Project description:Pigment particles used in tattooing may exert long terms effect by releasing diffusible degradation products. In the present work, aqueous suspensions of the organic orange pigment Po13 were aged by exposure to simulated sunlight at 40°C. The morphology and the surface charge of Po13 particles were barely modified upon aging, but their size was reduced. Soluble photoproducts were detected in the liquid fractions. One of this photoproduct (DCBP) was produced in large amount in suspension in isopropanol and purified. The toxicological profiles of aged suspensions, their soluble fractions and DCBP were then determined on the keratinocyte cell line HaCaT. Impact of suspensions of Po13 on viability was hardly affected by aging. In contrast, the soluble fractions were more toxic after aging. Suspensions and filtrates induced neither release of reactive oxygen species nor formation of DNA strand breaks. The samples exhibited only limited effects on the proteome of HaCaT cells. Conversely, DCBP was cytotoxic and induced the production of ROS, but was not genotoxic. DCBP was found to activate CYP450 monooxygenases known to be involved in the metabolism of xenobiotics. Altogether, our results show that aging of Po13 lead to the release of toxic soluble compounds.

Project description:Sweat plays a crucial role in thermo-regulation and skin health and potentially antimicrobial defense. Its composition is highly dynamic and its homeostasis involves a fine tuning of metabolic pathways. In-depth profiling of sweat protein composition will increase our understanding of skin disorders. Cystic Fibrosis (CF) is associated with high NaCl sweat concentration due to the absence of the CFTR protein. Patients with CF (pwCF) often display aquagenic palmoplantar keratodermia, a rare skin disorder characterized by skin wrinkling with oedema and whitish papules on the palms and/or soles, the pathophysiology of which is unknown. We report an in-depth analysis of the human sweat proteome of pwCF patients in comparison with that of healthy subjects (S).

Project description:Modified nucleotides in non-coding RNAs, such as tRNAs and snRNAs, represent an important layer of gene expression regulation through their ability to fine-tune mRNA maturation and transla-tion. Growing evidences support important roles of tRNA/snRNAs modifications and hence the enzymes that install them, in eukaryotic cell development and their dysregulation has been linked to various human pathologies including neurodevelopmental disorders and cancers. Human TRMT112 (Trm112 in Saccharomyces cerevisiae) functions as an allosteric regulator of several methyltransfer-ases (MTases) targeting molecules (tRNAs, rRNAs and proteins) involved in protein synthesis. Here, we have investigated the interaction network of human TRMT112 in intact cells and identify three poorly characterized putative MTases (TRMT11, THUMPD3 and THUMD2) as direct part-ners. We demonstrate that these three proteins are active N2-methylguanosine (m2G) MTases and that TRMT11 and THUMPD3 methylate positions 10 and 6 of tRNAs, respectively. In contrast, we discovered that THUMPD2 directly associates with the U6 snRNA and is required for the for-mation of m2G in this core component of the catalytic spliceosome. Consistently, our data reveal the combined importance of TRMT11 and THUMPD3 for optimal protein synthesis and cancer cell proliferation as well as a role for THUMPD2 in fine-tuning pre-mRNA splicing.

Project description:Alternative splicing is a potent modifier of protein function. Stromal interaction molecule 1 (Stim1) is the essential activator of store-operated Ca2+ entry (SOCE) triggering activation of transcription factors. Here, we characterize Stim1A, a splice variant with an additional 31 amino acid domain inserted in frame within its cytosolic domain. Prominent expression of exon A is found in astrocytes, heart, kidney and testes. Full length Stim1A functions as a dominant-negative regulator of SOCE and ICRAC, facilitating sequence specific fast calcium dependent inactivation and destabilizing gating or Orai1. Downregulation or absence of native Stim1A results in increased SOCE. Despite reducing SOCE, Stim1A leads to increased NFAT translocation. Differential proteomics revealed interference of Stim1A with the cAMP-SOCE crosstalk by altered modulation of phosphodiesterase (PDE8B), resulting in reduced cAMP degradation and increased PIP5K activity, facilitating an increased NFAT activation. Our study uncovers a hitherto unknown mechanism regulating NFAT activation and indicates that cell type specific splicing of Stim1 is a potent means to regulate the NFAT signalosome and cAMP-SOCE crosstalk.

Project description:Analysis of gene expression associated with exercise response. The hypothesis tested was that individuals with Type 2 Diabetes that failed to demonstrate exercise-induced metabolic improvements would also reflect this lack of response in their skeletal muscle transcriptional profile at baseline. Of 186 genes identified by microarray analysis, 70% were upregulated in Responders and downregulated in Non-responders. Several genes involved in substrate metabolism and mitochondrial biogenesis differed significantly between the groups at baseline. This differential baseline gene expression indicated that Non-responders had blunted oxidative capacity. Total RNA extracted from baseline samples of skeletal muscle of obese individuals with Type 2 Diabetes who were characterized as either Responders or Non-responders was examined for differential expression of exercise response-assocated genes.

Project description:The ProMetIS dataset corresponds to the proteomic and metabolomic analysis of mouse lines lacking the LAT (linker for activation of T cells; OMIM: 602354) or MX2 gene (MX dynamin-like GTPase 2; OMIM: 147890), as well as the control (WT) line. Besides its role in T-cell receptor (TCR) signaling (Roncagalli et al., 2010), LAT has been shown to be involved in neurodevelopmental diseases (Loviglio et al., 2017). The systematic generation of mouse models is part of the functional characterization of the genome by the IMPC consortium (Brown et al., 2018). This characterization is currently based on a battery of animal phenotypic tests (anatomy, behaviour, histology, haematology, physiology), the results of which feed the IMPC database (https://www.mousephenotype.org). In particular, gene knockouts associated to metabolic diseases have been identified (Rozman et al., 2018). To further characterize these models, global molecular approaches are required, such as the metabolomics analysis of plasma samples which has been described recently (Barupal et al., 2019). The originality of ProMetIS is to provide, in addition to the clinical data, the proteomics and metabolomics analysis of the LAT, MX2 and WT mouse models in liver and plasma. ProMetIS provides access to unique molecular functional information on the LAT and MX2 genes. In addition, the dataset has been generated by the four national infrastructures for mouse phenogenomics (PHENOMIN-ICS), proteomics (ProFI), metabolomics (MetaboHUB) and bioinformatics (IFB) for optimal quality, reproducibility and accessibility. The protocols and computational workflows provided here can be easily extended to a larger number of individuals and tissues. Finally, ProMetIS will be of great interest to develop and evaluate bioinformatics and biostatistical methods for the processing and integration of proteomic and metabolomic data.

Project description:The goal of this study is to report that breast cancer associated exosomes contain microRNAs (miRNAs) associated with the RISC Loading Complex (RLC) and display cell-independent capacity to process precursor microRNAs (pre-miRNAs) into mature miRNAs. Pre-miRNAs, along with Dicer, AGO2, and TRBP, are present in exosomes of cancer cells. CD43 mediates the accumulation of Dicer specifically in cancer exosomes. Cancer exosomes mediate an efficient and rapid silencing of mRNAs to reprogram the target cell transcriptome. Exosomes derived from cells and sera of patients with breast cancer instigate non-tumorigenic epithelial cells to form tumors in a Dicer-dependent manner. These findings offer opportunities for the development of exosomes based biomarkers and therapies. Exosomes from cancer cells were isolated using established ultracentrifugation methods. The global miRNA content of non-tumorigenic cells was investigated before and after exosomes treatment to study the role of microRNA biogenesis in exosomes for cancer progression and the transformation process of normal cells.
