Temporal phosphoproteomic analysis of VEGF-A signaling in HUVECs: An insight into early signaling events associated with angiogenesis


ABSTRACT: Aim of the study was to elucidate the phosphoproteomics alterations caused by the treatment of VEGF-A in HUVEC cells at early timepoints of 1, 5 and 10 minutes. Label-free quantitative temporal phosphoproteomics experiments were carried out using LC-MS/MS methods. LTQ orbitrap XL ETD (Thermo Electron, San Jose, CA, USA) connected to UltiMate 3000 nanoLC Pump (Dionex Co., Sunnyvale, CA, USA) was used for data acquisition.

Vascular endothelial growth factor-A (VEGF-A) is one of the primary factors promoting angiogenesis in endothelial cells. Although defects in VEGF-A signaling are linked to diverse pathophysiological conditions, the early phosphorylation-dependent signaling events pertinent to VEGF-A signaling remain poorly defined.
