Proteomic identification and analysis of FoAPY1 transgenic tomato plants

ABSTRACT: In this study, we obtain FoAPY1 transgenic tomato plants (2417-GFP) and control plants (GFP). All target tomato plants were identified by WB using anti-GFP antibody and all transgenic tomato plants has no differences in morphological and growth rate compared with wild type tomato plants. In order to detect the potential functional mechanism of FoAPY1 protein in the host plant, the proteomic analysis was performed to analyze differentially abundant proteins in two type tomato plants

Global regulation of cell development and aflatoxin biosynthesis in Aspergillus flavus by Afper1

Project description:Aspergillus flavus contaminates crops during preharvest and post-harvest periods and produce carcinogenic mycotoxin aflatoxins posing severe threat to food safety and human health. To identify potential targets to tackle aflatoxin contamination, we have characterized a novel Afper1, sharing sequence identity with the yeast protein per1, regulating cell development and secondary metabolism in A. flavus. Notably, proteomics analysis revealed that the enzymes involved in extracellular hydrolases, conidial development, ER homeostasis, and aflatoxin biosynthesis significantly varied. Unexpectedly, enzymes participated in scavenging ROS including catA, catB, cpeB, and sodM were significantly downregulated and the accumulated ROS and sensitivity to hydrogen peroxide were confirmed by experiments. Surprisingly, Afper1 deletion significantly unregulated heterochromatin protein (HepA) and downregulated GNAT family acetyltransferase involved in heterochromatin formation. Accompanying the accumulated ROS and chromatin remodelling, enzymes participating in aflatoxins, ustiloxin B, and gliotoxin were impaired. These results implied that Afper1 affected chromatin remodelling and disturbed ER homeostasis leading to the accumulation of ROS, and ultimately resulted in growth defect and impaired secondary metabolites biosynthesis.

Global transcriptome profiling of AtJUB1-OX and WT fruits at mature green (MG) and breaker+7 developmental stages.

Project description:To understand the AtJUB1 mediated regulation of tomato fruit development. To test how the NAC transcription factor JUNGBRUNNEN1 from Arabidopsis thaliana (AtJUB1, identical to ANAC042; At2g43000) affects the development of tomato fruits, AtJUB1-GFP was expressed in transgenic tomato plants under the control of the CaMV 35S promoter. AtJUB1-GFP fusion protein was previously shown to encode a functionally active transcription factor in Arabidopsis (Wu et al., 2012). We then performed gene expression profiling of fruits from AtJUB1-GFP lines (AtJUB1-OX plants) and non-modified wild-type (WT) plants. Fruits at two developmental stages, namely mature green (MG) and Breaker+7 days (B+7), were analysed. To this end, the pericarp was separated from the rest of the fruits and quickly frozen in liquid nitrogen for subsequent RNA extraction. Gene expression analysis was performed using GeneChip Tomato Genome Arrays (Affymetrix). Probe preparation and microarray hybridizations were performed by ATLAS Biolabs (Berlin, Germany).

Project description:Winged bean (WB), Psophocarpus tetragonolobus, is a tropical legume, the potential of which is not yet been understood. We found that a 5 week-oral administration of WB seed extract inhibited wrinkle formation induced by repeated tape stripping (TS), which is a model of lichenification in human chronic eczematous dermatitis. To elucidate mechanism of the effect of WB on this model, we applied microarray analysis. Skin barrier was disrupted by repeated application and removal (tape stripping; TS) of cellophane tape on the dorsal skin of the left side. This procedure was started after 1 week-administration of winged bean (WB) extract, and was conducted 3 times per week for 4 weeks. Dorsal skin samples from three experimental groups were used for microarray experiment. These groups were the non-TS (NT), TS, and TS with oral administration of WB extract (TS/WB). The DNA microarray experiment was performed using Affymetrix Mouse Genome 430 2.0 Array.

RNA seq analysis of pathogen-induced transcriptome changes in tomato plants overexpressing the Arabidopsis ELP4 gene

Project description:The goal of the RNA seq was to investigate the transcriptome changes induced by Pseudmonas syringae pv. tomato J4 in wild-type tomato 'Moneymaker' and transgenic 'Moneymaker' overexpressing the Arabidopsis ELP4 (AtELP4) gene. Results showed that P. syringae pv. tomato J4 induced dramatic transcriptional changes in both the wild-type and transgenic tomato plants. Interestingly, a group of defense genes including PR-5x, Pti5, PR1b1, and CHI3/9/14/17, which are associated with resistance to the hemibiotrophic bacterial pathogen Ralstonia solanacearum, were induced to higher levels in the AtELP4 transgenic tomato than in the wild type at 8 and 24 hr after P. syringae pv. tomato J4 infection. These results indicate that overexpression of AtELP4 in tomato leads to faster and/or stronger induction of some defense genes.

Transcriptome profiling of RNAi-trasgenic Solanum lycopersicum plants resistant to Tomato yellow leaf curl virus

Project description:RNA interference (RNAi) is a widely-used approach to generate virus-resistant transgenic crops. However, durability of RNAi-mediated resistance under extreme field conditions and side-effects of stable RNAi expression have not been thoroughly investigated. Here we performed field trials and molecular characterization of two RNAi-transgenic Solanum lycopersicum lines resistant to Tomato yellow leaf curl virus (TYLCV) disease, the major constraint for tomato cultivation in Cuba and worldwide. In order to determine potential impact of the hairpin RNA transgene expression on tomato genome expression and development, differences in the phenotypes and the transcriptome profiles between the transgenic and non-transgenic plants were examined. Transcriptome profiling revealed a common set of up- and down-regulated tomato genes, which correlated with slight developmental abnormalities in both transgenic lines.
