Mass spectrometry (MS)-based proteomics has recently attracted the attention from forensic pathologists. Project description:Î²-catenin signaling is required for hair follicle development, but it is unknown whether it is sufficient to activate expression of hair follicle genes in embryonic skin. To address this we profiled gene expression in dermis from E15.5 KRT14-Cre Ctnnb1(Ex3)fl/+ embryos carrying an activating mutation in epithelial beta-catenin, and control littermate embryos. Experiment Overall Design: Total dermal RNA from two KRT14-Cre Ctnnb1(Ex3)fl/+ and two control littermate E15.5 embryos was hybridized to Affymetrix GeneChip Mouse Genome MOE430 2.0 oligonucleotide microarrays. Experiment Overall Design: Appended below is Table S2: Full list of differentially expressed genes in KRT14-Cre Ctnnb1(Ex3)fl/+ mutant compared with control littermate dermis at E15.5, including normalization and filter parameters. Proteomic Analysis of Loricrin Knockout Mouse Epidermis

Project description:The cross-linked envelope of the mammalian epidermal corneocyte is important as a scaffold for assembly of the lipid barrier of the epidermis. Illustrating its importance, deficient envelope cross-linking by keratinocyte transglutaminase (TGM1) is a major cause of the autosomal recessive congenital ichthyoses, characterized by barrier defects. Expectations that loss of some of the known envelope protein components would also confer an ichthyosis phenotype have been difficult to demonstrate. To help rationalize this observation with one major component, the protein profile of epidermis from loricrin knockout mice has been compared to that of the wild type. Despite the mild phenotype of the knockout, some 40 proteins have been seen to be incorporated into envelope material to significantly different extents. Nearly half of these proteins were also seen to be incorporated to similarly altered extents into the disulfide bonded keratin network of the corneocyte. The results suggest that loss of loricrin alters their incorporation into envelopes as a consequence of protein-protein interactions during cell maturation. Mass spectrometric protein profiling revealed that keratin 1, keratin 10 and loricrin are prominent envelope components and that dozens of other proteins are also components. This finding helps rationalize the potential formation of functional envelopes despite loss of a single component due to availability of many alternative transglutaminase substrates

Narrow-band UVB phototherapy reverses DNA methylation changes in psoriatic epidermis

Project description:To obtain a comprehensive DNA methylation signature specific for the epidermal component of psoriasis and to analyze methylation changes during therapy, we performed genome-wide DNA methylation profiling of epidermal cells from 12 patients undergoing narrow-band UVB phototherapy and 12 corresponding healthy controls. A distinct DNA methylation pattern was seen in psoriasis compared to controls. An overall hypomethylation was found in psoriasis and 3665 methylation variable positions were identified. The DNA methylation pattern was reversed at the end of phototherapy in patients with clinical response. Bisulphite converted DNA from the 47 samples were hybridised to the Illumina Infinium 450k Human Methylation Beadchip.
