Comparative protein profiling of wild type and ILDR2 knockout podocytes

ABSTRACT: The role of ILDR2 in podocytes, specialized epithelial cells in the kidney, is still unknown. We investigated the role of ILDR2 in glomeruli and its influence on blood filtration. Ildr2 knockout mice exhibited glomerular hypertrophy and decreased podocyte density, however, did not develop effacement of podocyte foot processes or proteinuria. LC-MS/MS proteomic analysis of isolated glomeruli showed an increase in matrix proteins such as fibronectin and agrin. This suggests a protective role of ILDR2 in glomerulopathies.

Project description:Podocyte detachment due to mechanical stress is a common issue in hypertension-induced kidney disease. The present study investigated the role of zyxin in mechanically stretched podocytes, a model for glomerular hypertension. Conditionally immortalized differentiated podocytes were seeded on flexible silicon membranes of a six well plate, which was mounted on a manifold connected to a custom-built stretch apparatus (NIPOKA GmbH, Greifswald, Germany), which induced cyclic pressure variations resulting in upward and downward motion of the silicone membranes. We found that zyxin is essential for the adhesion of the postmitotic cell type, the podocytes, in vitro during mechanical stretch as well as in vivo for the maintenance of the kidney filtration.

LC MS/MS of calcium-sensing receptor (CaSR) knockdown in cultured murine podocytes versus wildtype

Project description:Calcimimetic agents allosterically increase the calcium (Ca2+) sensitivity of the calcium-sensing receptor (CaSR), which is expressed in the tubular system and to a lesser extent in podocytes. Activation of this receptor can reduce glomerular proteinuria and structural damage in proteinuric animal models. However, the precise role of the podocyte CaSR is still unclear. A CaSR knockdown in cultured murine podocytes and a podocyte-specific CaSR knockout in BALB/c mice were generated to study its role in proteinuria and kidney function. Podocyte CaSR knockdown abolished the calcimimetic R-568 mediated Ca2+-influx, disrupted the actin cytoskeleton, reduced cellular attachment and migration velocity. Adriamycin (ADR)-induced proteinuria enhanced glomerular CaSR expression in wild type mice. Albuminuria, podocyte foot process effacement, podocyte loss and glomerular sclerosis were significantly more pronounced in ADR-treated podocyte-specific CaSR knockout mice compared to wild type littermates. The co-treatment of WT mice with ADR and the calcimimetic cinacalcet reduced the proteinuria in WT, but not in podocyte specific CaSR knockout mice. In addition, four children with nephrotic syndrome, objecting glucocorticoid therapy, were treated with cinacalcet for 1 to 33 days. Proteinuria declined transiently by up to 96% and edema resolved. The activation of podocyte CaSR regulates key podocyte functions in vitro and reduces toxin induced proteinuria and glomerular damage in mice. Our findings suggest a potential novel role of CaSR signaling in control of glomerular disease. Conditionally immortalized differentiated podocytes were seeded on flexible silicon membranes of a six well plate, which was mounted on a manifold connected to a custom-built stretch apparatus (NIPOKA GmbH, Greifswald, Germany), which induced cyclic pressure variations resulting in upward and downward motion of the silicone membranes. Pressure amplitude was chosen to give a maximum up- and downward deflection of the membrane centre of 6 mm (low stretch).

Pathogenesis of focal segmental glomerulosclerosis and minimal change disease

Project description:Podocyte injury is the hallmark of both focal segmental glomerulosclerosis (FSGS) and minimal change disease (MCD) and is ultimately reflected in foot process effacement proteinuria. Triggers and pathogenic pathways leading to podocyte cytoskeleton rearrangements are however incompletely explained. Here, we aimed to contribute to the understanding of these pathways using tissue bottom-up proteomic profiling of laser capture micro dissected glomeruli from MCD and FSGS.

SMPDL3b in Podocytes: Decoupling Proteinuria from CKD Progression in Experimental Alport Syndrome

Project description:Alport Syndrome (AS) is a rare genetic disease with impaired production of collagen type IV alpha 3, 4 and 5 chains in the glomerular basement membranes (GBM), which results amongst others in progressive loss of kidney function. In AS, abnormalities in the GBM and associated podocyte detachment may potentially result from the dysregulation of sphingolipid metabolism. Here we investigated whether renal sphingomyelin phosphodiesterase acid-like 3b (SMPDL3b) overexpression modulates the generation of sphingosine-1-phosphate (S1P) and contributes to renal failure in Col4a3 knockout mice, a mouse model of AS. We found a 3-fold increase in SMPDL3b expression in glomeruli and murine podocytes isolated from Col4a3 knockout mice. Increased SMPDL3b expression occurred in association with increased glomerular S1P levels, while podocyte specific Smpdl3b deletion in Col4a3 knockout mice was sufficient to restore S1P levels and to reduce proteinuria and podocyte foot process but not sufficient to protect from renal failure. Thus, podocyte S1P may be a key modulator of proteinuria and podocyte integrity in AS. Our study in experimental AS suggests that SMPDL3b-derived S1P may dissociate proteinuria from renal failure, and suggests that improvement of glomerular structure and function may not always translate in protection from CKD progression in AS.

