Proteomic analysis of the secretome of a esxaA mutant Mycobacterium tuberculosis strain

ABSTRACT: Extracellular Mycobacterium tuberculosis (Mtb) aggregates can evade phagocytosis and intracellular host-cell defenses by inducing macrophage killing. We showed that Mtb aggregates require a functional ESX-1 type VII secretion system to induce cell death upon contact with macrophages. We used quantitative proteomics to compare the secretome of a esxA mutant strain with the wild type reference to identify secreted bacteria factors involved in macrophage death induction.

Project description:Extracellular Mycobacterium tuberculosis (Mtb) aggregates can evade phagocytosis and intracellular host-cell defenses by inducing macrophage killing. We showed that Mtb aggregates require a functional ESX-1 type VII secretion system and production of the surface-exposed lipid phthiocerol dimycocerosate (PDIM) to induce cell death upon contact with macrophages. We used quantitative proteomics to compare the secretome of a panel of Mtb mutant strains that induce or do not induce macrophage death and identified secreted bacteria factors involved in macrophage death induction.

The global transcriptional profile of Mycobacterium tuberculosis during human macrophages infection

Project description:During lung infection Mycobacterium tuberculosis (Mtb) resides in macrophages and subverts the bactericidal mechanisms of these professional phagocytes. In this work we have analyzed by DNA microarray technique the global transcription profile of Mtb infecting primary human macrophages in order to identify putative bacterial pathogenic factors that can be relevant for the intracellular survival of Mtb. Keywords: time course We compared the global gene expression of the H37Rv strain of Mtb after 4 hours and 24 hours of infection of human macrophage-like THP-1 cells with the gene expression profile of the strain growing exponentially in broth cultures.

Sigma factor E of Mycobacterium tuberculosis controls the expression of bacterial components that modulate macrophages

Project description:Sigma factor E (SigE) controls the expression of genes that are essential for Mtb virulence. In this work, we have identified the SigE regulon during infection of macrophages Our results indicate that SigE regulates the expression of genes involved in the maintenance of Mtb cell envelope integrity and function (i.e., detoxification and secretion). Keywords: strains comparison We compared the global gene expression of the H37Rv strain and the sigE mutant strain of Mtb in different conditions: growing exponentially in liquid media; after 2h of incubation in RPMI; or after 24 hours of infection of human macrophage-like THP-1 cells.

Genome-wide maps of H3K4me1 in Mycobacterium tuberculosis H37Rv-infected macrophages

Project description:Host macrophage transcriptional responses to intracellular pathogens remain poorly characterized. We screened transcriptional enhancers engaged in response to M. tuberculosis (Mtb) infection by ChIPseq analysis of histone H3 lysine 4 monomethylation (H3K4me1). De novo monomethylation during infection was associated with genes implicated in host defense and apoptosis. These regions were enriched for binding sites for ETS transcription factor family members and response elements for nuclear receptors, including liver X receptors (LXRs) and peroxisomal proliferator activated receptors (PPARs), many of which were encompassed by transposable elements. LXRa expression was strongly induced by infection, whereas that of PPARs was unaffected. LXR DNA binding and NCoR corepressor recruitment increased proportionately in infected cells but coactivator association was unchanged, consistent with a lack of induction of endogenous agonists. However, treatment of infected cells with LXR agonist T0901317 strongly increased coactivator recruitment and induced a gene expression program characterized by enhanced innate immune signaling and lipid metabolism. Remarkably, T0901317 treatment selectively induced apoptosis in infected macrophages, and was accompanied by Mtb death, reducing mycobacterial burden 18-fold relative to vehicle 5d after infection. These studies define macrophage transcriptional responses to Mtb infection, and suggest that tissue-specific LXRa agonists may be efficacious in clinical management of tuberculosis.

The adaptation of Mtb to the intracellular life depends on its ability to regulate the expression of its genes. Among the most important bacterial transcription activators are the sigma factors that bind to the RNA polymerase and give it promotor specificity. Sigma factor E (SigE) controls the expression of genes that are essential for Mtb virulence. Analysis of the macrophage transcriptional response indicated that proteins encoded by the sigE regulon are involved in the modulation of the macrophage inflammatory response. We compared the global gene expression of mouse bone marrow macrofages infected with H37Rv and SigE to the gene expression profile of the uninfected macrophages.

Ample glycosylation in membrane and cell envelope protein explains phenotypic diversity, virulence and AMR in the Mycobacterium tuberculosis complex

Project description:Glycoproteomics is likely to identify Mtb virulence factors because glycoproteins on the bacterial cell envelope are used by mycobacteria to enter the primary human host cell, the macrophage. It has been proposed that Mtb interacts with mannose receptors on host cells via mannosylated proteins to enter the macrophages. Despite the vital importance of these proteins in Mtb pathogenesis, our current knowledge of Mtb glycoproteins is still limited, and only a few secreted and cell wall-associated glycoproteins have to date been described. Previous studies have used laboratory strains as model systems to study glycosylation in Mtb. However, only a few sub-groups within the genetically conserved MTBC appear to cause extensive outbreaks with different clinical presentation and AMR. In this study, we employed qualitative and quantitative mass spectrometry and bioinformatics to explore the glycoproteomic patterns of clinical isolates from four lineages of the MTBC, lineages 3, 4, 5 and 7, to investigate the role of protein glycosylation in Mtb adaptation, survival and AMR.

Transcriptional changes in cells upon exposure and during internalisation of aggregating peptides

Project description:The goal of the study is the analysis of the transcriptional adaptation of cultured cells to the exposure and uptake of peptide aggregates of different sizes. Synthetic aggregating peptides with low and high aggregation propensities that form small (<500 nm, Peptide PepS) versus large (>1 µm, PepL) aggregate size distributions were designed. We found that soluble peptides and small aggregates (less than 500 nm diameter) were taken up by non-specific endocytosis as part of the fluid phase and travelled through the endosomal compartment to lysosomes. By contrast, aggregates bigger than 1 µm in diameter, which could not be internalized along with the fluid phase, were taken up through a mechanism dependent of cytoskeletal reorganization and membrane remodelling with the morphological hallmarks of phagocytosis. The microarray analysis aims to determine what's the impact of these different mechanisms of aggregate endocytosis on the transcriptional activity of the cell.

Infection of bone marrow derived macrophages with M. tuberculosis CDC1551

Project description:I infected 8 flasks of resting murine bone marrow derived macrophages with M. tuberculosis CDC1551. Individual samples of intracellular Mtb were collected at 2hr, and days 2,4,6,8,10,12, and 14 post-infection using the GTC (guanidine thiocyanate) method as previously published. A 2hr "no macrophage" control was used as a common denominator in the two-color hybridizations. I have two independent biological replicates of this experiment. Groups of assays that are related as part of a time series. Time: culture time post-infection time_series_design
