Quantitative LC-MS/MS Proteomics of human liver microsomes (healthy controls and normal-tumour from cancer patients)

ABSTRACT: We report quantification of proteins in human liver microsomal samples from 15 healthy volunteers and 18 patients with cancer in the liver (mainly, colorectal cancer liver metastasis). These data can be used in physiologically based pharmacokinetic models to predict appropriate drug doses in patients with cancer in their liver, especially colorectal cancer liver metastasis.

Introduction
Alterations in expression and activity of human receptor tyrosine kinases (RTKs) are associated with cancer progression and in response to therapeutic intervention.
Methods
Thus, protein abundance of 21 RTKs was assessed in 15 healthy and 18 cancerous liver samples [2 primary and 16 colorectal cancer liver metastasis (CRLM)] matched with non-tumorous (histologically normal) tissue, by a validated QconCAT-based targeted proteomic approach.
Results
It was demonst ...[more]
