Insights into enhancing Centella asiatica organ cell biofactories via hairy root protein profiling

Project description:The objectives of this study encompassed a thorough exploration of the potential implications of protein profiling in hairy roots, specifically focusing on optimizing and enhancing C. asiatica organ cell biofactories. In this pursuit, we categorized established C. asiatica hairy root lines according to their capacity for centelloside production, classifying them into HIGH, MID, or LOW categories. For comparative analysis, wild adventitious (Adv) roots were extracted from in vitro C. asiatica seedlings and cultivated in solid MS medium at 25°C in complete darkness, serving as control specimens. This meticulous, label-free proteomic analysis enabled the successful identification of several proteins. Our research substantially builds upon and extends the findings presented by Alcalde et al. (2022) (DOI: 10.3389/fpls.2022.1001023). In their study, distinctive morphological and metabolic variations were noted among different C. asiatica hairy root lines. Such differences are presumably attributable to the random incorporation of a selective set of genes from the T-DNA, with particular emphasis on the rol and aux genes.

RNA-Seq analysis of Medicago truncatula hairy root lines treated with methyl jasmonate

Project description:In hairy roots of the model legume Medicago truncatula the saponin production can be elicited by methyl jasmonate treatment. To identify genes potentially involved in saponin biosynthesis or its regulation we carried out transcript profiling by RNA-Seq of M. truncatula hairy roots treated with 100 μM of methyl jasmonate (dissolved in ethanol) for two or 24 hours. As control, M. truncatula hairy roots treated with an equivalent amount of ethanol were profiled.

Project description:This dataset belongs to a set of three RNA-Seq experiments that were carried out to study the regulation of monoterpenoid indole alkaloid production in the medicinal plant Catharanthus roseus. For this dataset, C. roseus hairy roots overexpressing the well-known MIA biosynthesis regulator ORCA3 were analyzed by RNA-Seq. As control, C. roseus hairy roots expressing GUS were used. Each analyzed sample consisted of an independent hairy root line; three hairy root lines per construct were analyzed.

TSAR1 and TSAR2 activate triterpene saponin production in Medicago truncatula

Project description:We identified two novel transcription factors ‘Triterpene Saponin Activation Regulator’ (TSAR) 1 and 2 that each activate a specific branch of saponin synthesis in M. truncatula. Therefore we generated three independent TSAR1 and three independent TSAR2 overexpression (OE) hairy root lines. As a control we raised three independent hairy root lines expressing a non-functional GUS gene. By qRT-PCR analyses and metabolite profiling of these lines, we found that TSAR1 OE leads to accumulation of non-haemolytic saponins whereas TSAR2 instigates accumulation of haemolytic saponins. To identify additional targets of TSAR1 and TSAR2 we carried out a genome-wide transcript profiling study by RNA-Seq.

Gene expression during early stages of arbuscular mycorrhizal root formation in Medicago truncatula.

Project description:Arbuscular mycorrhiza (AM) interactions between plants and Glomeromycota fungi primarily support phosphate aquisition of most terrestrial plant species. To unravel gene expression during early stages of Medicago truncatula root colonization by AM fungi, we used genome-wide transcriptome profiling based on mycorrhizal root fragments enriched for early fungal infection stages. We used Medicago GeneChips to detail the global programme of gene expression in response to early stages of colonization by arbuscular mycorrhizal fungi and identified genes differentially expressed during these early stages. Medicago truncatula GFP-HDEL hairy roots (genotypes A17 and DMI3) were grown in vertically-oriented petri dishes, incubated at 26M-BM-0C and inoculated with 8 Gigaspora margarita spores, which were positioned between the lateral roots. G.margarita spores germinated in 2 to 4 days. Hyphopodia were observed after 5-6 days. Root fragments which reacted to the fungal contact were collected and frozen. Non-inoculated control root fragments were harvested at a comparable age.

RNA-seq of soybean hairy roots in which GmMYB176 transcription factor was silenced or overexpressed

Project description:GmMYB176, an R1 MYB transcription factor regulates isoflavonoid biosynthesis in soybean. In the current experiment, GmMYB176 was silenced (GmMYB176-Si) or overexpressed (GmMYB176-OE) in soybean hairy roots and their effect on transcriptome was studied. RNA-Seq analyses of GmMYB176-Si and GmMYB176-OE along with control non-transformed soybean hairy roots revealed that alteration of gene expression of GmMYB176 affects gene regulation of hundreds of genes in soybean.
