Specific milk composition in miR-30b-transgenic mice associated to offspring early gut maturation with lasting consequences on their growth

Project description:Milk composition is complex and includes numerous components essential for the offspring growth and development. Besides a high abundance of the miR-30b microRNA, milk produced by the transgenic mouse model of miR-30b-mammary deregulation displays significant changes in its fatty acid profils. Moreover, wild-type adopted pups fed with this milk present an early growth defect. Therefore, the consequences of miR-30b milk feeding on neonate gut development, a prime target of suckled milk, were investigated, along with further characterization of changes in milk composition. A broad characterization of the duodenum of wild-type pups fed with miR-30b milk was performed, using histological, transcriptomic, proteomic and intestinal permeability analyses. Milk of miR-30b foster dams was extensively analyzed using proteomic, metabolomic and lipidomic approaches and hormonal immunoassays. Pups fed with miR-30b milk showed a maturation of their gut tissue, presenting an earlier reduction in paracellular and transcellular permeability at postnatal day 5. MiR-30b milk displayed significant changes in its total lipid content, ceramides and sphingomyelin concentrations, an overabundance in nine proteins and an increase in insulin and leptin levels. These molecules were associated with neonatal gut integrity and maturation, notably by acting on tight junctions. Their significant changes in miR-30b milk could be clearly involved in the early intestinal closure phenotype of the pups, in connection with the observed early growth defect. Further investigations are now needed to determine their specific mode of action, with the aim to modulate infant diet in regard with a benefic effect on growth and health.

Transcription profiling by array of mouse BALBc strain mesenteric lymph nodes after response to common allergens.

Project description:In this study, MLN transcriptome profiling of female BALB/c mice was conducted in order to explore the molecular mechanism(s) of antigen sensitization using three of the most common food allergens; BLG, OVA and PNA. Analysis of cDNA microarray data revealed a complex network of genes that is modulated during BLG, OVA or PNA sensitization. Results from this study implicate the role of differentially expressed genes in the pathogenesis of food allergy. Results from this study will also help better understanding the molecular mechanism of food allergy in animal models and may help in developing new drug target for the therapy of food allergy. This study is likely to add a new dimension in the development of biomarker genes for the determination of potential allergenicity of genetically modified (GM) foods.

Discovery and quantification of non-human proteins in human milk

Project description:The question whether and which non-human peptides or proteins are present in human milk was raised many decades ago. However, due to cross-reactivity or nonspecific antibody recognition, the accuracy of detection by immunochemical methods has been a concern. Additionally, the relative low-abundance of non-human peptides/proteins in the complex milk sample makes them a challenging target to detect. Here, by deep proteome profiling we detected several non-human peptides, which could be grouped as non-human proteins. We next estimated their concentration in human milk by combining data-dependent shotgun proteomics and parallel reaction monitoring. First, we fractionated human milk at the protein level by using sodium dodecyl sulfate polyacrylamide gel electrophoresis followed by reduction, alkylation and in-gel digestion. The resulted peptide mixtures were subjected to proteomics analysis. We were able to detect 1577 human proteins in human milk. Additionally, we identified 109 non-human peptides, of which 71 were grouped in to 36 non-human proteins. In the next step, we targeted 37 non-human peptides and 9 of them could be repeatedly quantified in human milk samples. Peptides/proteins originating from bovine milk products were the dominant non-human proteins observed, notably bovine caseins (α-S1-, α-S2-, β-, κ-caseins) and β-lactoglobulin.

Heat-treatments affect protease activities and peptide profiles of ruminants’ milk

Project description:Proteases present in milk are heat-sensitive, and their activities increase or decrease depending on the intensity of the thermal treatment applied. The thermal effects on the protease activity are well-known for bovine milk but poorly understood for ovine and caprine milk. This study aimed to determine the nonspecific and specific protease activities in casein and whey fractions isolated from raw bovine, ovine and caprine milk collected in early lactation, and to determine the effects of low-temperature, long-time (63°C for 30 min) and high-temperature, short-time (85°C for 5 min) treatments on protease activities within each milk fraction.

Project description:Streptomyces bingchenggensis is a soil bacterium that produces a family of macrolide antibiotics, milbemycins, which is commercially important in crop protection, human and veterinary medicine. After the complete genome sequence, and annotation, for further development of our gene expression approach to biosynthesis, we have employed whole genome microarray expression profiling as a discovery platform to obtain improved specificity and sensitivity of gene expression analysis, allowing a global and at the same time detailed picture of how gene clusters for secondary metabolism are modulated. In the result, we confirmed the expression mil and nan gene cluster, furthermore, pks3, pks5 and nrps7, nrps8 also showed significant gene expression, but no obvious products detected. In Streptomyces bingchenggensis, there are also corresponding genes belonging to Defense mechanisms, which is much more than other Streptomyces, for the resistance of own metabolites and dealing with complex environmental factors. 16 time points were designed at 12, 18, 24, 30, 33, 39, 42, 45, 51, 57, 60, 69, 72, 96, 120, and 144 hours. The samples were collected at each time point named L1, L3, L5, L7, L8, L10, L11, L12, L14, L16, L17, L20, L21, L22, L23, and L24.
