Targeting Supramolecular Active Complexes of Nav 1.7/Nav 1.8 to Relieve Chronic Neuropathic Pain


ABSTRACT: The aim of this project was to identify the proteins participating in assemble of supramolecular active complex containing Nav1.7 and Nav1.8 in dorsal root ganglia (DRG) of mice with nerve injury 6 weeks. DRG and spinal dorsal horn (SDH) tissues were collected from spared nerve injury mice and sham-operated mice.

Neuropathic pain (NP) affects 7%-10% of population, with current treatments often proving inadequate. Here we show that Na<sub>v</sub>1.7 and Na<sub>v</sub>1.8 form supramolecular active complexes (SMACs) with polygonal lattice structure in dorsal root ganglion (DRG) neurons of mouse models and patients with severe chronic NP.
