Project description:The objective of the study was to analyze the changes in proteomics that occur in saliva in acute abdominal disease (AAD) by proteomics in order to identify new proteins that could allow gaining knowledge about the physiopathological changes that can occur in this fluid and identify possible new biomarkers of acute abdominal disease. Samples from two groups of horses were analyzed by TMT proteomic analysis: - Diseased horses (n=7; 4 stallions and 3 mares; mean age = 8 years (range 1-18); Andalusians (n=5) and crossbred (n=2)). Presence of AAD was diagnosed based on history, physical examination (abdominal auscultation, rectal examination and nasogastric intubation) and additional diagnostic tests incl. complete blood count (CBC), serum biochemistry profile, abdominal ultrasound and or abdominocentesis. Final diagnoses were impaction of ascending colon (n=3), right dorsal colon displacement (n=2) and spasmodic colon (n=2). - Healthy horses (n=6, 3 geldings, 2 stallions and 1 mare; mean age = 12 years (range 4-15); Andalusians (n=4) and crossbred (n=2)). Horses were found healthy based on history, clinical examination, CBC and serum biochemistry profile. These procedures were approved by the ethical committee of the University of Murcia (CEEA 288/2017).

Project description:The objective of this study was to evaluate the impact of dietary Spirulina and lysozyme supplementation over the muscle proteome of piglets during the post-weaning stage. Thirty piglets were randomly distributed among three diets: control (no microalga), SP (10% Spirulina) and SP+L (10% Spirulina + 0.01% lysozyme). They were fed ad libitum for 4 weeks, after which they were sacrificed and samples of the longissimus lumborum muscle were taken. The muscle proteome was analysed using a Tandem Mass Tag (TMT)-based quantitative approach.

Project description:This study examines whether maternal low ω6:ω3 ratio diet and offspring seaweed (SW) supplementation can improve offspring immunity and performance by elucidating the effects on piglet serum proteome. A total of 16 sows were given either a standard (CR, 13:1) or low ω6:ω3 ratio diet (LR, 4:1) during pregnancy and lactation and their male weaned piglets were supplemented with SW powder (4 g/kg, SW) or not (CT) in a 21-day post-weaning (PW) diet. Four PW piglet groups were then identified based on dam and piglet treatment, namely CRCT, CRSW, LRCT, and LRSW (n = 10 each). Piglet serum collected at weaning and d21 PW were analyzed (n = 5 each) using TMT-based quantitative proteomics and validated by appropriate assays.

Project description:Background: Low birth weight is associated with an increased adult metabolic disease risk. It is widely discussed that poor intrauterine conditions could induce long-lasting epigenetic modifications, leading to systemic changes in regulation of metabolic genes. In a unique cohort of 17 monozygotic (MZ) monochorionic female twins very discordant for birth weight (relative differences ranging from 21.3-35.7%), we examined if adverse prenatal growth conditions experienced by the smaller co-twins lead to systemic long-lasting DNA methylation changes. Genome-wide DNA methylation profiles were acquired from saliva DNA using the Infinium HumanMethylation450 BeadChip, targeting ~2% of all CpGs in the genome. Results: Overall, co-twins showed very similar genome-wide DNA methylation profiles. Since observed differences were almost exclusively caused by variable cellular composition, an original marker-based adjustment strategy was developed to eliminate such variation at affected CpGs. Among adjusted and unchanged CpGs 3153 were differentially methylated between the heavy and light co-twins at nominal significance (p<0.01), of which 45 showed absolute mean β-value differences >0.05 (max=0.08). Deep bisulfite sequencing of eight such loci revealed that differences remained in the range of technical variation, arguing against a reproducible biological effect. Analysis of methylation in repetitive elements using methylation-dependent primer extension assays also indicated no significant intra-pair differences. Conclusions: Severe intrauterine growth differences observed within these MZ twins are not associated with long-lasting DNA methylation differences in cells composing saliva, detectable with up-to-date technologies. Additionally, our results indicate that uneven cell type composition can lead to spurious results and should be addressed in epigenomic studies. Project description:Oral cavity cancer is one of the most common cancer worldwide and oral cavity squamous cell carcinoma (OSCC) accounts for up to 90 percent of oral cancers. In Taiwan, OSCC is the fifth most common malignancy of male and causes more than 250 deaths per year. The poor outcome of OSCC patients is principally ascribed to the fact that this disease is often advanced at the time of diagnosis, suggesting that early detection and prevention of OSCC are urgently needed to reduce the cancer burden. Analysis of cancer-related body fluids is one of promising approaches to identify cancer-related molecules and biomarker candidates. To identify salivary biomarkers of OSCC, salivary samples of OSCC patients, individuals with oral potentially malignant disorders (OPMD), and healthy volunteers were collected. With isobaric tags for relative and absolute quantitation (iTRAQ)-based mass spectrometry (MS) analysis, 1838 proteins were identified. The levels of 102 proteins are found to be elevated in the saliva samples of OSCC patients compared to that of OPMD and healthy groups. To improve the feasibility of biomarkers, we used a two-phase verification to measure these candidates. Among them, salivary levels of AHSG, CFH, FGA, and SERPINA1 were determined to be significantly higher in the OSCC patients than in the healthy or in the OPMD group with both of MRM-MS and ELISA. Furthermore, the levels of these proteins were highly related to tumor size, lymph-node metastasis, and cancer stages. The results of this study not only provides a powerful strategy for verification of biomarker candidates, but also suggested that AHSG, CFH, FGA, and SERPINA1 could act as salivary biomarkers for OSCC detection.

