Parallel genetic excisions of the cardiac troponin I N-terminal extension in tachycardic mammals

ABSTRACT: A study of cardiac troponin I evolution in mammals with high heart rates, including shrews, moles, and bats. With genomic, mRNA and protein level analyses we showed repeated loss of the N-terminal extension. Here, liquid chromatography with tandem mass spectrometry was used to verify cardiac troponin I (TNNI3) protein identity in ∼22 kDa bands from Pyrenean desman (Galemys pyrenaicus) and northern short-tailed shrew (Blarina brevicauda) hearts.

Genetic excision of the regulatory cardiac troponin I extension in high heart rate mammal clades

Project description:Mammalian cardiac troponin I (cTnI) contains a highly conserved N-terminal extension harboring protein kinase A targets (Ser23/24) which are phosphorylated during ß-adrenergic stimulation to increase cardiomyocyte relaxation rate. Here, we show that the Ser23/24 encoding exon 3 of TNNI3 was pseudoexonized multiple times in shrews and moles to mimic Ser23/24 phosphorylation without adrenergic stimulation, thereby facilitating the evolution of exceptionally high resting heart rates (~1000 beats min-1). We further reveal alternative splicing of TNNI3 exon 3 in distantly related bat families with both cTnI isoforms being incorporated into cardiac myofibrils.

Transcription profiling of zebrafish embryos following cardiac troponin T2 knockdown

Project description:We examined the transcriptional effect of preventing cardiac contraction in zebrafish embryos which can be deprived of circulation without experiencing hypoxia since the fish obtain sufficient oxygen via diffusion. Morpholino antisense knockdown of cardiac troponin T2 (tnnt2) prevented cardiac contraction without affecting vascular development. We concluded that absence of hemodynamic force induces endothelial CXCR4a up-regulation and promotes recovery of blood flow. We used microarrays to define the genetic signatures of the development of collateral vessels in zebrafish. Experiment Overall Design: One cell zebrafish embryos were injected with morpholino antisense against control or cardiac troponin t2, which prevents cardiac development. Whole embryo RNA was extracted at 36, 48, and 60h post fertilization and transcriptionally profiled.

Cardiac transcription factors in HL-1 cells: gene expression and genome binding profiling

Project description:[1] Gene expression profiling in Gata4, Mef2a knockdown in HL-1 cells. HL-1 cells infected with adenovirus expressing either control siRNA or Gata4, and Gata4 & Mef2a siRNA. [2] Genome-wide maps of cardiac transcription factors binding region in HL-1 cells. We performed bio-ChIP-seq using streptavidin beads immunoprecipitation of biotinylated 5 cardiac transcription factors (fbio) and P300 antibody ChIP-seq. We used a dox-inducible dual adenovirus system to express biotinylated Core Cardiac TFs in HL1. One adenovirus expressed the dox-activated reverse tet activator protein (rtTA) and the E. coli biotinylating enzyme BirA from the cardiac specific rat troponin T promoter. The second virus expressed a Core Cardiac TF fused to a C-terminal flag-bio epitope tag, where bio is the 15 amino acid substrate for BirA. This in vivo biotinylation approach permits pulldown of different factors to be performed under identical, stringent conditions, and circumvents limitations posed by available antibodies. We titrated adenovirus and dox doses to express GATA4flbio and MEF2Aflbio at near endogenous levels. The same conditions were then used to express SRFflbio, TBX5flbio, and NKX2-5flbio. Mass spectrometric ITEM-TWO analyses of interactions between human cardiac Troponin T (hcTnT) peptides and the M7 or the M11.7 antiTnT antibodies.

Project description:Molecular characterization of antibodies M7, and M11.7, as well as the antiTNF alpha antibody in addition to the synthetic peptides P11-P17 and P21-P32. The M7 and M11.7 anti-human cardiac Troponin T (antihcTnT) antibodies from Roche Diagnostics Deutschland GmbH (Mannheim, Germany) and the antiTNF alpha antibody from Thermo Fisher Scientific (Waltham, MA, USA, cat.no. 28401) were reconstituted and diluted with 200 mM ammonium acetate, pH 6.7, for nano-electrospray mass spectrometry. Molecular masses were determined. Synthesized peptides P11-P17 and P21-P32, delivered by 'Peptide&Elephants' (Henningsdorf, Germany), were dissolved in 200 mM ammonium acetate, pH 6.7, and also forwarded to nano-electrospray mass spectrometric analysis. Molecular masses and amino acid sequences were determined by mass spectrometric fragmentation of precursor ions. ITEM TWO: The antibodies M7, M11.7 and antiTNF alpha were incubated with selected synthesized peptides to generate solutions which contained the antibody and one synthesized peptide, dissolved in 200 mM ammonium acetate, pH 6.7. Antibody – peptide solutions were subjected to nano electrospray mass spectrometric ITEM-TWO analyses to investigate the binding behavior of the antibody and synthesized peptides, such as the apparent binding energies and dissociation constants of the complex dissociation reactions in the gas phase.

Small RNA Sequencing in Polytrauma Patients: High vs. Low Troponin T Levels

Project description:In this study, small RNA sequencing was performed on plasma samples from 10 polytraumatized patients (Injury Severity Score, ISS ≥ 16), who were stratified into two groups based on their initial Troponin T (TnT) levels at the emergency room. The high TnT group (n = 5) included patients with TnT concentrations > 50 pg/mL, while the low TnT group (n = 5) included those with TnT concentrations < 12 pg/mL. The aim of this dataset is to identify differentially expressed miRNAs associated with elevated TnT levels, a marker of cardiac injury in trauma patients. Differential expression analysis revealed several candidate miRNAs that may serve as potential biomarkers for cardiac dysfunction. This dataset highlights the diagnostic potential of miRNAs in post-trauma cardiac injury.
