9-oxo-ODAs suppresses the proliferation of human cervical cancer cells through the inhibition of CDKs and HPV oncoproteins

ABSTRACT: We evaluated the growth inhibitory effects of 9-oxo-ODAs in cervical cancer caused by HPV infection. We confirmed the antitumor effects of 9-oxo-ODAs in vitro, identified their mechanisms of action using omics analysis, and verified their effects ex vivo and in vivo.

Analysis of RA vs 4-oxo-RA gene expression regulation in Xenopus laevis

Project description:We analyzed anterior halves of NF stage 15 embryos that were treated with 10uM DMSO (control), or 1uM RA and 10uM 4-oxo-RA (experimental) for changes in gene expression induced by the two mophogens. RNA-sequencing revealed significant overlap in genes upregulated by both RA and 4-oxo-RA, and to a lesser extent, those downregulated by them. We report all Zic-1 targets described in accompanying manuscript to be regulated by both RA and 4-oxo-RA.

Project description:Mucosal human papillomavirus (HPV) subtypes 16 and 18 are causative agents of cervical cancer, a leading cause of cancer-related deaths among women worldwide. In Japan, eggplant calyx is a folk remedy used to treat common warts. 9-oxo-(10E,12E)-octadecadienoic acid, isolated from eggplant calyx, may have antitumor effects. This study investigated the antitumor effects of 9-oxo-(10E, 12Z)-octadecadienoic acid and 9-oxo-(10E,12E)-octadecadienoic acid (9-oxo-ODAs) on human cervical cancer cells. 9-oxo-ODAs suppressed the proliferation of human cervical cancer cell lines (HeLa, and SiHa) in a concentration-dependent manner (IC50 = 25-50 µM). FCM analysis revealed that 9-oxo-ODAs induced apoptosis. Transcriptome, proteomics, and enrichment analyses revealed that treatment with 9-oxo-ODAs significantly altered the cell cycle and p53 pathways and decreased cyclin-dependent kinase 1 (CDK1) protein expression. Real-time PCR analysis demonstrated that 9-oxo-ODAs reduced CDK1 mRNA expression in a concentration-dependent manner. In vitro, 9-oxo-ODAs reduced the HPV oncoprotein expression. In ex vivo human cervical cancer tissues, 9-oxo-ODAs decreased CDK1 expression and increased cleaved caspase 3, an apoptosis marker. Further, 9-oxo-ODAs showed the potential to suppressed metastatic formation and growth of cervical cancer in vivo. These findings suggest that 9-oxo-ODAs induce cell cycle arrest and apoptosis in HPV-positive human cervical cancer cells, and this process involves CDK1. Consequently, 9-oxo-ODAs may be potential therapeutic agents for cervical cancer.

AUF1 recognizes 8-oxo-guanosine embedded in DNA and stimulates APE1 endoribonuclease activity

Project description:The presence of rNMPs embedded in DNA represents a frequent genomic event able to destabilize the DNA backbone and to alter DNA replication and transcription. Usually, rNMPs are excised by RNase H2 and fully repaired by other enzymes belonging to the ribonucleotide excision repair pathway. The existence of modified rNMPs embedded in DNA, including 8-oxo-guanosine (8-oxo-G) and ribose monophosphate abasic site (rAP), have been recently highlighted by several works including ours. While eukaryotic RNase H2 is unable to process rAP and 8-oxo-G embedded in DNA, we demonstrated a role of APE1, a key enzyme of the base excision repair pathway, on these lesions. Notably, though APE1 activity on the repair of rAP sites results as efficient as the canonical dAP site, its incision repair activity on 8-oxo-G is very weak. Therefore, in order to guarantee an efficient repair activity of 8-oxo-G, additional processing mechanisms should be active in cells. In this work, by using an unbiased proteomic approach to isolate proteins able to recognize 8-oxo-G embedded in DNA, we discovered that AUF1 actively and specifically recognize 8-oxo-G and, interestingly, stimulates the APE1 enzymatic activity on this substrate. Although further structural analysis should be performed, it is reasonable that AUF1 facilitates the endoribonuclease activity of APE1 on 8-oxo-G by stabilizing the lesion substrate. Furthermore, by two orthogonal approaches including GST-pulldown and proximity ligation assay, we uncovered a novel protein-protein interaction between AUF1 and APE1. Remarkable, this interaction is modulated by H2O2-treatment, playing a putative biological role during cell response to oxidative stress.

Microarray analysis of effect of N-(3-oxo-dodecanoyl) homoserine lactone on S. cerevesiae

Project description:We examined whether the budding yeast Saccharomyces cerevisiae can sense chemical signals from prokaryotes, specifically a variety of quorum sensing molecules from different bacteria species and from Candida albicans. We found that only N-acyl-3-oxo-dodecanoyl homoserine lactone (C12) from the opportunistic human pathogen Pseudomonas aeruginosa induces a stress response in yeast. Microarray experiments were performed in order to continue investigating the stress response. We treated S. cerevesiae (WT strain W303) with N-(3-oxo-dodecanoyl) homoserine lactone (C12), a quorum sensing molecule of Pseudomonas aeruginosa, which we found causes a stress response using a GFP reporter for HSP-12. Treatment conditions: 100 uM C12, 100 uM C12-lactam (control: synthetic analogue of C12 that is inactive in P. aeruginosa), DMSO (control: solvent), and 0.3 mM H2O2 (for comparison to oxidative stress).

Project description:Study of the impact of 4-oxo-RA on LSK cell characteristics post-myocardial infarction in RarB KO mice. Myocardial infarction was induced in female RarB KO mice aged 6 to 12 weeks by LAD occlusion. On the 1st and 2nd post-MI days, mice were intraperitoneally injected with 30 mg/kg 4-oxo-RA (MI+4-oxo-RA) or DMSO in PBS (MI+vehicle). Three days post-MI, mice were euthanized. ScRNA-seq was performed on 25k LSK cells (Lin-, Sca1+, c-Kit+) that were facs sorted.

HPV Oncoproteins and the Ubiquitin Proteasome System: A Signature of Malignancy?

Project description:Human papillomavirus (HPV) E6 and E7 oncoproteins are critical for development and maintenance of the malignant phenotype in HPV-induced cancers. These two viral oncoproteins interfere with a plethora of cellular pathways, including the regulation of cell cycle and the control of apoptosis, which are critical in maintaining normal cellular functions. E6 and E7 bind directly with certain components of the Ubiquitin Proteasome System (UPS), enabling them to manipulate a number of important cellular pathways. These activities are the means by which HPV establishes an environment supporting the normal viral life cycle, however in some instances they can also lead to the development of malignancy. In this review, we have discussed how E6 and E7 oncoproteins from alpha and beta HPV types interact with the components of the UPS, and how this interplay contributes to the development of cancer.
