Meltome of P. falciparum across intraerythrocytic developmental cycle

ABSTRACT: Intact cell cellular thermal shift assay (CETSA) was performed on P. falciparum parasites and quantified by mass spectrometry. The experiment was performed in 7 blood stage timepoints (4, 10, 16, 22, 28, 34, 40 hpi) of a continuous parasite culture and repeated three times.

Project description:8-cell stage rat embryos were sired by males treated with saline or a chemotherapeutic cocktail, BEP, consisting of Bleomycin, Etoposide, and Cisplatin for 9 weeks. Animals were given an additional 9 weeks of recovery, without any treatment, prior to mating. qPCR gene expression profiling for 84 stem cell transcription factors. Total RNA extracted from 25-35 embryos at the eight-cell stage sired by control or BEP-treated males after a 9-wk recovery period (2 females mated per male, n= 4 males/treatment).

Response of malaria parasites to artemisin compounds and oxidative stress

Project description:Artemisin and its derivates are important antimalarials used for treatment of P. falciparum malaria. While the mode of action has been attributed to oxidative stress, methylation and unfolded protein response, the exact mechanism of action of these antimalarials is not known. Here, we used cellular thermal shift assay (CETSA) on live P. falcparum cultures to investigate the MOA of artemesinins using different artemisinin derivates (DHA, ART, AS, ATM and a control drug deoxyART) and different oxidative (di-amide and tert-BOOH) and reducing (DTT,NAC) agents.

Investigating the target and off-target landscape of PLK1 small molecule inhibitor volasertib

Project description:In this study we aimed to identify off-targets of polo-like kinase 1 (Plk1) small molecule inhibitor volasertib. Plk1 is an important cell cycle kinase and an attractive target for anticancer treatment. Volasertib is ATP-competitive small molecules that may also block the ATP-pocket of other proteins. Despite the fatal infections and negative survival in a phase III trial on volasertib in acute myeloid leukemia volasertib has been a promising treatment option in children with rhabdomyosarcoma. Therefore, there is a need to understand the nature of adverse effects that possibly originate from the off-target proteins. We used thermal proteome profiling (TPP) to identify proteins that have a change in the thermal stability after treatment with volasertib. The temperature of aggregation of several proteins involved in prostaglandin and phosphatidyl-inositol phosphate metabolism increased after treatment with volasertib. PIP4K2A and ZADH2 were stabilized both by treatment in living cells and cell lysate. Functional disruption of these proteins affects the immune response and fatty acid metabolism. In addition, volasertib was found to affect transcriptional coactivators, normal and alternative RNA splicing regulators and proteins involved in the intracellular transport regulation. Our data suggests that the identified proteins may contribute more to the understanding of anti-tumor effect of volasertib.

The RT2 Profiler PCR Arrays PAHS-30C (SA Biosciences, MD, USA) was designed to analyze 84 genes related to human insulin signaling pathway. The RT-PCR was carried out using an ABI PRISMM-BM-. 7000 Sequence Detection System (Applied Biosystems, Foster City, CA).The expression of HSD11B2 for the 3 experiments concerned was monitored in parallel by real time PCR which confirming significant downregulation by insulin. qPCR gene expression profiling. RNA from 3 separated experiments was collected.
