Comparative Thyroglobulin Interactome Profiling

Project description:Thyroglobulin (Tg) is a secreted iodoglycoprotein serving as the precursor for T3 and T4 hormones. Many characterized Tg gene mutations produce secretion-defective variants resulting in congenital hypothyroidism (CH). Tg processing and secretion is controlled by extensive interactions with chaperones, trafficking, and degradation proteins comprising the secretory proteostasis network. While dependencies on individual pro-teostasis network components are known, the integration of proteostasis pathways mediating Tg protein quality control and the molecular basis of mutant Tg misprocessing remain poorly understood. We employ a multiplexed quantitative affinity purification–mass spectrometry approach to define the Tg proteostasis interactome and changes between WT and several CH-variants. Mutant Tg processing is associated with common imbalances in proteostasis engagement including increased chaperoning, oxidative folding, and routing towards ER-associated degradation components, yet variants are inefficiently degraded. Furthermore, we reveal mutation-specific changes in engagement with N-glycosylation components, suggesting distinct requirements of one Tg variant on dual engagement of both oligosaccharyltransferase complex isoforms for degradation. Modulating dysregulated proteostasis components and pathways may serve as a therapeutic strategy to restore Tg secretion and thyroid hormone biosynthesis.

Changes in golden Syrian hamster HDL proteome caused by altered cholesteryl ester transfer protein expression

Project description:Cholesteryl ester transfer protein (CETP) transfers cholesteryl ester (CE) and triglyceride (TG) between lipoproteins, which alters lipoprotein metabolism. Hamster and human CETPs have very different preferences for CE versus TG as substrate. To assess the impact of altering CETP’s substrate preference on lipoproteins in vivo, human CETP was expressed in hamsters (Mesocricetus auratus). Chow-fed hamsters received adenoviruses expressing no CETP (Ad-Null), hamster CETP (Ad-hamster CETP) or human CETP (Ad-human CETP). High density lipoproteins were isolated from hamster plasma 6 days after adenovirus injection by ultracentrifugation as the 1.063 - 1.21 g/ml density fraction. HDL proteins were precipitated with cold acetone and subjected to LC+MS/MS analysis

Alpha-Synuclein pathology and reduced neurogenesis in the olfactory system affect olfaction in a mouse model of Parkinson's disease

Project description:Parkinson’s disease (PD) is characterized by multiple symptoms including olfactory dysfunction, whose underlying mechanisms remain unclear. Here, we explored pathologic changes in the olfactory pathway of transgenic (Tg) mice of both sexes expressing the human A30P mutant α-synuclein (α-syn; α-syn-Tg mice) at 6–7 and 12–14 months of age, representing early and late-stages of motor progression, respectively. α-Syn-Tg mice at late stages exhibited olfactory behavioral deficits, which correlated with severe α-syn pathology in projection neurons (PNs) of the olfactory pathway. In parallel, olfactory bulb (OB) neurogenesis in α-syn-Tg mice was reduced in the OB granule cells at six to seven months and OB periglomerular cells at 12–14 months, respectively, both of which could contribute to olfactory dysfunction. Proteomic analyses showed a disruption in endocytic and exocytic pathways in the OB during the early stages which appeared exacerbated at the synaptic terminals when the mice developed olfactory deficits at 12–14 months. Our data suggest that (1) the α-syn-Tg mice recapitulate the olfactory functional deficits seen in PD; (2) olfactory structures exhibit spatiotemporal disparities for vulnerability to α-syn pathology; (3) α-syn pathology is restricted to projection neurons in the olfactory pathway; (4) neurogenesis in adult α-syn-Tg mice is reduced in the OB; and (5) synaptic endocytosis and exocytosis defects in the OB may further explain olfactory deficits.

