Intestinal inflammation induces remodelling in cerebrospinal fluid protein composition


ABSTRACT: Inflammatory bowel disease has been reported to trigger a variety of extra-intestinal manifestations in other organs, including the central nervous system (CNS). However, the direct impact of peripheral inflammation on the CNS is largely unknown. Cerebrospinal fluid (CSF) is a clear, colourless body fluid present in the brain and spinal cord. It performs vital functions since it protects the brain from injury and helps in maintaining a stable chemical environment for the central nervous system, acting as waste removal. Because CSF is in direct contact with the extracellular space of the brain, it provides insight into the status of the CNS. The analysis of changes in CSF proteome of murine models could help researchers in understanding the pathophysiology of several neurological diseases; however, the small volume of CSF in mice has thus far limited individual mouse proteome characterization. Moreover, the wide dynamic range of protein concentrations in CSF requires specific sample preparation for label-free proteomics. In order to address the impact of inflammatory bowel disease on CNS, we used a mouse model of intestinal painful inflammation, i.e. mice treated with 2.5 % dextran sodium sulphate (DSS) (w:v) in tap water ad libitum from day 0 until day 5, followed by DSS-free water in the next three days. By comparing the CSF protein content from 8 DSS-treated mice to that one derived from 7 control mice, we unveiled a pathogenic association between peripheral colonic inflammation and signals of early CNS alterations. Marked changes in cerebrospinal fluid protein composition were observed by untargeted proteomics, revealing enrichment in proteins related to inflammatory, immune response, complement, neuronal, and lipid metabolic pathways.
