Two circulating biomarkers are associated with the serological status of Rheumatoid Arthritis patients: a tool for precision medicine strategies

ABSTRACT: In the present work, we aimed to carry out a pipeline based on quantitative proteomics tools to discover, verify and validate circulating proteins that are associated with the presence in serum of RF and ACPA and may therefore have potential for the stratification of RA patients and the application of precision medicine strategies based on these molecular signatures.

Analysis of gene expression in mouse brain regions after exposure to pulse-modulated 1.9 GHz radiofrequency fields

Project description:This study assessed changes in gene expression in 7 mouse brain regions using Illumina Mouse WG-6 (v2) beadchips after exposure to 1.9 GHz pulse-modulated radiofrequency field exposure for 4h/day for 5 days. High dose RF field exposure was whole body average SAR and brain average SAR were 1.45 W/kg and 0.16 W/kg, respectively. Total RNA isolated from the amygdala, auditory cortex, caudate, cerebellum, hippocampus, hypothalamus and medial prefrontal cortex of 1.9 GHz pulse-modulated radiofrequency field exposed mice and compared to that in sham (handled) exposed animals. An unhandled control group was also included in the study design. A total of 20 RNA samples (5 independent biological tissues for each of 4 treatment groups) were collected for each brain region. When analysis of gene expression was conducted within individual brain regions when controlling the false discovery rate (FDR), no differentially expressed genes were identified relative to the sham control. However, it must be noted that most fold changes among groups were observed to be less than 1.5 fold and this study had limited ability to detect such small changes. While some genes were differentially expressed without correction for multiple-comparisons testing, no consistent pattern of response was observed among different RF-exposure levels or among different RF-modulations. The current study provides the most comprehensive analysis of potential gene expression changes in the rodent brain in response to RF field exposure conducted to date. Within the exposure conditions and limitations of this study, no convincing evidence of consistent changes in gene expression was found in response to 1.9 GHz pulse-modulated RF field exposure.

Using Model Organism Saccharomyces Cerevisiae to Evaluate the Differential Effects of ELF-MF and RF-EMF exposure on Global Gene Expression

Project description:The potential health hazard of exposures to electromagnetic fields (EMF) continues to cause much public concern. However, the biological and health effects of exposures to EMF remain controversial and their biophysical mechanisms are unclear. In the present study, we used Saccharomyces cerevisiae to identify genes responding to extremely low frequency magnetic fields (ELF-MF) and to radiofrequency (RF) EMF exposures. The expression of genes was analyzed by microarray screening and confirmed by real-time reverse transcription -polymerase chain reaction. In confirmation experiments, we found that there was no statistically significant change in three of the ELF-MF responsive candidate genes (P>0.05). On the other hand, out of the forty genes that responded to RF-EMF, the confirmation experiments found that only five were affected: structural maintenance of chromosomes 3-gene (SMC3), aquaporin 2 –gene (AQY2), halotolerance protein 9 –gene (HAL9), yet another kinase 1 -gene (YAK1) and one of unknown function gene (open reading frame: YJL171C) (P<0.05). Overall, this study has demonstrated that the yeast cells did not respond to 50 Hz ELF-MF and that the response to RF-EMF is limited to only five genes. The biological consequences of the observed gene expression changes induced by RF-EMF await further investigation. The yeast cells were exposed to 0.4 mT 50 Hz ELF-MF or 1800 MHz RF-EMF at a specific absorption rate of 3.5 W/kg for 6 hours.

Effects of global system for mobile communications 1800 MHz radiofrequency electromagnetic fields on gene expression in MCF-7 cells

Project description:Despite many studies over a decade, it still remains ambiguous as to the real biological effects induced by radiofrequency electromagnetic fields (RF EMF). Epidemiological studies indicates that long-term exposure to EMF could increase the risk of breast cancer. Some reports have showed that in vitro EMF exposures change cellular gene expression. In this study, we investigated global gene expression responses to RF EMF simulating the Global System for Mobile Communications (GSM) 1800 MHz signal in human breast cancer cell line MCF-7 using transcriptomic approaches. MCF-7 cells were intermittently (5 min fields on/10 min fields off) exposed to RF EMF at an average specific absorption rate (SAR) of 2.0 W/kg (2 W/kg exposure group) or sham-exposed (2 W/kg control group) for 24 h, or exposed to RF EMF at a higher level of 3.5 W/kg (3.5 W/kg exposure group) or sham-exposed (3.5 W/kg control group).
