Comparison of three methods for isolating human plasma EVs by LC-MSMS

Project description:The lack of standardized protocols for isolating extracellular vesicles (EVs), particularly from biobank-stored plasma, poses a substantial limitation for the study of biomarkers. Combining current isolation methods could enhance the specificity and purity of EVs isolation for their further use in diagnosis and personalized medicine. In this study, plasma samples from healthy individuals were subjected to extracellular vesicle isolation using ultracentrifugation (UC), size exclusion chromatography (SEC), and a combined approach of SEC and UC (SEC+UC). Characterization of the isolated EVs with NTA and TEM indicated that all isolation methods successfully isolated EVs, being not altered due to the long-term storage. Also, the western blot analysis confirmed the presence of exosome markers in all isolates but showed a higher expression of albumin in EVs-UC, suggesting potential plasma protein contamination. Proteomic analysis of samples from different isolation methods identified 542 proteins, with SEC+UC yielding the most complex proteome at 364 proteins. GO analysis of cellular component revealed differences in terms related to EVs and plasma, where SEC+UC proteins had the highest proportion of exosomal proteins. As a result of further study of the unique proteins detected in each isolation method, it was revealed that the SEC+UC method detected 181 unique proteins, demonstrating its superiority in the detection of low plasma concentration proteins. These findings support the robustness of the SEC+UC approach for EV isolation and proteomic studies due to its high efficiency and purity in isolating EVs. This study emphasizes the efficacy of the SEC+UC approach in obtaining highly pure and diverse EV populations suitable for comprehensive proteomic analysis with application to the discovery of biomarkers from biobank-stored plasma.

Coordinate β-adrenergic inhibition of mitochondrial activity and angiogenesis arrest tumor growth

Project description:Mitochondrial metabolism has emerged as a promising target against the mechanisms of tumor growth. Herein, we have screened an FDA-approved library to identify drugs that inhibit mitochondrial respiration. The β1-blocker nebivolol specifically hinders oxidative phosphorylation in cancer cells by concertedly inhibiting Complex I and ATP synthase activities. Complex I inhibition is mediated by interfering the phosphorylation of NDUFS7. Inhibition of the ATP synthase is exerted by the overexpression and binding of the ATPase Inhibitory Factor 1 (IF1) to the enzyme. Remarkably, nebivolol also arrests tumor angiogenesis by arresting endothelial cell proliferation. Altogether, targeting mitochondria and angiogenesis triggers a metabolic and oxidative stress crisis that restricts the growth of colon and breast carcinomas. Nebivolol holds great promise to be repurposed for the treatment of cancer patients.

Re-localization of components of the guided entry of tail-anchored proteins targeting pathway to cytosolic foci upon glucose starvation

Project description:Protein targeting is an essential step in the biogenesis of membrane proteins and the guided-entry of tail-anchored proteins (GET) pathway is a key targeting route to the endoplasmic reticulum. The adaptive response to cellular stress is multifaceted but a hallmark is the formation of membraneless organelles, which can act as sequestration sites for particular factors, allowing regulation of cellular processes during stress and/or facilitating efficient recovery upon stress alleviation. Upon glucose withdrawal, we observe that the Get3 ATPase of the GET pathway re-locates to dynamic cytosolic foci, here termed GET bodies, that also contain proteins of the GET pathway pre-targeting complex. Our data demonstrate that Get3 and the Get4-Get5 heterodimer play important roles in the assembly of these structures, and we further uncover the requirement of the TPR domain of Sgt2 for the GET body formation. Compositional analyses of GET bodies by enrichment coupled to mass spectrometry as well as high-throughput microscopy-based screening reveal a chaperone-rich core and the association of an inventory of proteins involved in metabolic processes. Temporal analyses of GET body assembly demonstrate the sequential recruitment of different chaperones and we discover the requirement of the co-chaperones Sis1 and Sti1 for maintaining the dynamic properties of these structures. As lack of Adh2, which converts ethanol to acetaldehyde in a reaction coupled to NADH production, impairs GET body disassembly, and the addition of NADH promotes GET body dissolution in vitro, our results suggest a mechanistic basis for the formation of GET bodies in response to the metabolic shift caused by glucose withdrawal.
