Identification of proteins bound with RPA194 at rDNA loci in hypoxic breast cancer cells

ABSTRACT: We combined chromatin immunoprecipitation (ChIP) with mass spectrometry to identify key bidning partners of RPA194 (the major subunit of RNA polI) at the rDNA loci in breast cancer cells subjected to hypoxia.

Project description:Solid tumors such as breast cancer have intratumoral regions of low oxygen tension, which influence a vast array of cellular responses. Hypoxia-Inducible Factor 1-alpha (HIF1α) represents the principal transcription factor orchestrating cellular responses to hypoxic conditions, mediating the regulation of genes implicated in oxygen homeostasis. The nucleolus is central stress response hub in the cell, as such we sought to identify novel binding partners of HIF1α in the nucleolar compartment during hypoxic stress.

Identification of ribosome proteins bound to IRES in hypoxia

Project description:Hypoxic conditions prompt internal ribosome entry site (IRES)-mediated translation of some of the hallmark cancer genes, such as VEGF. This translational switch is extremely vital for cell survival and tumor progression. Heterogeneity in ribosomes due to the diversity of ribosomal RNA (rRNA) and protein composition has been postulated to generate ‘specialized ribosomes’ that differentially regulate translation. A VEGF IRES sequence was used as bait to identify unique proteins bound at the IRES in breast cancer cells grown in hypoxia.

Project description:Breast cancer is one of the most common types of cancer in women. One key signaling pathway known to regulate tumor growth, metabolic adaptation, and cellular stress response in breast cancer is Wnt signaling. Breast cancer patients, specifically triple negative breast cancer (TNBC), with upregulated Wnt signaling often have a poor clinical prognosis. However, the effects of Wnt/β-catenin signaling on the nucleolus and the resultant impact on cancer development and progression remain unclear. A notable reduction was observed in the number of nucleoli per nucleus in response to Wnt/β-catenin signaling inhibition in multiple TNBC cell lines. Our comparative proteomic analysis revealed several changes in the composition of the nucleolar proteome of TNBC cells upon inhibition of Wnt signaling. Overall, we demonstrate that Wnt/β-catenin signaling will affects nucleolar functionality and thus influences breast cancer progression. Understanding the role of Wnt signaling in the nucleolus and breast cancer is a critical step towards developing novel therapeutic options for the treatment of breast cancer.

Of the different subtypes of breast cancer, the triple negative breast cancer subtype of breast cancer is the most aggressive. A proteomic screen of nucleolar content across breast cancer subtypes found that triple negative breast cancer cell lines have a distinct nucleolar proteome signature in comparison to non-TNBC breast cancer cell lines.

Project description:The nucleolus, responsible for ribosome biogenesis, acts as a key center for detecting and responding to cellular stress. Additionally, growing evidence suggests that ribosome biogenesis may play a significant role in promoting metastasis. A proteomic screen of nucleolar protein content between metastatic and non-metastatic breast cancers found that metastatic breast cancer cell lines have a unique nucleolar proteome signature as compared to non-metastatic breast cancer cell lines.
