The plasma membrane localization of PLEKHA5 is regulated by the phosphorylation status of S161

ABSTRACT: The phosphorylation of PLEKHA5 serine 161 residue affects the subcellular localization of the protein. Mutations of this site led to changes in plasma mebrane association during mitosis. The IP-MS experiment was conducted to confirm the phosphorylation at S161.

NuRD-interacting protein ZFP296 regulates genome-wide NuRD localization and differentiation of mouse embryonic stem cells

Project description:The Nucleosome Remodeling and Deacetylase (NuRD) complex plays an important role in gene expression regulation, stem cell self-renewal, and lineage commitment. Yet little is known about the dynamics of NuRD during cellular differentiation. Here, we study these dynamics using genome-wide profiling and quantitative interaction proteomics in mouse embryonic stem cells (ESCs) and neural progenitor cells (NPCs). The genomic targets of NuRD are highly dynamic during differentiation, with most binding occurring at cell-type specific promoters and enhancers. We identify ZFP296 as a novel, ESC-specific NuRD interactor that also interacts with the SIN3A complex. ChIP-sequencing in Zfp296 knockout (KO) ESCs reveals decreased NuRD binding both genome-wide and at ZFP296 binding sites, although this has little effect on the transcriptome. Nevertheless, Zfp296 KO ESCs exhibit delayed induction of lineage-specific markers upon differentiation to embryoid bodies. In summary, we identify an ESC-specific NuRD interacting protein which regulates genome-wide NuRD binding and cellular differentiation.

The dynamic interactome of Polycomb family complexes during stem cell differentiation

Project description:While the core subunits of Polycomb group (PcG) complexes are well characterized, little is known about the dynamics of these protein complexes during cellular differentiation. We used quantitative interaction proteomics to study PcG proteins in mouse embryonic stem cells (mESCs) and neural progenitor cells (NPCs). We found the stoichiometry of PRC1 and PRC2 to be highly dynamic during neural differentiation.

GRK2/3-regulated, but not constitutive, phosphorylation of the G protein-coupled receptor GPR84 controls arrestin interactions and subsequent function

Project description:Although the GPR84 activators 2-HTP and 6-OAU promoted phosphorylation of human GPR84 and its interactions with arrestin-3, PSB-16671 and DL-175 did not. Replacement of all 21 serine and threonine residues within the third intracellular loop, but not the 2 serines in the C-terminal tail, eliminated incorporation of [32P] promoted by 2-HTP and greatly reduced receptor-arrestin-3 interactions. Mass spectrometry indicated that GPR84 was phosphorylated constitutively on residues Ser221 and Ser224 whilst a range of sites became phosphorylated in response to 2-HTP. An antiserum able to identify pSer221/pSer224 recognised GPR84 from cells treated both with and without 2-THP, whilst an antiserum able to identify pThr263/pThr264 recognised GPR84 only after exposure to 2-HTP. Treatment with neither PSB-16671 nor DL-175 was able to promote receptor recognition by this antiserum. 2-HTP-mediated phosphorylation of Thr263/Thr264 was prevented by two chemically distinct GPR84 antagonists but neither affected constitutive phosphorylation of Ser221/Ser224. 2-HTP-mediated phosphorylation of Thr263/Thr264 but not constitutive phosphorylation of Ser221/Ser224 was prevented by GRK2/3 inhibition. Mutation of residues Thr263 and Thr264 to alanine generated a variant of GPR84 as limited in 2-HTP-induced interactions with arrestin-3 as when all 21 serine and threonine were eliminated from the third intracellular loop. By contrast this mutant was unaffected in capacity to reduce cAMP levels in response to 2-HTP. Homology modelling and mutagenesis provided molecular insight into differences in agonist function. These studies define key residues, regulated by GRK2/3, that define effective interactions with arrestins and provide novel tools to monitor the phosphorylation and functional status of GPR84.