Project description:Haemophilus parasuis(HPS) is a prominent swine pathogen that causes GlM-CM-$sser's disease characterized by fibrinous polyserositis, meningitis and arthritis; however, the molecular mechanisms underlying disease pathogenesis remains poorly understood, particularly the host counteraction to HPS invasion by the immune system. Here, we investigated the global expression changes in spleen following HPS infection using the Affymetrix Porcine Genechip.Our findings indicate previously unrecognized gene transcription changes in case of HPS infection in vivo and many presumed cascades in the study are clearly merit further investigation. Our data should provide new clues for immune response in mammals and identification of candidate genes related to HPS resistance. All animal tissue collection procedures were performed according to protocols approved by The Hubei Province, PR China for Biological Studies Animal Care and Use Committee. Piglets at 30 days old were determined to be HPS-free by serum indirect haemagglutination test before artificial bacterial challenges. Each piglet was intratracheally challenged with 5M-CM-^W108 colony-forming units (CFU) per millilitre of HPS strain 0165 (serotype 5) . A total of 5 piglets were challenged, and 5 pigs were used as controls. These two groups were raised in isolated facilities. Six microarrays were used in the experiment, corresponding to the RNAs from spleen tissues of three most severe piglets with fibrinous polyserositis, meningitis and arthritis after HPS infection and three controls.

Project description:Objectives: Salivary glands are affected during radiotherapy in the head and neck region, leading to a reduction in salivary flow and changes its composition. Besides negatively affecting the oral soft tissues, this can also lead to dental impairment. Thus, we evaluated the effect of radiotherapy in the proteomic profile of the saliva in patients with head-and-neck-cancer (HNC). Materials and methods: HNC patients had their saliva collected before (BRT), during (2-5 weeks; DRT) and after (3-4 months; ART) radiotherapy. Saliva was also collected from healthy volunteers (control; C). Samples were processed for proteomic analysis. Results: In total 1.055 proteins were identified, among which 46 were common to all groups, while 86, 86, 286 and 395 were exclusively found in C, BRT, DRT and ART, respectively. Remarkably, alpha-enolase was increased 35-fold DRT compared with BRT, while proline-rich proteins were decreased. ART there was a 16-fold increase in scaffold attachment factor-B1 and a 3-fold decrease in alpha-enolase and several cystatins. When compared with C, salivary proteins of BRT patients showed increases cystatin-C, lysozyme C, histatin-1 and proline-rich proteins. Conclusion/Clinical revelance: Both HNC and radiotherapy remarkably change the salivary protein composition. Altogether, our results, for the first time, suggest investigating alpha-enolase levels in saliva DRT in future studies as a possible biomarker and strategy to predict the efficiency of the treatment. Moreover, our data provide important insights for designing dental products that are more effective for these patients and contribute to a better understanding of the progressive changes in salivary proteins induced by radiotherapy.
