Palmitoylation proteins in extracellular vesicles

ABSTRACT: The aim of this project is to identify and quantify palmitoylated proteins present in the extracellular vesicles among four experimental groups. Four experimental groups are WT+SHAM, WT+CLP, DEP+SHAM, and DEP+CLP.

Differential Expression of microRNAs following activated protein C treatment in a rat model of septic shock

Project description:Sepsis induces systemic stress by augmenting inflammatory and pro-coagulant responses resulting in microvascular dysfunction and end organ failure, events modulated by the Protein C pathway. MicroRNAs (miRNAs) are small non-coding RNAs involved in post-transcriptional regulation of gene transcription yet their role in sepsis remains poorly defined. We hypothesized that aPC selectively alters the expression of specific miRNAs implicated in protection of hepatic function during septic shock. Male Sprague-Dawley rats underwent sham surgery or cecal ligation and puncture (CLP). Twenty-four later, animals were randomized and treated with aPC (1mg/kg) or vehicle (0.9% (w/v) saline) via an indwelling venous catheter at 12 hour intervals for 24 hours. Gene array was performed on hepatic RNA to determine miRNA expression, and predicted mRNA targets determined using a bioinformatics approach. Of 351 rat miRNAs examined by microarray hybridization, 17 were highly expressed during sepsis and restored to basal levels after aPC treatment. In silico analysis identified 9 miRNAs significantly regulating target genes of the focal adhesion pathway. These data suggest aPC treatment coordinates beneficial cytoprotective effects during sepsis by modulating miRNA expression. While translational effects remain to be fully elucidated in a clinical setting, we demonstrate herein the potential experimental and computational benefits for use of microRNA analysis in sepsis. 12 samples were analyzed. Microarray experiments were performed, in which liver tissue was harvested from variuous groups (Sham+Vehicle, Sham+aPC, CLP+Vehicle, CLP+aPC; n=3/group) and pooled.

Influence of the Soluble Guanylate Cyclase Stimulator Riociguat on Myocardial Left Ventricular Proteome and microRNA expression after Experimental Aortic Stenosis in Mice

Project description:We analyzed global proteomic adaptations during heart failure (HF) progression in a mouse model, suffering from left ventricular pressure overload due to transverse aortic constriction (TAC) and in response to riociguat (RIO) treatment, to gain deeper insights in beneficial effects due to sGC stimulation on cardiac remodeling and HF. TAC and sham animals were treated with either riociguat (RIO; 3 mg/kg/day) or vehicle (VEH; Transcutol®/Cremophor®/water: 10%/20%/70%) for five weeks, starting three weeks post-op when cardiac hypertrophy was established, resulting in four treatment groups ( n=5-6 per group). At the end of the study, hearts were dissected and proteomes of the left ventricles (LV) were analyzed by mass spectrometry (MS).

Project description:The mechanisms by which diesel exhaust particles act as an adjuvant are unknown. We hypothesized that these would be mediated through monocyte interactions with DEP. We sought to identify pathways with a role in inflammation or other signaling mechanisms that might demonstrate the mechanism of response to DEP. Cultured human monocytes from healthy donors were cultured in the presence or absence of diesel exhaust particles. RNA was extracted following culture to determine changes in gene expression pathways.

RNA-SEQ of WT and iMSN-D2RKO Parkinson's mice model treated with L-dopa

Project description:We analyzed the RNA expression profiles of Parkinson's model (6-hydroxydopamine lesion, 6-OHDA) mice in two different genotypes, WT and iMSN-D2RKO mice. Mutant iMSN-D2RKO mice were generated by mating D2Rflox/flox males with D2Rflox/flox females carrying the CRE under the control of the D1 receptor promoter (D1)-Cre. This way, we generated the D2Rflox/flox/D1-Cre/+ line; the D2Rflox/flox mice are thereafter called WT in our study. The D1R gene is expressed during development in MSNs precursors allowing the deletion of D2R as previously shown selectively in iMSNs. The RNA expression profiles were analyzed in three experimental groups: 1) sham-lesioned (sham); 2) 6-OHDA-lesioned (6-OHDA); and 3) 6-OHDA lesioned/L-DOPA-treated (15 mg/kg ). (L-DOPA). Mice of both genotypes were sacrificed 1h after the treatment and brains were flash frozen. Punches of the dorsolateralstriatum(DLS) were obtained on cryostat sections and processed for RNA extraction to prepare the library.

Project description:Expression data from Total RNA extracted from murine spleen. Sepsis was induced in C57Bl/6J mice by cecal ligation and puncture (CLP), followed 6 hours later by an intravenous injection of Mesenchymal Stem Cell (MSC) or saline. Twenty-eight hours after CLP, plasma, bronchoalveolar lavage (BAL) fluid and tissues were collected for analyses. Total RNA was extracted using Trizol (as per manufactures' instruction) followed by clean-up procedure using Qiagen RNA easy Prep (as per manufactures instructions) In the following study we hypothesized that mesenchymal stem cells (MSCs), which have documented immunomodulatory properties, would reduce sepsis-associated inflammation and organ injury in a clinically relevant model of sepsis. To identify the molecular changes associated with decreased inflammation in CLP-injured mice treated with MSCs, we analyzed the gene expression profiles from spleens collected at 28 hours from 4 animals per group: sham/saline, CLP/saline, and CLP/MSCs.

Multisite phosphorylation of intrinsically disordered region of Dishevelled

Project description:When WNT binds to its receptor Frizzled (FZD) to activate the signaling, Dishevelled (DVL), the central component of Wnt signaling, is phosphorylated by redundant Casein kinase 1 (CK1) isoforms ε or δ, to transduce the signal downstream. Although the basic principles of Wnt signal transduction are known, the mechanistic aspects of DVL phosphorylation remain elusive. Here we identify Wnt-dependent multisite phosphorylation of a large and conserved serine/threonine (S/T) cluster within intrinsically disordered region (IDR), located between the PDZ and DEP domains. The detailed analysis of the phosphorylation and its effect on the flanking domains was performed with the construct containing both domains PDZ and DEP flanking the IDR (hDVL3 PDZ-DEP, aa 243-496).
