Till now, the biological effects of combined exposure to EMP and 4.9 GHz RF have not been fully understood. Project description:Chloroplasts are indispensable for higher plants. The growth and development of plants are very sensitive to environmental temperature changes, and chloroplast development is also regulated by adverse environmental temperatures. However, the molecular mechanism of how plants coordinate chloroplast development and environmental temperature changes remains largely unknown. Here, a temperature-conditioned chloroplast development defect mutant tsl2 (thermo-sensitive mutant in leaf color 2) of Arabidopsis was obtained through forward genetic screening. The tsl2 mutant showed a weak yellowing phenotype at 22 °C which was more pronounced when the growth temperature was reduced to 16 °C, and was similar to that of the wild type when the temperature was increased to 29 °C. Genomic resequencing analysis revealed that TSL2 gene encodes the FtsHi5 which was shown to function in chloroplast protein import. Genetic complementation analysis showed that TSL2 rescued the chlorophyll content and thylakoid development defects in tls2 mutants at 16 °C. Quantitative mass spectrometry analysis with Tandem Mass Tag (TMT) isobaric labeling revealed broad changes in the proteome composition of tsl2 chloroplasts at low temperature, reflecting impaired chloroplast biogenesis and function. Together, our data suggest that TSL2 has a significant role in chloroplast development and proteostasis especially at low environmental temperatures.

Here, we used proteomics analyses to reveal the underlying research target in type 2 diabetic mouse model treated with spermine. Left ventricular tissue was excised from 9 mice (Control group, 3; T2D group（db/db mouse）, 3; T2D+SP group, 3) and analyzed. Quantitative analysis of the global proteome were detected using 4D label-free technique,and differentially expressed proteins (DEPs) were used to perform bioinformatic analyses.

Project description:Pseudomonas aeruginosa (P. aeruginosa) can cause severe acute infections, including pneumonia and sepsis, and also cause chronic infections commonly in patients with structural respiratory diseases. However, the molecular and pathophysiological mechanisms of P. aeruginosa respiratory infection are largely unknown. Here, we profiled performed to assay for transposase-accessible chromatin using sequencing (ATAC-seq), transcriptomics, and quantitative mass spectrometry-based proteomics and ubiquitin-proteomics in P. aeruginosa-infected lung tissues for multi-omics analysis, while ATAC-seq and transcriptomics were also examined in P. aeruginosa-infected mouse macrophages. To find the pivotal transcription factors that are likely involved in host immune defense, we integrally investigated systematic changes in chromatin accessibility and gene expression in P. aeruginosa-infected lung tissues combined with proteomics and ubiquitin-proteomics studies. We discovered that Stat1 and Stat3 were altered in various omics and found similar results in mouse alveolar macrophages. Taken together, these findings indicate that these crucial transcription factors and their downstream signaling molecules play a critical role in the mobilization of host immune response against P. aeruginosa infection and may serve as potential targets for bacterial infections and inflammatory diseases, as well as provide clear insights and resources for using integrative histological analyses.

Project description:Background: Importin 7 (IPO7), a karyopherin-β protein, is involved in various tumorigenesis and progression abilities by mediating the nuclear import of oncoproteins. However, the exact biological functions and IPO7 remain to be further elucidated. Materials and Methods: TCGA and GEO datasets were used to identify dysregulated expression of IPO7 in various cancers. Gain-of-function and loss-of-function analyses were used to identify the oncogenic functions of IPO7 in vitro and in vivo. Moreover, LC-MS/MS and parallel reaction monitoring analysis were used to comparatively profiled IPO7-related proteomics and potential molecular machinery. Results: Our works demonstrated that the expression of IPO7 was upregulated and was correlated with poor prognosis in cervical cancer. In vitro and in vivo experiments demonstrated that knockdown of IPO7 inhibited the proliferation of HeLa and C-4 I cells. LC-MS/MS analysis showed that IPO7-related cargo proteins mainly were enriched in gene transcription regulation. Then independent PRM analysis for the first time demonstrated that 32 novel IPO7 cargo proteins, such as GTF2I, RORC1, PSPC1, and RBM25. Moreover, IPO7 contributed to activating the PI3K/AKT-mTOR pathway by mediating the nuclear import of GTF2I in CC cells. Intriguingly, we found that the IPO7 expression was negatively correlated with CD8 T cell infiltration via regulating the expression of CD276 in CC. Conclusion: This study enhances our understanding of IPO7 nuclear-cytoplasmic translocation and might reveal novel potential therapeutic targets. The results of a negative correlation between the IPO7 and CD8 T cell infiltration indicate that the IPO7 might play an important impact in the immune microenvironment of CC.

