistosoma mansoni is a dioecious species, that is, it has two differentiated sexes. Interestingly, this sexual species evolved from a hermaphrodite ancestor. Indeed, most Platyhelminthes are hermaphrodites. Here we characterize the microRNAs of S. mansoni and quantify their differential expression between males and females. Mice were infected with Schistosoma mansoni 1-2 weeks prior to dissection. RNA from two independent samples were extracted and sequenced with Illumina MiSeq technology and AB SOLiD 4 technology. Reads were mapped to the reference genome and microRNA detected and analyzed.

After 48hr germ line tissues were removed and RNA was extracted from remaining somatic tissues. Samples were processed for RNAseq using Illumina procedures.

Effect of capacitation on proteomic profile and mitochondrial parameters of spermatozoa in bulls

Project description:Sperm capacitation is a critical process for fertilization. This process involves several biochemical and physiological changes that have been extensively studied, but little is known about the changes that occur in the sperm’s proteomic profile. This work aims to analyze the effect in vitro capacitation had on the proteome and mitochondrial parameters of bull spermatozoa. Samples of non-capacitated (NC) and in-vitro capacitated (ICV) sperm were analyzed using flow cytometry to assess sperm viability, mitochondrial membrane potential (MMP), and reactive oxygen species (mROS). Proteome was evaluated using SWATH-MS. In-vitro capacitation induced a significant decrease in sperm viability, high MMP, and a significant increase in mROS production. Within the group of living spermatozoa, the capacitation process induced a significant decrease in healthy mitochondrial spermatozoa, as well as a significant increase in mROS production, without affecting the MMP intensity

Bacillus thuringiensis Cry5, Cry21, App6 and Xpp55 proteins to control the plant pest nematodes Meloidogyne javanica and M. incognita

Project description:The global imperative to enhance crop protection while preserving the environment has increased interest in the application of biological pesticides. Bacillus thuringiensis (Bt) is a Gramm-positive bacterium that can produce nematicidal proteins and accumulate them in parasporal crystals. Root-knot nematodes are obligate root plant parasitic which are distributed worldwide, causing severe damages to the infested plants and, consequently, large yield reductions. In this work, we have evaluated the toxicity of the crystal proteins Cry5, Cry21, App6, and Xpp55 against two root-knot nematodes belonging to the Meloidogyne genus (M. incognita and M. javanica). The results show that all four proteins, when solubilized, were highly toxic for both nematode species. To check the potential of using Bt strains producing nematicidal crystal proteins as biopesticides to control plant parasitic nematodes in the field, in planta assays were conducted, using two wild Bt strains which produced Cry5 or a combination of App6 and Cry5 proteins. The tests were carried out with cucumber or with tomato plants infested with M. javanica J2, subjected to irrigation with spore+cristal mixtures of the respective strains. The results showed that the efficacy of the nematicidal activity was plant-dependent, as Bt was able to reduce emerged J2 in tomato plants but not in cucumber plants. In addition, the toxicity observed in the in planta assays was much lower than expected, highlighting the challenge of the crystal proteins to exert their toxicity. This emphasizes the delivery of the Bt proteins as crucial for its use to control root-knot nematodes.

Microarray based analysis of temperature and oxidative stress induced messenger RNA in Schistosoma mansoni

