Project description:Neuroinflammation driven by microglial overactivation is a significant contributor across a diverse range of neurological disorders. While MEF2C mutations have been associated with a syndromic form of autism spectrum disorder (ASD) and various other neurological disorders in humans, the role of MEF2C as a key immune checkpoint to restrain microglial overactivation remains elusive. In this study, we established MEF2C-knockout (KO) induced microglia-like cells (iMGLs) via differentiation of corresponding human pluripotent stem cells, and subsequently treated with LPS, as a disease model of overactivated microglia. Through high-throughput screening, we identified BMS265246, a CDK2 inhibitor, which effectively normalized overactivated phenotypes in LPS-stimulated MEF2C-KO iMGLs, nearing levels seen in WT counterparts. Mechanistically, absence of MEF2C instigated a sequential cascade encompassing p21 downregulation, CDK2 activation, RB phosphorylation and degradation, and NF-κB p65 subunit nuclear translocation, thereby intensifying inflammatory responses. Remarkably, BMS265246 treatment significantly rectified microglial overactivation and ASD behavioral defects in both global and microglia-specific Mef2C heterozygous mice. Overall, our findings elucidate a novel protective mechanism governed by MEF2C, and unveil CDK2 as a promising therapeutic target for treating various diseases influenced by overactivated microglia with aberrant MEF2C expression and/or CDK2 activation.

Project description:Intrinsic immune checkpoints in microglia are vital for maintaining homeostatic immune response and preventing overactivation. Neuroinflammation, frequently driven by microglial overactivation, significantly influences a wide range of neurological disorders. While MEF2C has been associated with a syndromic form of autism spectrum disorder (ASD) and various other neurological disorders in humans, its specific role as an immune checkpoint remains poorly defined. By harnessing MEF2C-knockout (KO) induced microglia-like cells (iMGLs) through differentiation from human pluripotent stem cells (hPSCs), here we observed that these cells, following LPS stimulation, exhibited overactivation similar to patterns seen in various neurological disorders. High-throughput screening identified BMS265246, a CDK2 inhibitor, as effective in specifically suppressing the overactivated phenotypes of MEF2C-KO iMGLs, and in restoring inflammatory response closer to normal levels. Mechanistically, we uncovered that MEF2C regulates p21 transcription, a critical step in preventing CDK2 activation in microglia. Loss of MEF2C results in CDK2-induced RB phosphorylation and degradation, leading to enhanced NF-κB p65 subunit nuclear translocation and exacerbated inflammatory responses. Remarkably, BMS265246 treatment rectified microglial overactivation and ASD-like behaviors in both global and microglia-specific Mef2C heterozygous knockout mice. Overall, our findings elucidated a previously unknown immune checkpoint mechanism governed by MEF2C, and highlighted CDK2 as a potential key factor driving neuroinflammation through microglial overactivation. These insights positions CDK2 as a promising therapeutic target for treating various neurological diseases influenced by overactivated microglia.

Project description:Two genes have a synthetic lethal relationship when silencing or inhibition of one gene is only lethal in the context of a mutation or activation of the second gene. This situation offers an attractive therapeutic strategy, as inhibition of such a gene will only trigger cell death in tumor cells with an activated second oncogene but spare normal cells without activation of the second oncogene. Here we present evidence that CDK2 is synthetic lethal to neuroblastoma cells with MYCN amplification and overexpression. Neuroblastomas are childhood tumors with an often lethal outcome. Twenty percent of the tumors have MYCN amplification and these tumors are ultimately refractory to any therapy. Targeted silencing of CDK2 by three RNA interference techniques induced apoptosis in MYCN-amplified neuroblastoma cell lines, but not in MYCN single copy cells. Silencing of MYCN abrogated this apoptotic response in MYCN-amplified cells. Inversely, silencing of CDK2 in MYCN single copy cells did not trigger apoptosis, unless a MYCN transgene was activated. The MYCN induced apoptosis after CDK2 silencing was accompanied by nuclear stabilization of P53 and mRNA profiling showed up-regulation of P53 target genes. Silencing of P53 rescued the cells from MYCN-driven apoptosis. The synthetic lethality of CDK2 silencing in MYCN activated neuroblastoma cells can also be triggered by inhibition of CDK2 with a small molecule drug. Treatment of neuroblastoma cells with Roscovitine, a CDK inhibitor, at clinically achievable concentrations induced MYCN-dependent apoptosis. The synthetic lethal relation between CDK2 and MYCN indicates CDK2 inhibitors as potential MYCN-selective cancer therapeutics. CDK2 shRNA in a tet repressor system was stably transfected in the IMR32 cell line. Time course analysis was performed in triplicate after induction of CDK2 shRNA at 5 time points.

Project description:Aneuploidy, a cancer hallmark, drives chromosomal instability, drug resistance, and clinically-aggressive tumors. Cyclin-dependent kinase 2 (CDK2) antagonism with independent inhibitors or CDK2 knock-down triggered anaphase catastrophe. This disrupts supernumerary centrosome clustering, causing multipolar division and apoptosis. Time-lapse fluorescent microscopy of FUCCI cell cycle probes transduced into aneuploid lung cancer cells revealed distinct fates of bipolar and polyploid cells after CDK2 inhibition. Apoptosis occurred in multipolar progeny but was repressed in persistent polyploid cancer cells. RNA-seq analyses after CDK2 inhibition of 4N versus 2N lung cancer cells were enriched for CDK1 pathway and KIF family members. The Cancer Genome Atlas (TCGA) analysis of lung cancers indicated CDK1 and KIF family member overexpression was associated with an unfavorable survival. Intravital microscopy of transplanted lung cancer cells in mice extended findings from the in vitro to in vivo settings. CDK2 inhibition of tumor-bearing mice produced polyploid cancer cells in vivo. These cancer cells were resistant to apoptosis and proliferated despite CDK2 inhibition. In contrast, polyploid populations were rarely detected in CDK2 inhibited human alveolar epithelial cells. These findings are translationally relevant. Combined targeting of CDK2 with CDK1 or kinesin family member antagonists should eliminate polyploid cancer cells, promote apoptosis and augment antineoplastic effects.

CRISPR screens identifiy CDK2 loss as a mediator of resistance to INX-315. Furthermore, CDK2 deletion reverses the G2/M block induced by CDK2 inhibitors and restores cell proliferation. Complementary drug screens define multiple means to cooperate with CDK2 inhibition beyond G1/S. These include the depletion of mitotic regulators as well as CDK4/6 inhibitors cooperate with CDK2 inhibition in multiple phases of the cell cycle.
