Identification of p53-regulated microRNAs in Hapatocellular Carcinoma (HCC).

Project description:p53 suppresses tumor progression and metastasis by regulating a large set of genes and microRNAs. By profiling 92 primary hepatocellular carcinomas (HCCs) and 9 HCC cell lines, we found that p53 upregulates microRNAs including miR-200 and miR-192 family members. By sequencing TP53 in 92 HCC samples, we classified the 92 samples into two groups (wt and mut). We also classified 9 HCC cell lines by testing p21 expression after DNA-damage mediated p53 activation. We then profiled microRNA expression in 92 HCC tissue samples and 9 HCC celll lines to identify p53-regulated microRNAs.

Project description:DUBs are involved in various pathways and signalling networks by pairing with E3 ubiquitin ligases in protein complexes. In order to test whether OTUD5 associates with an E3 ubiquitin ligase to regulate cell proliferation, the stable Flag-OTUD5/Hep3B cell line was established by transfecting Hep3B cells with plasmids expressing pCIN4-Flag-OTUD5. The cellular factors interacting with OTUD5 were purified by anti-Flag antibody-conjugated M2 beads.

The isoform PP2A (protein phosphatase 2)/B55alpha inhibits the activity of the prolyl hydroxylase 2 (PHD2) and activates the hypoxia inducing factor-1alpha (HIF-1alpha), playing a key role in in vessel remodeling. Thus, PP2A/B55alpha activators may have the potential to induce angiogenesis and arteriogenesis. Herein we investigated the pharmacological attributes of VCE-004.8 (Etrinabdione) and its effectiveness in a murine model of critical limb ischemia.

Proteomic characterization of the effect of CBD in animal models of PWA

Project description:The effect of CBD was investigated in a preclinical model of PCOS generated by chronic exposure of female mice to DHT from weaning onwards. To exacerbate the metabolic complications in this model, PWA mice were fed a high-fat diet (HFD) for 18 weeks before initiation of the pharmacological treatments. This model phenocopies a subtype of women with this condition who suffer metabolic comorbidities. PWA exacerbated the metabolic disturbances induced by the high-fat diet (HFD), as HFD-fed, non-androgenized female mice exhibited a smaller increase in body weight (BW) compared to androgenized females on the same diet before treatment initiation (Fig. 1A). Daily treatment with CBD for 28 days caused a clear reduction of BW through the treatment period. This datset contains the results of a proteomic characterization of the livers from control mice, PWA mice and CBD treated mice.