Laser microdissection and mass spectrometry shows loss of podocyte proteins in minimal change disease and focal segmental glomerulosclerosis.

Project description:Minimal change disease and primary focal segmental glomerulosclerosis (FSGS) are common causes of nephrotic syndrome. Both diseases present with massive proteinuria and widespread foot process effacement on electron microscopy examination. On the other hand, secondary FSGS can also present with significant proteinuria, although the extent of foot process effacement can be variable. In this study, we performed laser microdissection of glomeruli followed by mass spectrometry to study podocyte proteins and compare the differences between the 3 diseases. Our study shows significant decrease in total spectral counts of nephrin, podocin, CD2-associated protein, alpha actinin 4, inverted formin 2 and dystroglycan 1 in all 3 diseases compared to time zero kidney transplant controls and membranous nephropathy biopsies (p <0.001). On the other hand, the decrease in the podocyte proteins in minimal change disease, primary FSGS and secondary FSGS was comparable and there was no significant difference. Surprisingly, there was no significant decrease in the podocyte proteins in membranous nephropathy compared to time zero kidney transplant control biopsies. In conclusion, our study shows significant decrease in major podocyte proteins in minimal change disease, primary and secondary FSGS, but not in time zero kidney transplant controls and in membranous nephropathy.

Arctigenin attenuates diabetic kidney disease through the activation of PP2A in podocytes

Project description:Arctigenin (ATG) is a major component of Fructus Arctii, which as a traditional herbal remedy reduced proteinuria in diabetic patients. However, whether ATG specifically provided renoprotection in DKD was not known. Here we report that ATG administration is sufficient to attenuate proteinuria and podocyte injury in mouse models of diabetes. Transcriptomic analysis of diabetic mouse glomeruli showed that cell adhesion and inflammation are two key pathways affected by ATG treatment, and mass spectrometry analysis identified protein phosphatase 2A (PP2A) as one of the top ATG-interacting proteins in renal cells. Enhanced PP2A activity by ATG reduces p65 NF-κB-mediated inflammatory response and high glucose-induced migration in cultured podocytes via its interaction with Drebrin-1. Importantly, podocyte-specific Pp2a deletion in mice exacerbates DKD injury and abrogates the ATG-mediated renoprotection. Collectively, our results clearly demonstrate a renoprotective mechanism of ATG via PP2A activation and establish PP2A as a potential target for DKD progression.

Cytoskeleton-associated protein 4 (CKAP4) affects podocyte cytoskeleton dynamics in diabetic kidney disease.

Project description:Cytoskeleton-associated protein 4 (CKAP4) stabilizes the endoplasmic reticulum, by regulating its folding and anchoring to the microtubular cytoskeleton. Since podocytes are highly dependent on an intact cytoskeleton to exert their function, the role of CKAP4 was explored during normal conditions and in diabetic kidney disease (DKD). The differences in CKAP4 gene expression between patients with chronic kidney disease (CKD) and controls were explored in publicly available databases. Expression of CKAP4 in patients with DKD was investigated with in situ hybridization. Localization in human kidney tissue was determined using immunofluorescence, qPCR, and western blot. The CKAP4 homolog was knocked down (KD) in zebrafish. Proteinuria and glomerular morphology were investigated. CKAP4 KD and overexpression in podocytes were investigated using proteomics, immunofluorescence, and western blot. Glomerular CKAP4 gene expression was reduced in DKD patients compared to healthy individuals, but not in other CKDs. The glomerular expression of CKAP4 was most pronounced in podocytes. CKAP4 KD zebrafish became proteinuric and podocyte FPE (foot process effacement) was observed. CKAP4 KD in podocytes in vitro led to disruption of actin stress fibers, microtubular rearrangement and loss of integrin expression. CKAP4 reduction causes podocyte FPE and proteinuria in vivo. Loss of CKAP4 disrupts the podocyte actin cytoskeleton, its microtubular orientation, and integrin-based glomerular basement membrane attachment. This indicates that CKAP4 is a crucial connector between cell body and structural elements in podocytes. The reduction of CKAP4 observed in glomeruli from DKD patients may thus be coupled to impaired podocyte function and proteinuria.
