Project description:Chlorpyrifos oxon (CPO), the toxic metabolite of the organophosphorus (OP) insecticide chlorpyrifos, causes developmental neurotoxicity in humans and rodents. CPO is hydrolyzed by paraoxonase-1 (PON1), with protection determined by PON1 levels and the human Q192R polymorphism. To examine how the Q192R polymorphism influences fetal toxicity associated with gestational CPO exposure, we measured biomarker inhibition and fetal-brain gene expression in wild-type (PON1+/+), PON1-knockout (PON1-/-), and tgHuPON1R192 and tgHuPON1Q192 transgenic mice. Pregnant mice exposed dermally to 0, 0.50, 0.75 or 0.85 mg/kg/d CPO from gestational days (GD) 6 through 17 were sacrificed on GD18. Biomarkers of CPO exposure inhibited in maternal tissues included brain acetylcholinesterase (AChE), RBC acylpeptide hydrolase (APH), plasma butyrylcholinesterase (BChE) and carboxylesterase (CES). Fetal plasma BChE was inhibited in PON1-/- and tgHuPON1Q192, but not PON1+/+ or tgHuPON1R192 mice. Fetal brain AChE and plasma CES were inhibited in PON1-/- mice, but not in other genotypes.

We used both wt (PON1+/+) and PON1 knockout (PON1-/-) mice and PON1-/- mice carrying transgenes encoding the human alloforms tgHuPON1Q192 and tgHuPON1R192 to gain insight into the mechanisms of neurotoxicity of CPO throughout postnatal development, and to ascertain the importance of the PON1Q192R polymorphism for protecting against developmental toxicity of CPO. Whole-genome microarrays were used to measure gene expression changes associated with chronic CPO exposure of developing (PND 4-21) PON1-/-, tgHuPON1Q192R transgenic and PON1+/+ mice. Expression profiles are derived from cerebella from wild-type C57/Bl6 and PON1-/- on a C57/Bl6 background and two transgenic strains (tgHuPON1Q192, tgHuPON1R192) expressing either human PON1Q192 or human PON1R192 on the PON1-/- C57/Bl6 background. Project description:Nitric oxide is a pleiotropic free radical produced by three nitric oxide synthases (NOS1-3), of which the inducible NOS2 is involved in carcinogenesis. In this study, RNA-seq, ChIP-seq, and qRT-PCR experiments combined with bioinformatic analysis showed that NRF2 is a repressor of the NOS2 gene by maintaining a distal enhancer located 22 Kb downstream of the transcription start site (TSS) in a poised state. NRF2 depletion primed the activation of this enhancer, exerting a pioneering activity before achieving full activation. Specifically, NRF2 controls the inducible expression of NOS2 in response to intracellular oxidative stress and extracellular oxygen partial pressure. Abrogation of NOS2 expression using esiRNAs partially reduced or severely impaired the ability to form 3D spheroids respectively of WT and NRF2-depleted Panc-1 cells. Mechanistically, NOS2 and nitric oxide release are required to sustain the epithelial-to- mesenchymal transition (EMT) process. Importantly, silencing of NOS2 blocked 3D Matrigel invasion of NRF2-depleted Panc-1 cells, demonstrating that a short circuit in the reciprocal regulation of NOS2 and NRF2 attenuates PDAC cell malignancy. Overall, our data demonstrate for the first time that: i) NRF2 is a repressor of the NOS2 gene in pancreatic cancer cells; ii) identify a poised enhancer whose priming is triggered in conditions that simulate or stimulate NRF2 depletion; iii) NOS2 activity is required for NRF2-depleted Panc-1 cells to maintain their malignancy and invasiveness.

Project description:We previously reported that high expression of the extracellular glutathione peroxidase GPX3 is associated with poor patient outcome in ovarian serous adenocarcinomas, and that GPX3 protects ovarian cancer cells from oxidative stress in culture. Here we tested if GPX3 is necessary for tumor establishment in vivo and to identify novel downstream mediators of GPX3’s pro-tumorigenic function. GPX3 was knocked-down in ID8 ovarian cancer cells by shRNA to test the role of GPX3 in tumor establishment using a syngeneic IP xenograft model. RNA sequencing analysis was carried out in OVCAR3 cells following shRNA-mediated GPX3 knock-down using two hairpins to identify GPX3-dependent gene expression signatures. GPX3 knock-down abrogated clonogenicity and intraperitoneal tumor development in vivo, and the effects were dependent on the level of GPX3 knock-down. RNA sequencing showed that loss of GPX3 leads to decreased gene expression patterns related to pro-tumorigenic signaling pathways. Validation studies identified GDF15 as strongly dependent on GPX3. GDF15, a member of the TGF-beta growth factor family, has known oncogenic and immune modulatory activities. Similarly, GPX3 expression positively correlated with pro-tumor immune cell signatures, including regulatory T-cell and macrophage infiltration, and displayed significant correlation with PD-L1 expression. We show for the first time that tumor produced GPX3 is necessary for ovarian cancer growth in vivo and that it regulates expression of GDF15. The immune profile associated with GPX3 expression in serous ovarian tumors suggests that GPX3 may be an alternate marker of ovarian tumors susceptible to immune check-point inhibitors.

