The proteomic changes in MCF-7 cells in exposure to cytotoxin

ABSTRACT: In this study, the proteomic changes of MCF-7 cells exposed to different concentrations of cytotoxin were investigated. At different time points, differentially expressed protein families were detected in MCF-7 cells exposed to cytotoxin. At 8 h, the cytotoxin effects mainly associate with the interference of cell cycle and different intracellular pathways. Among these differentially expressed proteins, pro-apoptotic proteins were also highly expressed. At 24 h, it appeared that cytotoxin induced cell death in MCF-7 cells by involving various cell death pathways.

The label-free quantitative secretome of MCF-7 cells treated with sumaCTX

Project description:This study aimed to investigate the alteration of intracellular proteome of MCF-7 cells following membrane permeabilization by sumaCTX at increasing concentration, using a label-free quantitative (LFQ) approach. The secretome represents the leakage of intracellular proteins in MCF- 7 cells were subjected to trypsin digestion and LC-MS/MS. Based on the LFQ secretome, there was a total of 105 distinct abundance proteins (DAPs) identified in different experimental groups. Further bioinformatics analyses revealed that most of these DAPs were confined to cytoplasmic, nucleus, and membrane. The DEPs involved in metabolic pathways, membrane integrity, inflammatory responses, and necroptosis. There were relatively higher detectable levels of DAPs in the secretome treated with a higher concentration of sumaCTX, suggesting prominent membrane permeabilization. The necroptosis was possibly a stress response in MCF-7 upon exposure to the high concentration of sumaCTX.

LC-MS peptide analysis of cytotoxin digested with multiple-proteolytic enzyme

Project description:In this study, multiple enzyme digestion method was used to enhance the peptide sequence coverage of cytotoxin to characterize its epitope properties. This epitope excision method exhibits no preference for either consecutive or assembled epitopes, based on the specific binding between an antibody and cytotoxin. Following the binding of cytotoxin to its respective antibody, the epitope peptide sequences were shielded from enzymatic proteolysis experiment. Thesedigested peptides were then identified by LC-MS.

Biosynthesis of polycyclopropanated high energy biofuels

Project description:Cyclopropanes-functionalized hydrocarbons are excellent fuels. however, their synthesis is challenging and harmful for the environment. In this work we produced polycyclopropanated fatty acids in bacteria. These molecules can be easily converted into renewable fuels for high energy applications such as shipping, long-haul transport, aviation, and rocketry. We explored the chemical diversity encoded in the genome of thousands of bacteria to identify and repurpose naturally occurring cyclopropanated molecules. We identified a set of candidate iterative Polyketide Synthases (iPKSs) predicted to produce polycyclopropanated fatty acids (POP-FAs), expressed these PKSs in Streptomyces coelicolor and produced the POP-FAs. We determined the structure of the molecules and increased their production 22-fold. Polycyclopropanated fatty acid methyl esters (POP-FAMEs) were obtained by methyl esterifying the POP-FAs. Finally, we calculated the enthalpy of combustion of several POP fuel candidates to assess their potential as replacement for fossil fuels in energy demanding applications. Our research shows that POP-FAMEs and other polyketide derived POPs have the energetic properties for energy demanding applications for which sustainable alternatives are scarce.

Comparative proteomics analysis of growth-primed adult dorsal root ganglia reveals key molecular mediators for peripheral nerve regeneration

Project description:Peripheral nerve injuries due to physical insults or chronic diseases are quite common, yet no pharmacological therapies are available for the effective repair of injured nerves. The slow growth rate of adult nerves and insufficient access to growth factors pose major hurdles in timely reinnervating target tissues and restoring functions after nerve injuries. A better understanding of the molecular changes that occur during the immediate regenerative reprogramming of neurons, stated here as in vivo priming, following nerve injury may reveal ideal candidates for future therapies. Hence, molecular profiling of neuronal soma within the first week of nerve injury has been the gold standard for revealing molecular candidates critical for nerve regeneration. A complementary in vitro regenerative priming approach was recently shown to induce enhanced outgrowth in adult sensory neurons. In this work, we exploited the in vitro priming model to reveal novel candidates for adult nerve regeneration. We performed the whole tissue proteomics analysis of in vitro primed DRGs and compared their molecular profile with that of the in vivo primed, and control DRGs. Through this approach, we identified several commonly and uniquely altered molecules in the in vitro and in vivo primed DRGs that have the potential to modulate adult nerve regrowth. We further validated the growth inducing potential of mesencephalic astrocyte-derived neurotrophic factor (MANF), one of the hits identified in our proteomics analysis, in primary adult sensory neurons. Overall, this study showed that in vitro priming partially reproduces the molecular features in in vivo primed adult sensory neurons. The shortlisted candidates presented here from the two priming approaches may serve as potential therapeutic targets for adult nerve regeneration.

