Fission yeast Cdc10-YFP interactome in MM-N 16 hours

ABSTRACT: Mass spectrometry analysis of proteins co-purifying with Cdc10, a component of the MBF transcriptional complex, was performed both in a wild-type and in a whi5∆ background. Cdc10 was tagged with YFP at its C-terminal end and expressed from its endogenous promoter at its chromosomal locus. Cdc10-YFP was immunoprecipitated from cells cultured in MM-N medium for 16 hours, and co-purifying proteins were analysed by mass spectrometry. An untagged wild-type strain was used as a control.

After one hour of nitrogen starvation, Pab1-YFP was pulled down and co-purifying proteins were analysed by mass-spectrometry. Our results revealed the presence of all components of PP2A protein complexes including Paa1, the catalytic subunits Ppa1, Ppa2, and Ppa3.

Project description:We performed mass-spectrometry analysis of proteins co-purifying with the Paa1, the structural subunit of the PP2A protein phosphatase complex. Paa1 was tagged with YFP at the C-terminus and expressed from its endogenous promoter at its chromosomal locus. After one hour of nitrogen starvation, Paa1-YFP was pulled down and co-purifying proteins were analysed by mass-spectrometry. Our results revealed the presence of all components of PP2A protein complexes including Paa1, the catalytic subunits Ppa1, Ppa2, and Ppa3, and the regulatory subunits Pab1, Par1 and Par2. Additionally, several PP2A regulators (Igo1, Zds1, Dis2, Ppe1 and Ekc1) and components of the PP2A SIP/STRIPAK complex were also detected, confirming that the pull-down approach was successful. The mass-spectrometry analysis also showed an over-representation of proteins related to ribosome structure, translation initiation, aminoacylation, and tRNA modification in the Paa1 interactome. We focused on Trm112, a widely conserved protein with a crucial role in translation.

Mass spectrometry analysis of chromosomal passenger complex proteins and kinetochore proteins in Trypanosoma brucei

Project description:We performed chemical crosslinking mass spectrometry of native protein complexes purified from Trypanosoma brucei procyclic cells to obtain protein-protein interaction information for the chromosomal passenger complex (CPC) and kinetochore complex. Crosslinks among CPC subunits were detected using YFP-AUK1, while those among KKT8 complex subunits were detected in the YFP-KKIP1 sample. We also performed immunoprecipitation and mass spectrometry of CPC subunits (full-length proteins and fragments) to identify co-purifying proteins.

Potential tear biomarkers for the early diagnosis of Parkinson's disease- A pilot study

Project description:Parkinson's disease (PD) is the second most common neurodegenerative disease after Alzheimer's disease. The purpose of this study was to analyze the profile of the tear proteome of patients with idiopathic PD (iPD), carriers of the E46K-SNCA mutation and healthy subjects (CT) and to identify biomarkers for the early diagnosis of PD. An observational, prospective and case-control pilot study was carried out in 24 patients with iPD, 3 carriers of the E46K-SNCA mutation and 27 CT subjects. The patients' neurological involvement was scored and their tear samples were analyzed using nano-liquid chromatography-mass spectrometry (nLC-MS/MS). These analyses led to the identification of 560 tear proteins in which some of the deregulated proteins were involved in immune response, apoptosis, collagen degradation, protein synthesis, defense and altered lysosomal function. Of these proteins, six related to neurodegenerative processes, showed a good capacity to classify patients and controls. These findings revealed that some proteins were upregulated in the tears of PD patients, mainly those involved in lysosomal function. It is worth highlighting the importance of this study in identifying tear proteins implicated in neurodegeneration and its relationship with PD patients with an aggressive disease phenotype.

Fission yeast Whi5 represses MBF-dependent transcription in quiescent cells II

Project description:When cells arrest the cell cycle in G1 to enter the non-dividing quiescent state, the transcriptional machinery that promotes G1/S transition must be inactivated. Central to this regulation is Whi5, in budding yeast, and the Retinoblastoma (Rb) protein, in mammalian cells, acting as repressors of the transcription factors SBF, and E2F, respectively. In fission yeast, the MBF complex is the functional homologue of E2F in metazoans. Fission yeast Whi5/Mug54 is an uncharacterized protein predicted to function as a cell cycle transcriptional repressor at the G1/S transition. In this work, we show that after induction of quiescence by nitrogen starvation, fission yeast Whi5 is localised to the nucleus, and it is required for the repression of MBF-dependent genes. Mass spectrometry and bimolecular fluorescence complementation (BiFC) analyses revealed that Whi5 physically interacts with components of both the MBF and the histone deacetylase Clr6-I complexes. Moreover, Whi5 is required for the interaction of the MBF complex with Clr6-I, suggesting a role for Whi5 in maintaining transcriptional repression of MBF-dependent genes in quiescent cells by the recruitment of HDACs to gene promoters.

MAZ induces MYB expression during the exit from quiescence via the E2F site in the MYB promoter

Project description:Most E2F-binding sites repress transcription through the recruitment of Retinoblasoma (RB) family members until the end of the G1 cell-cycle phase. Although the MYB promoter contains an E2F-binding site, its transcription is activated shortly after the exit from quiescence, before RB family members inactivation, by unknown mechanisms. We had previously uncovered a nuclear factor distinct from E2F, Myb-sp, whose DNA-binding site overlapped the E2F element and had hypothesized that this factor might overcome the transcriptional repression of MYB by E2F-RB family members. We have purified Myb-sp and discovered that Myc-associated zinc finger proteins (MAZ) are major components. We show that various MAZ isoforms are present in Myb-sp and activate transcription via the MYB-E2F element. Moreover, while forced RB or p130 expression repressed the activity of a luciferase reporter driven by the MYB-E2F element, co-expression of MAZ proteins not only reverted repression, but also activated transcription. Finally, we show that MAZ binds the MYB promoter in vivo, that its binding site is critical for MYB transactivation, and that MAZ knockdown inhibits MYB expression during the exit from quiescence. Together, these data indicate that MAZ is essential to bypass MYB promoter repression by RB family members and to induce MYB expression.