Project description:The balance between laminins containing the α5 (LAMA5) or the α4 (LAMA4) subunits is determinant for the diapedesis of different leukocyte subtypes in inflammatory conditions. We have shown that the extracellular superoxide dismutase (SOD3) is a positive regulator of LAMA4, and this was associated with enhanced intratumor T cell infiltration in experimental and primary human cancers. Here we show that SOD3 overexpression in neoplastic or endothelial cells (ECs) downmodulates LAMA5 levels in implanted tumors. The inhibitory effect of SOD3 on LAMA5 expression occurs at the transcriptional level and is specific for SOD3, since LAMA5 levels are not changed by in SOD1-overexpressing cells. Global transcriptomic analyses revealed that SOD3 overexpression changes the transcription of 1,682 genes in ECs exposed to tumor secreted factors, being the canonical and non-canonical Nuclear Factor kappa B (NF-κB) pathways major SOD3 targets. Indeed, SOD3 reduced the transcription of well-known NF-κB target genes as well as the NF-κB-driven promoter activity in endothelial cells stimulated with tumor necrosis factor (TNF)-α, an inducer of NF-κB signaling

Project description:Using primary human bronchial epithelial cells collected at bronchoscopy, we have perturbed signaling pathways important in regulation of response to tobacco smoke exposure and cancer development: ATM, BCL2, GPX1, NOS2, IKBKB, and SIRT1 Using gene expression profiles generated for each pathway and four independent gene expression datasets, we show that SIRT1 activity is significantly up-regulated in cytologically normal airway epithelial cells from active smokers compared to non-smokers; and in contrast, this activity is strikingly down-regulated in non-small cell lung cancer. 52 samples were run on Affymetrix Human Exon 1.0 ST microarrays (n=7 for ATM, n=8 for GPX1, n=7 for SIRT1, n=7 for NOS2, n=7 for IKBKB, n=8 for BCL2, and n=8 for GFP). The CEL files were processed using RMA and the ENTREZ Gene CDF file. 13 samples were removed either because of a low Pearson correlation between samples (n=1 SIRT1 sample) or a strong batch effect (n=2 ATM, n=2 BCL2, n=2 GFP, n=2 GPX1, n=1 IKBKB, n=2 NOS2, n=1 SIRT1 samples) as determined by PCA.

Project description:GPX3 is primarily synthesized and secreted by renal tubular epithelial cells and serves as the main source of GPX3 in plasma. A portion of GPX3 adheres to the renal basement membrane, suggesting that GPX3 may also regulate renal cell physiological functions. Our previous work has found that GPX3 expression is downregulated in the renal tubular epithelial cells of mice that have undergone ischemia-reperfusion-induced acute kidney injury, but the specific impact of this downregulation remains unclear. To address this, we constructed mice with specific deletion of GPX3 in renal tubular epithelial cells and subjected them to ischemia-reperfusion modeling. We reported the protective role of native GPX3 in the kidneys under IRI-AKI conditions in mitigating oxidative stress and mitochondrial damage in tubular epithelial cells. The deletion of GPX3 in tubular epithelial cells exacerbated oxidative stress, apoptosis, and mitochondrial dysfunction in IRI-AKI. Renal cortex tissue from control and IRI-modeled mice was used for RNA sequencing. Overall, our data provide an overview of the genetic changes in the kidneys of mice with GPX3 knockout in both non-modeled and IRI-AKI-modeled conditions, laying the groundwork for studying the specific mechanisms by which GPX3 regulates renal function.

Project description:Oxidative stress is a common phenomenon and is linked to a wide range of diseases and pathological processes. Tissue-specific variation in redox signaling and cellular responses to oxidative stress may be associated with vulnerability to toxic agents and carcinogenic exposures. In order to provide a basis for tissue-specific difference, we examined the tissue-specific transcriptional features of 101 oxidative stress-associated genes in 10 different tissues and organs of healthy mice under physiological conditions. Microarray analysis results, which were consistent with qPCR results, showed that catalase, Gpx3 and Gpx4 were most highly regulated in the liver, kidney and testes. We also found the tissue-specific gene expression of SOD1 (liver and kidney), SOD2 (heart and muscle) and SOD3 (lung and kidney). The current results will serve as a reference for animal models and help advance our understanding of tissue-specific variability in oxidative stress-associated pathogenesis.
