Project description:This project investigates the mechanisms by which growth factors selectively activate nuclear calcium signaling to regulate gene transcription. While it is established that growth factors trigger an increase in nuclear inositol 1,4,5-trisphosphate (IP3) and subsequent calcium release via intranuclear IP3 receptors (ITPRs), the downstream structural and functional consequences within the nucleoplasm remain under characterization. Our study reveals that this calcium signaling event leads to the transient assembly of an actin nucleoskeleton, which specifically associates with the intranuclear non-muscle myosin 2A (MYH9).

Dual regulation of SLC25A39 by AFG3L2 and iron controls mitochondrial glutathione homeostasis

Project description:Organelle transporters define metabolic compartmentalization and how this metabolite transport process can be modulated is poorly explored. Here, we discovered that SLC25A39, a mitochondrial transporter critical for mitochondrial glutathione uptake, is a short-lived protein under dual regulation at the protein level. Co-immunoprecipitation mass spectrometry and CRISPR KO in cells identified that mitochondrial m-AAA protease AFG3L2 is responsible for degrading SLC25A39 through the matrix loop 1. SLC25A39 senses mitochondrial iron-sulfur cluster using four matrix cysteine residues and inhibits its degradation. SLC25A39 protein regulation is robust in developing and mature neurons. This dual transporter regulation, by protein quality control and metabolic sensing, allows modulating mitochondrial glutathione level in response to iron homeostasis, opening new avenues to explore regulation of metabolic compartmentalization. Neuronal SLC25A39 regulation connects mitochondrial protein quality control, glutathione and iron homeostasis, which were previously unrelated biochemical features in neurodegeneration.

Surveying the proteome-wide landscape of Mitoxantrone and examining drug-sensitivity in BRCA1-deficient ovarian cancer using quantitative proteomics

Project description:Mitoxantrone (MX) is regularly used to treat several cancers, yet despite its long history in the clinic, recent studies continue to unveil novel protein targets that may contribute to the cytotoxic effects of the drug, as well as potential non-canonical antitumor activity. A better understanding of MX’s cellular targets is required to fully comprehend the molecular consequences of treatment and to interpret MX-sensitivity in homologous recombination (HR)-deficient cancer. Here, we describe two approaches used to evaluate MX-activity in HR-deficient UWB1.289 ovarian cancer cells and survey the binding profile of MX using TMT-labeled quantitative proteomics and chemoproteomics. Mass spectrometry (MS) analysis of cellular extracts from MX-treated BRCA1- UWB1.289 cells revealed a unique downregulation of pathways instrumental to maintaining genomic stability, including single-strand annealing. Moreover, the BRCA1- cells exhibited a significant upregulation of proteins involved in ribosome biogenesis and RNA processing. Additional MS analyses following affinity-purification using a biotinylated-mitoxantrone probe (MXP) corrobo-rated these findings, which showed considerable targeting of proteins involved in ge-nomic maintenance and RNA processing. Our results suggest that an interplay of both canonical and non-canonical MX-antitumor activity overwhelms the BRCA1- cells. Fur-thermore, this study aids in characterizing the target landscape of MX, ultimately providing insights into off-target effects and MX-action in HR-deficient cancer.

A Modular Chromosomal Passenger Complex Rewires Chromosome Segregation in Plasmodium berghei

Project description:Faithful chromosome segregation relies on precise kinetochore-microtubule interactions and checkpoint surveillance, yet the molecular basis of these processes varies widely across eukaryotes and is only beginning to be defined in apicomplexan parasites. In the malaria parasite Plasmodium berghei, chromosome segregation is especially critical during transmission from host to mosquito: rapid mitoses generate male gametes, and subsequent meiosis in the zygote seeds the next generation of infection. Here, we identify Aurora-related kinase 1 (ARK1) as a central regulator of chromosome segregation in both mitotic and meiotic contexts. ARK1 localises to spindle poles, spindles, and kinetochores, and its depletion results in short and multipolar spindles, kinetochore misalignment, and failed chromosome partitioning. ARK1 forms a minimal Chromosomal Passenger Complex (CPC) with INCENP1 during male gametogenesis, but a more elaborate CPC with INCENP2, kinetochores, centromeric histones, and spindle assembly checkpoint proteins during meiosis. This stage-specific modularity allows Plasmodium to prioritise male gamete formation whilst safeguarding faithful chromosome inheritance during zygote development, ensuring parasite transmission to the mosquito. Our findings demonstrate that conserved CPC principles are rewired in Plasmodium, highlighting both the plasticity of eukaryotic checkpoint control and a potential vulnerability for blocking malaria transmission.

Antisense oligonucleotide-mediated inclusion of the TRA2β ultraconserved poison exon induces the expression of a lncRNA with anti-tumor effects

Project description:Dysregulated expression of oncogenic splicing factors (SFs) occurs in human tumors in part through alterations in splicing of poison exons (PE), highly conserved exons that trigger RNA degradation. TRA2β is one such SF that undergoes PE suppression in tumor cells. Therefore, manipulation of TRA2β-PE can be a promising therapeutic strategy for cancer. Here, we demonstrate that low TRA2β-PE inclusion correlates with decreased patient survival across multiple tumor types. TRA2β-PE-targeting antisense oligonucleotides (ASOs) induce anti-cancer phenotypes and widespread transcriptomic alterations in cell culture models by causing both TRA2β protein knockdown and upregulation of TRA2β-PE-containing transcripts, which act as long-noncoding RNAs (lncRNAs) to sequester nuclear proteins. Finally, we demonstrate efficacy of TRA2β-PE-targeting ASOs in preclinical 3D organoid and in vivo patient-derived xenograft models. Together, this demonstrates that oncogenic SFs can be drugged using PE-targeting ASOs and elucidates a mechanism by which the TRA2β-PE acts both as a regulator of TRA2β protein expression and as a lncRNA that controls cancer cell growth.