Analysis of L1CAM-regulated signaling pathways and cellular responses to anti-L1CAM radioimmunotherapy in human ovarian cancer cells

Project description:L1 cell adhesion molecule (L1CAM) is overexpressed in various solid tumors, and its overexpression is linked to increased invasion, metastasis, angiogenesis, cancer stemness, and therapy resistance. However, the mechanisms of L1CAM-mediated carcinogenesis and radioresistance, particularly in high-grade serous ovarian carcinoma (HGSOC), the most common and lethal type of ovarian cancer, remain insufficiently understood. Anti-L1CAM radioimmunotherapy (RIT) using the radioisotopes Terbium-161 (Tb-161) and Lutetium-177 (Lu-177) has shown that Tb-161 is more cytotoxic than Lu-177, due to its distinct radioactive properties. Moreover, we identified a population of L1CAM+/CD133+ cells as cancer stem cells (CSCs) in ovarian cancer and demonstrated L1CAM's association with radioresistance. Understanding L1CAM-regulated downstream signaling and cellular responses to anti-L1CAM RIT with Tb-161, compared to Lu-177, is now critical. To address this, we established CRISPR-Cas9-mediated L1CAM knock-out (ΔL1CAM) in human ovarian cancer cells (OVCAR8) alongside a control cell line with a knockout of the nonessential gene AAVS1 (ΔAAVS1). In this study, quantitative phosphoproteomics, coupled with matching proteomics, revealed (i) L1CAM-dependent signaling pathways and biological processes by comparing ΔL1CAM with wild-type cells, and (ii) cellular responses to anti-L1CAM RIT with Tb-161 and Lu-177 in L1CAM-expressing ovarian cancer cells.

Discovery of genes regulated by the metastasis suppressor gene, RhoGDI2

Project description:A number of studies find that metastasis suppressor proteins, including RhoGDI2, may function in part though controlling expression of genes regulating metastasis (reviewed in Smith and Theodorescu, Nature Reviews Cancer, 2009, PMID: 19242414). Intracellular spatial localization regulated by the microtubule network.

Project description:The commonly recognized mechanisms for spatial regulation inside the cell are membrane-bounded compartmentalization and biochemical association with subcellular organelles. We use computational modeling to investigate another spatial regulation mechanism mediated by the microtubule network in the cell. Our results demonstrate that the mitotic spindle can impose strong sequestration and concentration effects on molecules with binding affinity for microtubules, especially dynein-directed cargoes. The model can recapitulate the essence of three experimental observations on distinct microtubule network morphologies: the sequestration of germ plasm components by the mitotic spindles in the Drosophila syncytial embryo, the asymmetric cell division initiated by the time delay in centrosome maturation in the Drosophila neuroblast, and the diffusional block between neighboring energids in the Drosophila syncytial embryo. Our model thus suggests that the cell cycle-dependent changes in the microtubule network are critical for achieving different spatial regulation effects. The microtubule network provides a spatially extensive docking platform for molecules and gives rise to a "structured cytoplasm", in contrast to a free and fluid environment.

Phosphoribosyl modification of poly-ubiquitin chains at the Legionella-containing vacuole prohibiting autophagy adaptor recognition

Project description:Ubiquitination is a crucial posttranslational modification in eukaryotes that plays a significant role in the infection of intracellular microbial pathogens, such as Legionella pneumophila, the bacterium responsible for Legionnaires’ disease. While the Legionella-containing vacuole (LCV) is coated with ubiquitin (Ub), it avoids recognition by autophagy adaptors. In this study, we report that the Sdc and Sde families of effectors work together to build ubiquitinated species around the LCV. The Sdc effectors catalyze canonical polyubiquitination directly on host targets or on the phosphoribosyl-Ub (PR-Ub) conjugated to host targets by Sde. Remarkably, the Ub moieties within the poly-Ub chains are either modified with a phosphoribosyl group by Sde and other PDE domain-containing effectors or covalently attached to other host substrates via Sde-mediated PR-ubiquitination. Furthermore, these modifications prevent the recognition by Ub adaptors, such as p62, and therefore exclude host autophagy adaptors from the LCV. Our findings shed light on the nature of the poly-ubiquitinated species present at the surface of the LCV and provide a molecular mechanism for the avoidance of autophagy adaptors by the Ub-decorated LCV.
