Mimic miR-497a-5p in vivo intervention

ABSTRACT: The use of microRNA mimics to increase microRNA levels in the developing organs of newborn mice. Livers from mice treated with 100 mg/kg of an in vivo miR-497a-5p LNA mimic at postnatal day (P)1 and P3 were harvested at postnatal day (P)5. Lungs and livers from mice treated with daily 10 mg/kg intraperitoneal injections of an in vivo miR-497a-5p mimic were harvested at postnatal day 14.

Project description:Proton pump inhibitors (PPIs) have become top-selling drugs worldwide. They are prodrugs activated by protonation in highly acidic environments. They inhibit stomach acid secretion by covalently modifying the gastric H+/K+-ATPase. However, little is known about alternative activation mechanisms and target proteins in non-acidic environments. Employing a chemoproteomic approach in living cells, we find rabeprazole to form covalent conjugates with numerous zinc-binding proteins. We show that one of the main target proteins, density-regulated protein (DENR), is conjugated to rabeprazole through a cysteine residue forming part of the zinc-binding site. We propose that zinc acts as a Lewis acid, obviating the need for low pH, to promote the activation of PPIs in non-acidic environments. Our findings may aid the understanding of secondary drug effects and/or the repurposing of PPIs.

Mitoprotein-induced stress launches a global transcriptional fail-safe program

Project description:Most mitochondrial proteins are synthesized as cytosolic precursor proteins before being imported into mitochondria. Cytosolic accumulation of mitochondrial precursors hazards cellular fitness and is associated with a growing number of diseases, but is not observed under physiological conditions. Individual mechanisms that allow cells to avoid cytosolic accumulation of mitochondrial precursors have recently been discovered, but their interplay and regulation remain elusive. Here we show that cells rapidly launch a global transcriptional program to restore cellular proteostasis after induction of a “clogger” protein that reduces the number of available mitochondrial import sites. Cells upregulate the protein folding and proteolytic systems in the cytosol and downregulate both the cytosolic translation machinery and many mitochondrial metabolic enzymes, presumably to relieve the workload of the overstrained mitochondrial import system. We show that this transcriptional remodeling is a combination of a “wideband” core response regulated by the transcription factors Hsf1 and Rpn4 and a unique mitoprotein-induced downregulation of the oxidative phosphorylation components, the most abundant group of precursorsmediated by an inactivation of the Hap2/3/4/5 complex. These findings depict the first comprehensive, time-resolved model of adaptations to mitochondrial import impairment. This reveals the regulation of previously described reactions and combines them with a so far unidentified mitochondrion-specific arm into an overarching coordinated network of the cellular mitoprotein-induced stress response.

The general aim is to identify putative hits in ALS that relates with miR-146a expression.

Mislocalization of pathogenic RBM20 variants in dilated cardiomyopathy is caused by loss-of-interaction with Transportin-3

Project description:Severe forms of dilated cardiomyopathy (DCM) are associated with point mutations in the alternative splicing regulator RBM20 that are frequently located in the arginine/serine-rich domain (RS-domain). Such mutations can cause defective splicing and cytoplasmic mislocalization, which leads to the formation of detrimental cytoplasmic granules. Successful development of personalized therapies requires identifying the direct mechanisms of pathogenic RBM20 variants. Here, we decipher the molecular mechanism of RBM20 mislocalization and its specific role in DCM pathogenesis. We demonstrate that mislocalized RBM20 variants retain their splice regulatory activity, which reveals that aberrant cellular localization drives the pathological phenotype. A genome-wide CRISPR knock-out screen combined with image-enabled cell sorting identified Transportin-3 (TNPO3) as the main nuclear importer of RBM20. We show that the direct RBM20-TNPO3 interaction involves the RS-domain, disrupted by pathogenic variants. Re-localization of pathogenic RBM20 variants to the nucleus restores alternative splicing and dissolves cytoplasmic granules in cell culture and animal models. These findings provide proof-of-principle for developing therapeutic strategies to restore RBM20’s nuclear localization in RBM20-DCM patients.

Project description:The brain is a lipid-rich organ that experiences rapid growth and development after birth in a period hallmarked by extensive lipid synthesis. We still lack a fundamental understanding of lipid metabolism during this critical time of brain development and how these dynamics occur in infants born extremely preterm (<28 weeks of gestation) suffering from brain injuries. Using an established model of neonatal brain injury due to intermittent hypoxemia, we recapitulated hippocampal-dependent cognitive impairments and examined the extent of changes in the brain’s lipid and protein profile. Specifically, proteomic analysis was performed in microdissected hippocampus from postnatal day (P) 11 male and female mice that underwent intermittent hypoxia (IHx) from P1 to P11 or serve as normoxia (Nx) controls. Our results show that the most significantly downregulated proteins belong to the metabolism pathway and within the metabolism pathway, metabolism of lipids was one of the top three most downregulated pathways.

Specifically, Juk+ and Juk- P. aeruginosa cells were infected with phiKZ at MOI 2.5. Infected cells were collected at five minutes post-infection and prepared for proteomics analysis.Â