Project description:The body’s defense against schistosome infection can take many forms. For example, upon developing acute schistosomiasis, patients often have fever coinciding with larval maturation, migration and early oviposition. As the infection becomes established, the parasite comes under oxidative stress generated by the host immune system. The most common treatment for schistosomiasis is the anti-helmenthic drug praziquantel. Its effectiveness, however, is limited due to its inability to kill schistosomes 2 - 4 weeks post-infection. Clearly there is a need for new anti-schistosomal drugs. We hypothesize that gene products expressed as part of a protective response against heat and/or oxidative stress are potential therapeutic targets for future drug development. Using a 12,166 element oligonucleotide microarray to characterize Schistosoma mansoni genes induced by heat and oxidative stress we found that 1,878 Schistosoma mansoni elements were significantly induced by heat stress. These included previously reported heat-shock genes expressing homologs of HSP40, HSP70 and HSP86. One thousand and one elements were induced by oxidative stress including those expressing homologs of superoxide dismutase, glutathione peroxidase and aldehyde dehydrogenase. Seventy-two elements were common to both stressors that could potentially be exploited in the development of novel anti-schistosomal therapeutics. However, aquatic species often exhibit reduced reproductive success under human care. Different factors, including water temperature, nutrition, and intrinsic genetic and epigenetic elements, influence their fertility. Conventional sperm analyses do not provide insights into the functional competence of semen. Therefore, proteomics is gaining relevance, increasing a better understanding of male physiology. In this study, we investigated the proteomic profiles of seminal plasma and spermatozoa from small-spotted catsharks in two different environments: natural environment and aquarium environment.

Bull sperm SWATH-MS-based proteomics reveals link between high fertility and energy production, motility structures, and sperm-oocyte interaction. Quantitation

Project description:Prediction of male or semen fertility potential remains a persistent challenge that has yet to be fully resolved. This work analyzed several in vitro parameters and proteome of spermatozoa in bulls cataloged as high (HF; n=5) and low field (LF; n=5) fertility after more than a thousand artificial inseminations. Sperm motility was evaluated by Computer-Assisted Sperm Analysis. Sperm viability, mitochondrial membrane potential (MMP), and reactive oxygen species (mROS) of spermatozoa were assessed by flow cytometry. Proteome was evaluated by SWATH-MS procedure. Spermatozoa of HF bulls showed significantly higher total motility than the LF group (41.4% vs. 29.7%). Rates of healthy sperm (live, high MMP, and low mROS) for HF and LF bull groups were 49% and 43%, respectively (p > 0.05). Spermatozoa of HF bulls showed higher presence of differentially abundant proteins (DAPs) related to both energy production (COX7C), mainly OXPHOS pathway, and to the development of structures linked with the motility process (TPPP2, SSMEM1 and SPAG16). Furthermore, we observed that EQTN, together with other DAPs related to the interaction with the oocyte, were overrepresented in HF bull spermatozoa. The biological processes related to protein processing, catabolism, and protein folding were found to be overrepresented in LF bull sperm in which the HSP90AA1 chaperone was identified as the most DAP

Extracellular vesicles from Trypanosoma cruzi-dendritic cell interaction show modulatory properties and confer resistance to lethal infection as a cell-free based therapy strategy

Project description:Extracellular vesicles (EVs) include a heterogeneous group of particles. Microvesicles and exosomes are the most characterised vesicles. They can be distinguished by their size, morphology, origin and molecular composition. To date, increasing studies demonstrate that EVs mediates intercellular communication. EVs reach considerable interest in the scientific community due to their role in diverse processes including antigen-presentation, stimulation of anti-tumoral immune responses, tolerogenic or inflammatory effects. In pathogens, EVs shedding is well described in fungus, bacteria, protozoan parasites and helminths. For Trypanosoma cruzi EVs liberation and protein composition was previously described. Dendritic cells (DCs) are key players promoting the immune response against pathogens and also maintaining self-tolerance. In previous reports we have demonstrate that T. cruzi downregulates DCs immunogenicity in vitro and in vivo. Here we analyze EVs from the in vitro interaction between blood circulating tripomastigotes (Tp) and bone-marrow-derived DCs. We found that Tp incremented the number and the size of EVs in cultures with DCs. EVs displayed some exosome markers and intracellular RNA. Protein analysis demonstrated that the parasite changes the protein-EV profile of DCs. We observed that EVs from the interaction of Tp-DCs were easily captured by unstimulated-DCs in comparison with EVs from DCs cultured without the parasite, and also modify the activation status of LPS-stimulated DCs. Noteworthy, we found protection in animal treated with EVs DCs+Tp and challenged with T. cruzi lethal infection. Our goal is to go deep into the molecular characterization of EVs from the DCs-Tp interaction, in order to identify mediators for therapeutic purposes.