Molecular recognition of thyroglobulin by sortilin

Project description:Sortilin is a ubiquitous membrane receptor mediating protein trafficking along different cellular routes. In the thyroid gland, sortilin was proposed as a post-endocytic receptor of the thyroid hormone precursor, thyroglobulin (TG). It was reported that sortilin preferably shuttles iodinated TG to the lysosome, thereby facilitating the proteolytic release of thyroid hormones. However, the molecular details of this interaction are unknown. In this work, using an integrative approach, we reveal the structural basis of human TG recognition by sortilin. We found that sortilin interacts with a TG C-terminal peptide, which is more exposed in the monomeric TG than in its commonly found dimeric form. Unlike previously hypothesised, we suggest that the TG iodination state does not affect interaction with sortilin, which could act as a scavenger for degraded TG, as proposed for other cargoes. By comparing the mode of binding of TG and other known sortilin partners, we then derived a consensus ligand recognition motif. Overall, this study broadens understanding on the role of sortilin in TG trafficking and provides insight into general signatures for sortilin cargoes in other tissues.

The structure of human thyroglobulin

Project description:Thyroglobulin protein is the precursor of thyroid hormones, which are essential for growth, development and control of metabolism in vertebrates. Hormone synthesis from thyroglobulin (TG) occurs in the thyroid gland via the iodination and subsequent coupling of pairs of tyrosine amino acids, whose positions in TG have not been clearly identified. Tyrosine proximity within TG is thought to enable the coupling reactions but the lack of a three-dimensional structure of TG has prevented mechanistic understanding. Here we determined the structure of full-length human thyroglobulin at ~3.4 Å resolution by cryo-electron microscopy (cryo-EM). To enable perturbation experiments we expressed human TG in human HEK cells and showed it to be active in hormone production. We identified all hormogenic tyrosine pairs in the structure and verified them via site-directed mutagenesis and in vitro hormone production assays. Analysis revealed that proximity, flexibility and solvent exposure of the tyrosines are key characteristics of the hormogenic sites. To support the validity of our insights, we engineered the small bacterial protein MBP (maltose binding protein) to produce thyroid hormones with similar efficiency as TG. Finally, our study provides an essential framework to further understand the production and regulation of thyroid hormones, including in thyroid diseases.

Conformational stability of Syrian hamster prion protein PrP(90-231).

Project description:Many transmissible spongiform encephalopathies (TSEs) are believed to be caused by a misfolded form of the normal cellular prion protein (PrP(C)) known as PrP(Sc). While PrP(Sc) is known to be exceptionally stable and resistant to protease degradation, PrP(C) has not shown these same unusual characteristics. However, using ion mobility spectrometry mass spectrometry (IMS-MS), we found evidence for at least one very stable conformation of a truncated form of recombinant PrP(C) consisting of residues 90-231, which resists unfolding in the absence of solvent at high injection energies and at temperatures in excess of 600 K. We also report the first absolute collision cross sections measured for recombinant Syrian hamster prion protein PrP(90-231).

Cardiopulmonary Injury in the Syrian Hamster Model of COVID-19.

Project description:The Syrian hamster has proved useful in the evaluation of therapeutics and vaccines for severe acute respiratory syndrome-coronavirus-2 (SARS-CoV-2). To advance the model for preclinical studies, we conducted serial sacrifice of lungs, large pulmonary vessels, and hearts from male and female Syrian hamsters for days 1-4, and 8 post-infection (dpi) following infection with a high dose of SARS-CoV-2. Evaluation of microscopic lung histopathology scores suggests 4 and 8 dpi as prime indicators in the evaluation of moderate pathology with bronchial hyperplasia, alveolar involvement and bronchiolization being key assessments of lung disease and recovery, respectively. In addition, neutrophil levels, red blood cell count and hematocrit showed significant increases during early infection. We present histological evidence of severe damage to the pulmonary vasculature with extensive leukocyte transmigration and the loss of endothelial cells and tunica media. Our evidence of endothelial and inflammatory cell death in the pulmonary vessels suggests endothelialitis secondary to SARS-CoV-2 epithelial cell infection as a possible determinant of the pathological findings along with the host inflammatory response. Lastly, pathological examination of the heart revealed evidence for intracardiac platelet/fibrin aggregates in male and female hamsters on 8 dpi, which might be indicative of a hypercoagulative state in these animals.
