A molecular glue degrader of the WIZ transcription factor for fetal hemoglobin induction

ABSTRACT: Here we report the discovery of molecular glue degraders of the WIZ gene control factor that potentially and robustly induce HbF expression in preclinical models.

A cereblon-based (CRBN) glue degrader of NEK7 regulates NLRP3 inflammasome in a context-dependent manner

Project description:Aberrant NLRP3 inflammasome activation is linked to many inflammatory diseases, driving the search for therapeutics inhibiting this pathway. NEK7 is proposed to mediate NLRP3 inflammasome assembly and activation by bridging adjacent NLRP3 subunits. Hence, reduction of NEK7 protein may block NLRP3 activation. We identified NK7-902, a potent and selective CRBN glue degrader of NEK7. NK7-902 degraded NEK7 in human immune cells and whole blood. However, full NEK7 degradation completely blocked NLRP3-dependent IL-1β release in vitro only in certain donors and experimental conditions. Unlike most CRBN glue degraders, NK7-902 effectively degraded NEK7 in murine cells and inhibited IL-1β release in mouse in vivo. By contrast, oral administration of NK7-902 in cynomolgus monkey caused long-lasting NEK7 degradation but only transiently blocked IL-1β in blood. These findings suggest NEK7 contributes to but is not absolutely required for NLRP3 activation in monkeys and humans.

TRI-611, a selective, brain-penetrant molecular glue degrader of ALK

Project description:Tyrosine kinase inhibitors (TKIs) targeting Anaplastic Lymphoma Kinase (ALK) have transformed the treatment landscape of ALK fusion-positive non-small cell lung carcinoma (NSCLC), but limited options for patients who progress on approved TKIs highlight a continued need for an orthogonal therapeutic approach. TRI-611 is a potent, brain-penetrant molecular glue degrader (MGD) of ALK fusion proteins with the potential to address this need . TRI-611 promotes the proximity of the ALK kinase domain and CRBN via a unique degron interface distal from the kinase active site. The unique binding interface of TRI-611 leads to selectivity across the proteome including known CRBN neo-substrates and other kinases. TRI-611 treatment induces degradation of all forms of oncogenic ALK fusion proteins, including TKI-resistant mutated versions of ALK, leading to regression of cell line and patient-derived subcutaneous and intracranial tumour models of ALK-positive NSCLC. TRI-611 can be combined with orthosteric ALK TKIs, achieving synergistic and durable tumour regressions. TRI-611 represents the first example of a clinical stage MGD targeting an oncogenic gene fusion and has the potential to expand the arsenal of therapeutic options for ALK-positive NSCLC patients.

Project description:Targeted protein degradation is a powerful pharmacological strategy that harnesses the ubiquitin proteasome system to eliminate disease-relevant proteins, including otherwise undruggable proteins. Here, we report an unbiased and broadly applicable platform for the systematic discovery of molecular glues across diverse E3 ligases. Using multiplexed mass spectrometry-based chemical screening, we identified a molecular glue, M12, that reprograms the E3 ligase DCAF11 to degrade DDX18. We found that M12 functions as a prodrug, activated by glutathionylation. The glutathione moiety then directly engages DCAF11, enabling neo-substrate recruitment. We demonstrate that this mechanism readily enables targeted degradation of a range of proteins. Our studies highlight a novel approach to redirect E3 ligase function through an enzyme-activated small molecule, expanding the scope of targeted protein degradation and chemically induced proximity.
