Label free comparison of the protein cargo of extracellular vesicles (EVs) isolated from SC5314 and ecm33∆ mutant cells of Candida albicans after 16 hours of growth in YPD medium


ABSTRACT: Given the tendency to substitute cellular vaccines with acellular ones, and the complete lack of virulence shown by the C. albicans cell wall mutant ecm33∆, we performed the characterization of its EVs and test their possible use as vaccine for invasive candidiasis. Label free proteomics was carried out comparing the protein cargo of extracellular vesicles (EVs) isolated from SC5314 and ecm33∆ mutant cells of Candida albicans after 16 hours of growth in YPD medium.

Extracellular vesicles (EVs) from <i>Candida albicans</i> can elicit immune responses, positioning them as promising acellular vaccine candidates. We characterized EVs from an avirulent <i>C. albicans</i> cell wall mutant (<i>ecm33</i>Δ) and evaluated their protective potential against invasive candidiasis. EVs from the yeast (YEVs) and hyphal (HEVs) forms of the SC5314 wild-type strain were also tested, yielding high survival rates with SC5314 YEV (91%) and <i>ecm33</i> YEV immunization (64%).  ...[more]
