Project description:mRNA export is central to gene regulation and expression. In trypanosomes, transcription is polycistronic, all mRNAs are processed by trans-splicing and export is mediated by non-canonical mechanisms. We have described several conserved mRNA export pathway components in Trypanosoma cruzi, including orthologs of Sub2, a component of the TREX complex, and eIF4AIII, a core component of the exon junction complex (EJC). Few orthologs of proteins involved in mRNA export in higher eukaryotes are detectable in trypanosome genomes and examples of mechanistic divergence well known. To uncover additional components of the trypanosome mRNA maturation and export system we undertook an unbiased search for protein interactors of TcSub2 and TceIF4AIII. Significant overlap between TcSub2 and TceIF4AIII interacting protein cohorts suggests that both proteins associate with similar machinery. We confirmed several interactions with conserved core components of the EJC and multiple additional complexes, together with identification of proteins specific to trypanosomatids. The highly interactive super-interactome uncovered here, capable of supporting RNA processing from splicing through to nuclear export and cytoplasmic events, highlights kinetoplastid-specific and conserved components creating an amalgam to support the unique mRNA maturation mechanisms in trypanosomes.

Project description:Trypanosoma cruzi, the etiologic agent of Chagas disease, cycles through different life stages characterized by defined molecular traits associated with the proliferative or differentiation state. In particular, T. cruzi epimastigotes are the replicative forms that colonize the intestine of the Triatomine insect vector before entering the stationary phase that is crucial for differentiation into metacyclic trypomastigotes, which are the infective forms of mammalian hosts. The transition from proliferative exponential phase to quiescent stationary phase represents an important step that recapitulates the early molecular events of metacyclogenesis, opening new possibilities for understanding this process. In this study, we report a quantitative shotgun proteomic analysis of the T. cruzi epimastigote in the exponential and stationary growth phases. More than 3000 proteins were detected and quantified, highlighting the regulation of proteins involved in different subcellular compartments. Ribosomal proteins were upregulated in the exponential phase, supporting the higher replication rate of this growth phase. Autophagy-related proteins were upregulated in the stationary growth phase, indicating the onset of the metacyclogenesis process. Moreover, this study reports the regulation of N-terminally acetylated proteins during growth phase transitioning, adding a new layer of regulation to this process. Taken together, this study reports a proteome-wide rewiring during T. cruzi transit from the replicative exponential phase to the stationary growth phase, which is the preparatory phase for differentiation

Project description:This work aimed to investigate the differential expression of proteins from two distinct genetic groups of T. cruzi (´DTUs´, TcI and TcII), isolated from chronically infected individuals displaying the cardiac form of Chagas’ disease. For this purpose, epimastigote forms of the two isolates were cultured in vitro and the cells recovered for protein extraction. Comparative two-dimensional gel electrophoreses were performed and differentially expressed spots selected for identification by mass spectrometry, followed by database searching and protein categorization. The two-dimensional electrophoretic profiles revealed the complex composition of the T. cruzi extracted proteome. In total 46 differentially expressed proteins were identified present in 40 distinct spots found in the comparative gel analyses. The comparative two-dimensional gel electrophoresis allowed detection of major differences in protein expression between two T. cruzi isolates, belonging to the TcI and TcII genotypes. Our findings suggest that patients displaying the cardiac form of the disease harbor parasites capable of exhibiting distinct proteomic profiles.

Project description:Chagas disease is one of the most lethal diseases in Latin America, but currently it has become a global health problem because of migration. It is caused by the parasite Trypanosoma cruzi (T. cruzi), an intracellular flagellated protist whose life cycle has been defined in detail, although the immunopathology of its infection is still quite unknown. Moreover, there are differences in pathogenicity between the strains in mice, Y produces a high lethality while VFRA remains in the host in a most chronic manner. Comparative proteomics studies between T. cruzi strains have not been performed extensively, they should allow the detection of relevant virulent factors or distinctive functions. We focused on the proteome analysis of trypomastigotes of Y and VFRA strains by mass spectrometry followed by gene ontology analysis to display similarities or differences in cellular components, biological processes and molecular functions. Also, we performed metabolic pathways enrichment analysis to see the most relevant pathways in each strain. There were a 20% of different proteins showing that Y strain has specific proteins for replication and growth, while VFRA for localization or transport, among others. We found a very similar profile in the different ontology groups, but with some punctual differences. We detected enriched antioxidant defenses in VFRA that could correlate with its behavior of chronic infection in mice controlling the ROS production, while Y strain displayed a great enrichment of pathways related with nucleotides and protein production, explaining the high parasite load and lethality of this strain.

Project description:As Trypanosoma cruzi, the etiological agent of Chagas disease, multiplies in the cytoplasm of nucleated host cells, infection with this parasite is highly likely to affect host cells. We performed an exhaustive transcriptome analysis of T. cruzi-infected HeLa cells using an oligonucleotide microarray containing probes for greater than 47,000 human gene transcripts. In comparison with uninfected cells, those infected with T. cruzi showed greater than threefold up-regulation of 41 genes and greater than threefold down-regulation of 23 genes. Real-time reverse transcriptase-polymerase chain reaction (RT-PCR) of selected, differentially expressed genes confirmed the microarray data. Many of these up- and down-regulated genes were related to cellular proliferation, including seven up-regulated genes encoding proliferation inhibitors and three down-regulated genes encoding proliferation promoters, strongly suggesting that T. cruzi infection inhibits host cell proliferation, which may allow more time for T. cruzi to replicate and produce its intracellular nests. These findings provide new insight into the molecular mechanisms by which intracellular T. cruzi infection influences the host cell, leading to pathogenicity. Keywords: infection response
