Integration of human adipocyte chromosomal interactions with adipose gene expression prioritizes obesity-related genes from GWAS

Project description:We perform promoter Capture Hi-C in human adipocytes to investigate interactions between gene promoters and distal elements as a transcription-regulating mechanism contributing to these phenotypes. We find that promoter-interacting elements in human adipocytes are enriched for adipose-related transcription factor motifs, such as PPARG and CEBPB, and contribute to heritability of cis-regulated gene expression. Transcript profiling of Arabidopsis thaliana swi3d mutant with inactivated SWI3D subunit of SWI/SNF complex

Project description:SWI3D subunit of SWI/SNF chromatin remodeling complexes (CRCs) is involved in the control of Arabidopsis development. We found, using microarray analysis that inactivation of SWI3D gene caused altered expression of genes belonging to various regulatory pathways, including metabolism, stress, response to hormone and development. On the basis of this experiment and our other data we concluded that Arabidopsis SWI3D modulates major regulatory pathways. Transcript profiling of Arabidopsis er/erl1/erl2 mutant line

Project description:The members of ERECTA family (ERf) leucine-rich receptor like kinases (ERECTA, ERECTA-LIKE1, ERECTA-LIKE2) play important role in various regulatory processes like epidermal patterning, stomata development, meristem size, inflorescence architecture control and hormonal signaling. In this experiment we showed that inactivation of all three ER's causes wide range of transcriptional changes affecting developmental, hormonal and metabolic processes. Based on our transcriptomic data and other experiments we concluded that ERf's kinases are involved in transcriptional control of genes expression. Overexpression of the selective autophagy receptor AtNBR1

Project description:Cargo receptors are well established elements of the selective autophagy in all eukaryotes. The NBR1-like family of such receptors is evolutionarily conserved, however plants have only one member of the family, which is a functional and structural hybrid of its two animal counterparts, p62 (SQSTM1) and NBR1. Examination of plants overexpressing NBR1 indicates that they are oversensitive to TOR inhibitors and have more autophagosomes than the parental lines. The observed changes in shoots suggested induction of ABA signalling pathway, what was further corroborated by the germination tests and the assay of ABA level. The transcriptome changes in roots, such as lower level of snoRNA and some tRNAs, suggested inhibition of ribosome biogenesis, what is supported by identification of RPS6, an important component of TOR-dependent translational control, as a novel partner of NBR1.
