Utilizing HXMS to Uncover Structural Basis of CPC Phase Separation

ABSTRACT: The centromere is the locus that directs accurate chromosome segregation, and a mater regulatory complex ,the chromosome passenger comlex (CPC) is enriched in a region of the mitotic centromre termed the inner centromre. Recent findings support a role of CPC phase separation in its localization and function in mitotic error correction. Currently there are no teqhniques to globaly map the motifs on proteins that drive the multivalent interactions. In this context, we now employ hydrogen/deuterium exchange coupled to mass spectrometry (HXMS) to study dynamics of CPC phase separation.

Identifying transcriptional targets of ONECUT3 by ChIP-seq

Project description:We report the direct target genes of the aberrant expression of ONECUT3 through next generation sequencing. We combine RNA-seq and ChIP-seq to identify target genes of ONECUT3 overexpression. RNA sequencing (RNA-seq) and chromatin immunoprecipitation sequencing (ChIP-seq) assays were performed on TP53-KO mouse embryonic fibroblast MEF with or without Doxycycline (100 ng/ml) to identify target genes both bound and regulated by ONECUT3. We found that genes related with sister chromosome exchange separation, mitotic nuclear division, and chromosome segregation were significantly upregulated by high level of ONECUT3. The top 10 directly activated target genes predicted by Binding and Expression Target Analysis (BETA) include Incenp and Cdca8, which are the critical components of Chromosomal Passenger Complex (CPC). Through this observation, we conclude that main role of ONECUT3 as transcription factors is to upregulate mRNA transcription of mitotic process. This sutdy provide new insight of genetic network of how dysfunctional ONECUT3 underlies mitotic defects and Chromosomal instability.

MKLP2 walks along microtubules and takes the CPC along for a ride

Project description:The mitotic kinesin-like Protein 2 (MKLP2, KIF20A) is essential for the proper execution of cytokinesis and required to ‘passage’ the chromosomal passenger complex (CPC) from the chromosomes in (pro)metaphase to the spindle midzone and equatorial cortex in anaphase. Together with MKLP1 (KIF23) and MPP1 (KIF20B), MKLP2 forms the kinesin-6 subfamily of motor proteins. Whilst MKLP1 and MPP1 are both plus-end directed processive motors, the typical structure of the MKLP2 motor domain along with the extended neck-linker region, suggested that MKLP2 might not be able to function as a transport motor but is more likely to act as a microtubule bundler. This notion contradicts the prevailing hypothesis that MKLP2 can transport the CPC in anaphase, but appears to be in line with in vitro and in cellulo studies showing that recombinant MKLP2 efficiently bundles microtubules, and that knock-down of MKLP2, disturbs the organization of the anaphase spindle midzone. Using TIRF microscopy and purified fluorescent proteins, we here demonstrate that MKLP2 is a plus-end directed processive motor that can transport the CPC along microtubules in vitro. Live imaging of MKLP2::GFP and INCENP::GFP in early anaphase cells revealed that a fraction of both MKLP2 and INCENP displays directed movements towards the (equatorial) cortex. In line, inhibition of MKLP2 ATPase activity at the start of anaphase disturbed the localization of MKLP2 and CPC at the equatorial cortex. Our data suggest that MKLP2 is a processive microtubule-based motor that transports itself and the CPC to the equatorial cortex.

Project description:Sgo1 makes multipartite interactions with CPC subunits Previous studies have suggested that the Sgo1-CPC (Chromosomal Passenger Complex) interaction is mediated via the N-terminal coiled-coil of Sgo1 and Borealin. However, our structural data revealed that the very N-terminus of Sgo1 can interact with the BIR domain of Survivin. Together, these studies suggest that multi-partite interactions between Sgo1 and different CPC subunits could facilitate CPC-Sgo1 complex formation. To gain further structural insights, we performed chemical cross-linking of the CPCISB10-280-Sgo11-415 complex using a zero-length cross-linker, 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide (EDC), followed by mass spectrometry analysis. Cross-linking-mass spectrometry (CLMS) data showed that: (1) consistent with our previous observations, the N-terminal region of Sgo1 (amino acids 1-34) makes extensive contacts with Survivin BIR domain (amino acids 18-89); (2) the N-terminal coiled-coil of Sgo1 (amino acids 10-120) interacts with the CPC triple helical bundle; (3) consistent with previous findings, the N-terminal coiled-coil also contacts the Borealin dimerisation domain; and (4) the Sgo1 region beyond the N-terminal coil-coil region, which is predicted to be unstructured, contacts both Survivin and Borealin with most contacts confined to the Sgo1 central region spanning amino acids (aa) 180-300. Thus, our cross-linking results suggests that Sgo1 interacts with CPC, mainly via two regions, the N-terminal coiled-coil domain and the unstructured central region.

Slx5/8 alleviates replication stress by promoting mitotic progression

Project description:MCM-10 vs. wild type yeast differential analysis using label-free relative quantification and directed mass spectometry using RIPPER. Loss of Mcm10 causes replication stress. We uncovered that S. cerevisiae mcm10-1 mutants rely on the SUMO-targeted E3 ubiquitin ligase complex Slx5/8 and SUMO network for survival. How Slx5/8 suppresses replication stress has remained elusive. We developed a novel intensity-based label-free quantitative mass spectrometry approach and identified SUMO conjugates that were either enriched or depleted in mcm10-1 cells. We reasoned that sumoylated proteins that were decreased in mcm10-1 cells were potential Slx5/8 targets. Such candidates included subunits of the chromosome passenger complex (CPC), Bir1 and Sli15, which we verified to be substrates of the Slx5/8 pathway. CPC is known to positively regulate the spindle assembly checkpoint (SAC), and we show here that the ablation of CPC components and other SAC proteins supports growth of mcm10-1 cells by relieving mitotic arrest. Together, our data provide mechanistic insight into how SUMO and Slx5/8 orchestrate the replication stress response to promote cell survival.

Survivin deficient liver of mice after partial hepatectomy

Project description:The liver possesses remarkable regenerative capacity in response to injury. Upon partial hepatectomy (PHx), terminally differentiated hepatocytes in the remaining liver enter the cell cycle and restore the liver mass and function within weeks. However, liver regeneration is often impaired in livers with chronic diseases. Survivin, an inhibitor of apoptosis protein (IAP) and member of chromosome passenger complex (CPC), plays versatile roles in cell mitosis and apoptosis. We and others found that the expression of Survivin was highly increased in liver during PHx-induced liver regeneration, which indicated that Survivin played important roles in this process. However, the function of Survivin in liver regeneration remains largely undefined. Here, using mice with genetic deletion of Survivin, we found that during PHx-induced liver regeneration Survivin regulated both hepatocyte G1/S phase transition by inhibiting the expression of p21 and G2/M phase transition by regulating the localization of CPC. Moreover, restoration of Survivin expression in Survivin-deficient hepatocytes inhibited p21 expression and promote both hepatocyte G1/S and G2/M transition during PHx-induced liver regeneration.
