The INO80 chromatin remodeling complex regulates histone H2A.Z mobility and the G1-S transition in oligodendrocyte precursors


ABSTRACT: This project explores the role of the chromatin remodelling complex in oligodendrogial development using a series of tools including conditional disruption of the INO80 gene within mice and primary oligodendrocyte precursors. This experiment sought to understand whether INO80 complex proteins interact with a major oligodendrocyte-specific transcription factor, OLIG2. OLIG2 was immuno-precipitated from cultured oligodendrocyte lineage cells differentiated from primary E13.5 cortical neural stem cells. Immunoprecipitated samples were then analysed for protein interaction partners via mass spectrometry.

Chromatin remodeling complexes (CRCs) participate in oligodendrocyte (OL) differentiation, survival, and maintenance. We asked whether CRCs also control the proliferation of OL precursors (OPs)-focusing on the INO80 complex, which is known to regulate the proliferation of a variety of other cell types during development and disease.