Project description:Coiled-coil domain-containing 68 (CCDC68) plays different roles in cancer and is predicted as a tumor suppressor in human colorectal cancer (CRC). However, the specific role of CCDC68 in CRC and the underlying mechanisms remain unknown. Here, we showed that CCDC68 expression was lower in CRC than in corresponding normal tissues, and CCDC68 level was positively correlated with disease-free survival. Ectopic expression of CCDC68 decreased CRC cell proliferation in vitro and suppressed the growth of CRC xenograft tumors in vivo. CCDC68 caused G0/G1 cell cycle arrest, downregulated CDK4, and upregulated ITCH, the E3 ubiquitin ligase responsible for CDK4 protein degradation. This increased CDK4 degradation, which decreased CDK4 protein levels and inhibited CRC tumor growth. Collectively, the present results identify a novel CDK4 regulatory axis consisting of CCDC68 and ITCH, which suggest that CCDC68 is a promising target for the treatment of CRC.

Project description:N-glycosylation is an important post-translational modification involved in protein folding, signal transduction, extracellular matrix (ECM) organization and immune response. Recent evidence shows that SARS-CoV-2 Spike protein is highly glycosylated and it may be potential target in viral pathology and drug/vaccine design. Therefore, the N-glycoproteomic profiling of coronavirus and its infected cells are of great importance in therapeutic targets screening for drug discovery. Here, we carried out 4D label-free LC-MS/MS-based N-glycoproteomics using a well-established SARS-CoV-2 cellular model, pangolin GX_P2V virus-infected Vero E6 cells, to study the mechanism of coronavirus infestation and potential drug targets. Meanwhile, we investigated the effect of Cepharanthine (CEP) on viral-induced aberrant N-glycoprotein changes in affected cells and on the viral proteins. The results showed that coronavirus GX_P2V could cause aberrant glycosylation of cell proteins at multiple levels, including extracellular matrix (ECM) and related signal transduction, whereas CEP can maintain 12 out of 69 GX_P2V-induced aberrant glycoproteins at normal glycosylation state. Functional enrichment and PPI analyses revealed that LAMB1 and FN1 were the pivotal proteins in regulating the aberrant glycosylation caused by coronavirus in presence of CEP, indicating that CEP might achieve its therapeutic intervention via these potential targets. Besides, CEP can regulate the glycosylation of viral proteins S, M and N. Nevertheless, there were still 57 out of 69 glycoproteins which cannot be significantly affected by CEP, indicating the combination of CEP with other drugs against the rest of targets should be considered.

Project description:Gametophytic self-incompatibility is the primary cause of low fruit set in almond. The mechanism of recognition that determines whether the gametophyte is successfully fertilized between pollen tube SCF (F-box-SSK1-Cul1-Rbx1) protein and pistil S-RNase protein during fertilization is unclear. In this study, the pistils of two almond cultivars 'Wanfeng' and 'Nonpareil' were used as the experimental materials after selfing- and nonselfing/cross-pollination, and pistils from the stamen-removed flowers were used as controls. We used fluorescence microscopy to observe the development of pollen tubes after pollination and 4-dimensional label-free quantitation (4D-LFQ) to detect the protein expression profiles of 'Wanfeng' and 'Nonpareil' pistils and in controls. The results showed that it took 24-36 h for the development of the pollen tube to 1/3 of the pistil, and a total of 7684 differentially accumulated proteins (DAPs) were identified in the pistil after pollinating 36 h, of which 7022 were quantifiable. The up- and down-regulation of the 12 differential protein expressions identified by parallel reaction monitoring (PRM) was the same as that identified by 4D-LFQ, with an average fold-change difference of 14.34%, a maximum of 31.37% and a minimum of 3.68%. Bioinformatics analysis based on the function of the differential proteins, including classification, enrichment, and clustering, identified RNA polymerases (4 DAPs), autophagy (3 DAPs), oxidative phosphorylation (3 DAPs), and homologous recombination (2 DAPs) pathways associated with the self-incompatibility process. The interaction between the serine/threonine kinase (MARK2) protein E3 ubiquitin ligase and the microtubule protein component microtubule-associated protein tau (MAPT/ACT) was found using the STRING database, which demonstrated the involvement of the MARK2 protein in the reaction of pollen tube recognition the nonself- and the self-S-RNase protein. It provides a new way to reveal the mechanism by which almond pollen tubes recognize the self and nonself S-RNase enzyme protein.