Facile metabolic reprogramming distinguishes mycobacterial adaptation to hypoxia and starvation: The role of ketosis in starvation-induced persistence

Project description:Mycobacteria have the ability to adapt to stressful infection conditions by entering a reversible state of non-replicating persistence (NRP) characterized by slow or no cell growth and increased tolerance to various antimicrobial agents. While hypoxia and nutrient deprivation are commonly used to study this NRP state, there is limited understanding of the molecular distinctions in how mycobacteria adapt to these different stresses to achieve a similar NRP outcome. In this study, we conducted a comprehensive analysis of Mycobacterium bovis BCG's response to starvation, shedding light on a coordinated metabolic shift away from glycolysis during nutrient-rich growth to the depletion of lipid stores, lipolysis, and fatty acid ß-oxidation during NRP. Interestingly, this response differs from BCG's NRP state under hypoxia, where it shifts towards cholesterol metabolism and triglyceride storage. Our investigation also unveiled hidden metabolic vulnerabilities in the NRP state induced by starvation, notably an increased sensitivity to H2O2. These findings open up possibilities for the development of precise therapeutic approaches against these otherwise challenging-to-treat pathogens.

Transcription profiling of human post-mortem conjunctivae and corneal epithelia

Project description:Whole human conjunctivae and human corneas were obtained from the National Disease Research Interchange (NDRI, Philadelphia, PA) and processed between 48 - 72 hr of death. No donor details apart from age, sex and cause of death were released. Criteria for inclusion were donor ages between 25 - 65 years old and overt physical integrity of the epithelium over the central cornea as revealed by a quick stain with 0.1 % Trypan blue. For each replicate experiment (Conjunctiva 1 and Cornea 1; Conjunctiva 2 and Cornea 2;Conjunctiva 3 and Cornea 3), RNA from two different corneal or conjunctival specimens was pooled and converted into cDNA. Appropriately fragmented biotin-labeled cRNA was hybridized to the HG-U133A GeneChips® (Affymetrix) oligonucleotide microarray following the manufacture protocol. Three separate experiments each including one conjunctival and one corneal cRNA pool were performed (Conjunctiva 1 and Cornea 1; Conjunctiva 2 and Cornea 2;Conjunctiva 3 and Cornea 3).

Alterations in tumour necrosis factor signaling pathways associated with cytotoxicity and resistance to taxanes in tumour cells.

Project description:The taxanes are widely used in the treatment of breast and other cancers. While their cytotoxicity has been attributed to the induction of cell cycle arrest in mitosis through the stabilization of microtubules, we found that docetaxel promotes soluble tumor necrosis factor alpha (TNF-alpha production in MCF-7 breast tumor cells. TNF-alpha induces apoptotic cell death in a variety of cell types by binding to one of its receptors (TNFR1) which promotes death-inducing signaling complex (DISC) formation. Consistent with this view, we also report that selection of MCF-7 cells for survival in increasing concentrations of paclitaxel or docetaxel results in selection of drug-resistant variants that are resistant to TNF-alpha cytotoxicity. MCF-7 cells selected to 3-5 nM docetaxel produced >30-fold more TNF-alpha than control MCF-7CC cells but had strongly reduced levels of TNFR1. In contrast, expression of TNFR2 was unchanged, resulting in enhanced cell survival through the activation of the NF-kappaB p50 and p65 subunits. Gene expression profiling of docetaxel resistant MCF-7 cells compared to parental MCF-7 cells was performed for the changes of TNF related genes, and also confirmed in ovarian cell line A2780. Two docetaxel resistant cell lines of breast MCF-7 and ovarian A2780 were generated for gene expression profilling. Two colour microarray of Agilent whole human genome nucleotide arrays was conducted with two replicate of both forward and reverse labellings for MCF-7. Four arrays were used for this experiments. And it was four replicate of both forward and reverse labellings for A2780 cells. Eight arrays were used for this experiments
