Quantitative proteomics of serum samples using tandem mass tags in dogs with recurrent epileptic seizures

ABSTRACT: This study included four groups of dogs (group A: healthy controls, group B: idiopathic epilepsy receiving antiepileptic medication (AEM), group C: idiopathic epilepsy without AEM administration, group D: structural epilepsy), for which comparative proteomic analysis of serum samples was performed.

Project description:This study included four dog groups (group A: 10 healthy dogs, group B: 9 dogs with idiopathic epilepsy receiving antiepileptic medication-AEM, group C: 8 dogs with idiopathic epilepsy without AEM administration and group D: 7 dogs with seizures due to structural brain abnormalities). The epileptic dogs were allocated into these 3 groups after a diagnostic investigation that included laboratory testing, thoracic radiographic and abdominal ultrasonographic examination, brain imaging, CSF routine and proteomic analysis. The purpose of the current study was to investigate and compare the proteomic profile among the four groups of dogs.

Project description:The aim here was to identify mRNA expression biomarkers associated with the disease epilepsy and the antiepileptic drug response. Gene expression profiles of drug-naïve patients with epilepsy were compared with that of healthy controls. The profiles were significantly different between the two groups as well as patients with different epilepsy types i.e. idiopathic, symptomatic and cryptogenic. Besides, patients showing differential response to antiepileptic monotherapies were also having differential blood gene expression profiles.

Plasma proteomics in epilepsy: network-based identification of proteins associated with seizures

Project description:Purpose Identification of potential biomarkers of seizures. Methods In this exploratory study, we quantified plasma protein intensities in 15 patients with recent seizures compared to 15 patients with long-standing seizure freedom. Using TMT-based proteomics we found fifty-one differentially expressed proteins. Results Network analyses including co-expression networks and protein-protein interaction networks, using the STRING database, followed by network centrality and modularity analyses revealed 22 protein modules, with one module showing a significant association with seizures. The protein-protein interaction network centered around this module identified a subnetwork of 125 proteins, grouped into four clusters. Notably, one cluster (mainly enriching inflammatory pathways and Gene Ontology terms) demonstrated the highest enrichment of known epilepsy-related genes. Conclusion Overall, our network-based approach identified a protein module linked with seizures. The module contained known markers of epilepsy and inflammation. The results also demonstrate the potential of network analysis in discovering new biomarkers for improved epilepsy management.

Changes in the gene expression in the inferior colliculus of the sound stimulated rats both Wistar and the strain susceptible to audiogenic seizures WAR

Project description:Using microarray analyses and subsequent verification by RT-PCR, we studied the changes in gene expression in the inferior colliculus after an ictal event in one models of audiogenic epilepsy, Wistar audigenic rat (WAR). WAR is a genetically selected strain susceptible to audiogenic seizures that was inbred in the School of Medicine of RibeirÃ£o Preto (Brazil) beginning in 1990. This strain is a model of audiogenic idiopathic epilepsy that develops tonic-clonic generalized seizures. Genetic animal models of epilepsy are an important tool for further understanding the basic cellular mechanisms underlying epileptogenesis and for developing novel antiepileptic drugs. We conducted a comparative study of gene expression in the inferior colliculus, a nucleus that triggers audiogenic seizures, using two animal models, the Wistar audiogenic rat (WAR) and the genetic audiogenic seizure hamster (GASH:Sal). For this purpose, both models were subjected to auditory stimulation, and 60 minutes after stimulation, the inferior colliculi were collected. As a control, intact Wistar rats and Syrian hamsters were subjected to identical stimulation and tissue preparation protocols to those performed on the experimental animals. A total of 15 animals were used in this study according to the following distribution: 9 male WAR and 6 male control rats (Rattus norvegicus, Wistar albino, Charles River Laboratories) at 12 weeks of age and a body weight of approximately 230 g. The animals were exposed to auditory stimulation, and 60 min after the seizures, we harvested the IC for all gene expression analyses. As controls, normal Wistar rats were exposed to the same stimulation according to the identical procedure. For gene microarray (Rat Gene 1.0 ST), the rats were randomly divided into two groups, and we used both sides of the IC (ipsilateral and contralateral) from each animal: stimulated Wistar rats and stimulated WAR rats.

It exerts effective antiepileptic effect against various epilepsy models. Exploring the gene transcriptomic profile of long-term treatment of Q808 is necessary. In the present study, hippocampus RNA-sequencing was performed to reveal the transcriptome profile of rats after treatment of gavage of Q808 for 28 day.

Disruption of KCC2 in parvalbumin-positive interneurons causes fatal epilepsy in mice

Project description:Since many years, we are interested in the regulation of the intraneuronal chloride concentration. We were the first group reporting a full knockout of the KCl-co-transporter KCC2, which dies immediately after birth due to respiratory failure. Notably, KCC2 loss-of-function mutations are associated with inherited febrile seizures, severe genetic generalized epilepsy and epilepsy of infancy with migrating focal seizures. Here, we used our floxed line to study the consequences of the disruption of KCC2 within parvalbumin-positive interneurons in mice. Remarkably, this leads to the disinhibition of PV-positive interneurons as evidenced by a decrease of E-S-Coupling and an increase in the sIPSC frequency. Nevertheless, these mice develop fatal epilepsy with progressive loss of parvalbumin-positive interneurons thus increasing network excitability.

Structural basis for antiepileptic drugs and botulinum neurotoxin recognition of SV2A

Project description:More than one percent of people have epilepsy worldwide. Levetiracetam (LEV) is a successful new-generation antiepileptic drug (AED), and its derivative, brivaracetam (BRV), shows improved efficacy. Synaptic vesicle glycoprotein 2a (SV2A), a putative membrane transporter in the synaptic vesicles (SV), has been identified as a target of LEV and BRV. SV2A also serves as a receptor for botulinum neurotoxin (BoNT), which is the most toxic protein and has paradoxically emerged as a potent reagent for therapeutic and cosmetic applications. Nevertheless, no structural analysis on AEDs and BoNT recognition by full-length SV2A has been available. Here we describe the cryo-electron microscopy structures of the full-length SV2A in complex with the BoNT receptor-binding domain, BoNT/A2 HC, and either LEV or BRV. The large fourth luminal domain of SV2A binds to BoNT/A2 HC through protein-protein and protein-glycan interactions. LEV and BRV occupy the putative substrate-binding site in an outward-open conformation. A propyl group in BRV creates additional contacts with SV2A, explaining its higher binding affinity than that of LEV, which was further supported by label-free spectral shift assay. Numerous LEV derivatives have been developed as AEDs and positron emission tomography (PET) tracers for neuroimaging. Our work provides a structural framework for AEDs and BoNT recognition of SV2A and a blueprint for the rational design of new AEDs and PET tracers.
